Two chiral coordination polymers involving amino acid backbone l-cysteic acid (H2l-cys) and N-donor ligand 4,4′-bipyridine (4,4′-bpy) [{Cd(l-cys)(4,4′-bpy)(H2O)}3.5H2O]n1, [{Zn2(l-cys)2(4,4′-bpy)2(H2O)4}·(H2O)·(4,4′-bpy)]n2 with rectangular grids have been synthesized. Both compounds are insoluble in common polar/non-polar solvents and well characterized by various physico-chemical techniques such as CHN, IR, TGA, NMR, solid state CD and single crystal X-ray diffraction methods. Complexes 1 and 2 comprise l-cysteate dianions self assembled into chiral coordination polymers by bridging the adjacent metal centres through the carboxylate oxygen generating one-dimensional helical chains. The helical chains are pillared by 4,4′-bpy generating two dimensional network. Complex 1 generates two dimensional (4,4) rectangular grid network with dimension 4.77 Å × 11.74 Å (based on dCd···Cd) involving with the edge sharing l-cys and 4,4′-bpy ligands, respectively. Complex 2 possesses a brick-wall type (6,3) network topology with edge lengths of the grids 11.42 Å × 10.11 Å based on dZn···Zn. Lattice water molecules are encapsulated between the adjacent rectangular grids via hydrogen bonding interactions. One 4,4′-bpy moiety is stacked between the adjacent layers of brick-wall network via weak π···π interaction with the edge sharing N-donor ligand. Even though both the complexes are rigid and stable, N2 adsorption studies by these complexes revealed not much promising results. The terminal protruding sulphonate groups, angular orientation of the grids within the two-dimensional network and interpenetration of the adjacent offset sheets concomitantly prevent the formation of through tubular channels. Flexible coordination geometry around the metal centre and the versatile coordination mode of the amino carboxylate group from the l-cys moiety are accountable for the variation of the appealing network topology in these complexes. Chiral nature is established by solid state CD spectrum which shows a positive cotton effect for both complexes. 相似文献

In an analysis of the possible association of endothelial constitutive nitric oxide synthase (ecNOS) gene polymorphism and plasma nitric oxide levels in patients with acute coronary syndrome, we investigated 106 patients with the syndrome and 100 healthy controls. Genotype was determined using the polymerase chain reaction; plasma nitric oxide levels were found using ELISA. The genotype frequencies for the a/b polymorphism in the control group were 77% for bb, 19% for ab, and 4% for aa. In the patients, genotype frequencies were 55% bb, 34% ab, and 11% aa. The allele frequencies were 28% a and 72% b among the patients and 13% a and 87% b among control subjects. Our findings showed a significant association of the ecNOS gene polymorphism with acute coronary syndrome in the South Indian population. 相似文献

Background
The burden of the pandemic (H1N1) 2009 influenza might be underestimated if detection of the virus is mandated to diagnose infection. Using an alternate approach, we propose that a much higher pandemic burden was experienced in our institution.Methodology/Principal Findings
Consecutive patients (n = 2588) presenting to our hospital with influenza like illness (ILI) or severe acute respiratory infection (SARI) during a 1-year period (May 2009–April 2010) were prospectively recruited and tested for influenza A by real-time RT-PCR. Analysis of weekly trends showed an 11-fold increase in patients presenting with ILI/SARI during the peak pandemic period when compared with the pre-pandemic period and a significant (P<0.001) increase in SARI admissions during the pandemic period (30±15.9 admissions/week) when compared with pre-pandemic (7±2.5) and post-pandemic periods (5±3.8). However, Influenza A was detected in less than one-third of patients with ILI/SARI [699 (27.0%)]; a majority of these (557/699, 79.7%) were Pandemic (H1N1)2009 virus [A/H1N1/09]. An A/H1N1/09 positive test was correlated with shorter symptom duration prior to presentation (p = 0.03). More ILI cases tested positive for A/H1N1/09 when compared with SARI (27.4% vs. 14.6%, P = 0.037). When the entire study population was considered, A/H1N1/09 positivity was associated with lower risk of hospitalization (p<0.0001) and ICU admission (p = 0.013) suggesting mild self-limiting illness in a majority.Conclusion/Significance
Analysis of weekly trends of ILI/SARI suggest a higher burden of the pandemic attributable to A/H1N1/09 than estimates assessed by a positive PCR test alone. The study highlights methodological consideration in the estimation of burden of pandemic influenza in developing countries using hospital-based data that may help assess the impact of future outbreaks of respiratory illnesses. 相似文献134.
The drosophilid assemblages of four cultivated areas (soy, bean, corn, and orange plantations) grown in the core of the Neotropical region were analyzed by comparing their abundances and compositions. The collections, which were gathered using 38 banana traps, captured 12,560 drosophilids, including nine Neotropical and six exotic species. Most of the flies were collected in the bean (43%) and soy (42%) fields. The composition and relative abundance of species also varied among cultivated areas, with orange orchards presenting the highest relative abundance of exotics due to the dominance of the Afrotropical Zaprionus indianus (Gupta). Crop plantations were dominated by a Neotropical species, Drosophila cardini (Sturtevant), which has been shown to be well adapted to dry and disturbed environments. We discuss the drosophilid assemblages of the cultivated areas, comparing them with assemblages from neighbor urban and natural environments. The low drosophilid richness found in this study is similar to the richness found in urban environments and lower than the drosophilid richness of forests, supporting a pattern already known for other taxa. The high abundance of drosophilids in cultivated areas, as well as the dominance of a Neotropical species (D. cardini) in the crop assemblages, was a surprising result. 相似文献

