The wall of the egg case of the dogfish (Scyliorhinus canicula) contains an analogue of collagen Types IV and VIII organised into a regular 3-dimensional network. It presumably provides both a protective and a filtering role for the eggs contained within it. Electron micrographs of longitudinal and transverse sections, including systematically tilted sections, have been used both to define, for the first time, the space group symmetry of the lattice and to carry out 3-D reconstruction of the unit cell contents. This cell was found to be tetragonal, space group I422, with a = b = 11 ± 1 nm, and c = 74 ± 4 nm. Consistent with this, projection symmetries were c2mm for the [1,0,0] view, p2mm for the [1,1,0] view, and p4mm for the projection down the c axis (the [0,0,1] view), and all observed reflections in the computed Fourier transform obeyed the ruleh+k+l= 2n(ninteger) for body-centred lattices. The 3-D reconstruction, the first electron micrograph 3-D reconstruction of a collagen-containing material, is interpreted in terms of variations of previous molecular models for this structure. Type IV collagen is a constituent of the basal lamina, where it forms a network with both structural and filtering properties. The dogfish egg case structure may throw light on the (less regular) collagen IV structure of the basal lamina.  相似文献

Genetic relatedness of 20 Salmonella isolates comprising 16 serotypes was analysed by restriction endonuclease digestion of the total DNA with six endonucleases individually. The rDNA fingerprints generated by EcoRI were more polymorphic, each serotype showed a unique fingerprint sharing several core (monomorphic) bands with several polymorphic bands. Eight characteristic NciI rDNA fingerprints were found. Similar rDNA RFLP patterns were observed in strains of Salmonella from different serotypes.S.I. Koh-Luar and E. Cytochrome c oxidase (COX) consists of 13 subunits, 3 encoded in the mitochondrial genome and 10 in the nucleus. Little is known of the role of the nuclear-encoded subunits, some of which exhibit tissue-specific isoforms. Subunit VIa is unique in having tissue-specific isoforms in all mammalian species examined. We examined relative evolutionary rates for the COX6A heart (H) and liver (L) isoform genes along the length of the molecule, specifically in relation to the tissue-specific function(s) of the two isoforms. Nonsynonymous (amino acid replacement) substitutions in the COX6AH gene occurred more frequently than in the ubiquitously expressed COX6AL gene. Maximum-parsimony analysis and sequence divergences from reconstructed ancestral sequences revealed that after the ancestral COX6A gene duplicated to yield the genes for the H and L isoforms, the sequences encoding the mitochondrial matrix region of the COX VIa protein experienced an elevated rate of nonsynonymous substitutions relative to synonymous substitutions. This is expected for relaxed selective constraints after gene duplication followed by purifying selection to preserve the replacements with tissue-specific functions.   相似文献

The phagocytic activity of neuroglial cells in adult feline degenerating optic nerve was investigated by immunocytochemistry at both light and electron microscopy levels. Degeneration was initiated by unilateral eye enucleation and the segment distal to the transection showing true Wallerian degeneration was examined. Following enucleation, twelve adult domestic cats were examined over a period of seven to 215 days. All cases showed slow clearance of myelin debris and absence of proliferating monocytes throughout the post-enucleation period. All phagocytic cells present were neuroglial cells, and many of these cells expressed oligodendroglial antigens. These findings demonstrate the persistence of an active population of oligodendrocytes that might play an additional functional role during Wallerian degeneration of feline optic nerve.  相似文献

HIV infection induces phenotypic and functional changes to CD8+ T cells defined by the coordinated upregulation of a series of negative checkpoint receptors that eventually result in T cell exhaustion and failure to control viral replication. We report that effector CD8+ T cells during HIV infection in blood and SIV infection in lymphoid tissue exhibit higher levels of the negative checkpoint receptor TIGIT. Increased frequencies of TIGIT+ and TIGIT+ PD-1+ CD8+ T cells correlated with parameters of HIV and SIV disease progression. TIGIT remained elevated despite viral suppression in those with either pharmacological antiretroviral control or immunologically in elite controllers. HIV and SIV-specific CD8+ T cells were dysfunctional and expressed high levels of TIGIT and PD-1. Ex-vivo single or combinational antibody blockade of TIGIT and/or PD-L1 restored viral-specific CD8+ T cell effector responses. The frequency of TIGIT+ CD4+ T cells correlated with the CD4+ T cell total HIV DNA. These findings identify TIGIT as a novel marker of dysfunctional HIV-specific T cells and suggest TIGIT along with other checkpoint receptors may be novel curative HIV targets to reverse T cell exhaustion.  相似文献

This study documents the monsoonal and lunar effects on species composition and abundance of microzooplankton in a tropical estuary. We investigated microzooplankton abundance in relation to the various environmental and biotic parameters, sampled in the Matang mangrove (Malaysia) from April 2013 to February 2014. A total of 39 microzooplankton taxa comprising four major groups, i.e. loricate ciliates (37.72%), aloricate ciliates (29.46%), dinoflagellates (24.33%) and meroplanktonic nauplii (8.49%) were identified. The loricate ciliates were the most diverse group with 31 taxa recorded. Four major species of loricate ciliates were identified, i.e. Tintinnopsis beroidea, Tintinnopsis rotundata, Stenosemella avellana and Tintinnidium primitivum, while Strombidiidae and Strobilidiidae dominated the aloricate ciliates. Although small loricate ciliates were ubiquitous, redundancy analysis shows marked shifts in microzooplankton community structure, from one that was dominated by loricate ciliates during the drier SW monsoon, to aloricate ciliates at the onset of the wet NE monsoon, and then to dinoflagellates towards the end of the drier NE monsoon period. These shifts were associated with rainfall, dissolved inorganic nutrients, salinity, temperature and microbial food abundance. There was no clear lunar effect on abundance of microzooplankton except for Favella ehrenbergii and copepod nauplii, which were more abundant during neap than spring tides.  相似文献

Retinylmonophosphatase (RMPase) activity in mouse brain paralleled the subcellular distribution of the plasma-membrane marker Na+ + K+-dependent ATPase. The enzyme had a pH optimum between 5.5 and 7.0. The enzyme demonstrated linear kinetics with respect to time and both protein and substrate concentrations. RMPase was saturated by low retinyl monophosphate (RMP) concentrations and exhibited an apparent Km of 4.6 microM. The enzyme did not require MgCl2 for activity, and in fact assays were routinely run in the presence of 10 mM-Na2EDTA. In general, detergents inhibited the enzyme, with 0.05% Triton X-100 causing a 30% loss of activity. Phosphatidic acid was also inhibitory, but phosphatidylcholine and sphingomyelin stimulated phosphatase activity. RMPase was inhibited 35% by 5 mM concentrations of fluoride, phosphate or pyrophosphate. A series of other phosphorylated compounds, including glucose 6-phosphate, alpha-glycerophosphate, ATP, AMP, p-nitrophenyl phosphate and thiamin pyrophosphate, showed little or no inhibition. RMPase activity differed in several characteristics from that previously reported for dolichylmonophosphatase. It is concluded that RMP could play a distinct role in the plasma membrane.  相似文献

Summary Distributions and abundances of ants were determined on the N, E, W and S upper slopes of a conical hill that had a gradient of conditions from moist-cool oakjuniper-pine woodland to warm-dry open shrub woodland. Species distributions varied remarkably with slope, even in terms of subfamilies and unique common species. A formicine was the most abundant species on the north slope. A different species of dolichoderine was most abundant on the other slopes. The assemblages of ant species were much more distinct for these small sites on each slope than are the known groups of species associated with vegetation types on a broad geographical basis. Interpretation of the causes of such specific assemblages awaits detailed information on species ecologies However, there is considerable circumstantial evidence that body size is an important factor. The percentage similarity of body size of pairs of species in the same site was least for common species of the same feeding guild; similarity was greater when one or both members of a pair were uncommon; similarities were highest for species in different guilds. Combinations of species that would have had very high size overlaps did not occur together. Some combinations that occurred were those that had the least overlap possible for the species known to occur on the entire hill.

On a constaté que la répartition des espèces variait de façon très marquée suivant la pente du versant, même en ce que concerne les sous-familles et les espèces uniques abondantes. Les assemblages d'espèces sont plus nettement démarqués dans ce cas que ne le sont ceux des groupes connus d'espèces associées avec des types de végétation sur des aires géographiques plus étendues. Avant de pouvoir donner une interprétation satisfaisante des causes qui déterminent les divers assemblages, il faudra disposer d'informations beaucoup plus détaillées sur l'écologie des diverses espèces. Il existe déjà, portant, beaucoup de preuves indirectes qui montrent l'importance de la taille spécifique. Les pourcentages de ressemblances des tailles spécifiques entre les espèces d'un même site étaient minimum chez les espèces communes appartenant au même groupe trophique. Les ressemblances étaient considérablement plus grandes lorsque l'un des membres d'une paire, ou les deux membres, appartenaient à des espèces rares. Les ressemblances maximum se rencontrent entre espèces appartenant à des groupes trophiques différents. On n'a trouvé aucune combinaison d'espèces communes dont les tailles spécifiques sont à peu près identiques. Les quelques combinaisons d'espèces communes qui ont été observées apparaissent entre espèces à la taille spécifique aussi divergentes que possible parmi toutes les expèces recensées dans l'air délimitée.