Objective
ANGPTL8 is a liver and adipose tissue produced protein that regulates the level of triglyceride in plasma as well as glucose homeostasis. This study was designed to evaluate the level of ANGPTL8 in obese and non-obese subjects before and after exercise training.Methods
A total of 82 non-obese and 62 adult obese were enrolled in this study. Subjects underwent a three months of exercise training. Both full length and C-terminal 139–198 form of ANGPTL8 were measured by ELISA.Results
Our data show that the full length ANGPTL8 level was increased in obese subjects (1150.04 ± 108.10 pg/mL) compared to non-obese (775.54 ± 46.12) pg/mL (p-Value = 0.002). C-terminal 139–198 form of ANGPTL8 was also increased in obese subjects 0.28 ± 0.04 ng/mL vs 0.20 ± 0.02 ng/mL in non-obese (p-value = 0.058). In obese subjects, the levels of both forms were reduced after three months of exercise training; full length was reduced from 1150.04 ± 108.10 pg/mL to 852.04 ± 51.95 pg/mL (p-Values 0.015) and c-terminal form was reduced from 0.28 ± 0.04 ng/mL to 0.19 ± 0.03 ng/mL (p-Value = 0.058). Interestingly, full length ANGPTL8 was positively associated with fasting blood glucose (FBG) in non-obese (r = 0.317, p-Value = 0.006) and obese subjects (r = 0.346, p-Value = 0.006) C-terminal 139–198 form of ANGPTL8 on the other hand, did not show any correlation in both groups.Conclusion
In conclusion, our data demonstrate that ANGPTL8 was increased in obesity and reduced after exercise training supporting the potential therapeutic benefit of reducing ANGPTL8. The effects of alterations in sulfur metabolism on hepatic and renal metallothionein and glutathione metabolism were studied in the adult rat using inhibition of two enzymes of these pathways, hepatic cystathionase and renal gamma-glutamyl transpeptidase. Rats were fed a diet containing both methionine (0.66%) and cystine (0.20%) for 1 week before receiving three consecutive daily intraperitoneal injections of propargylglycine, a selective cystathionase inhibitor, at various doses (2.5-375 mumol/kg). When hepatic cystathionase was inhibited greater than 90% (greater than or equal to 50 mumol propargylglycine/kg), renal and hepatic metallothionein and hepatic glutathione were unaltered except at the highest dose. On the other hand, renal glutathione was increased two-fold with a concomitant decrease in renal gamma-glutamyl transpeptidase activity (50% of control). In another experiment, when renal gamma-glutamyl transpeptidase was inhibited greater than 90% with three consecutive daily injections of acivicin, a selective gamma-glutamyl transpeptidase inhibitor (10 mg/kg IP), renal glutathione content was unaltered while hepatic glutathione was decreased. Renal and hepatic metallothionein were not changed. Thus, the cysteine pools for metallothionein and glutathione appear unrelated under the present experimental conditions. In addition, following either proparglyglycine or acivicin injections, renal and hepatic glutathione pools appear to be altered differently. Human mesenchymal stem cells (MSCs), with capacity to differentiate into adipocytes, osteoblasts and chondrocytes, offer potential
for the development of novel treatments. A critical question in MSCs biology is whether this cell population possesses a relatively
uniform differentiation capability or is comprised of distinct subsets of progenitors committed to differentiate in particular
pathways. To quantify the changes during growth of MSCs, we analyzed the mesenchymal phenotype and differentiation ability
using a multi-marker PCR with six primer sets specific for CD73, CD90, CD105, CD166, CD45 and β-actin allowing a gel-based
differential detection of the PCR products. To determine degree of variability of MSCs populations in terms of proliferation,
cell proliferation assays were performed on expanded MSCs up to the sixth passage. At each passage, the osteogenic and adipogenic
differentiation potentials of MSCs were verified by culture in inductive media. RT-PCR and cytochemical analysis revealed
that, despite the loss of multipotentiality during expansion, certain markers remain expressed, indicating that these markers
are unlikely to be reflective of the MSC’s true ‘stem cell’ nature. Our results suggest that decrease in the expression of
MSCs specific markers correlates with down-regulation of proliferation ability and differentiation efficiency of MSCs. 相似文献

Summary Evaluation of the potential adverse human health effects of low-level chronic exposure to heavy metals is dependent on the
basic knowledge of the cellular and molecular toxicology of these metals. The use of various cell culture systems has greatly
facilitated our knowledge of the cellular effects. Inasmuch as most of the acute and chronic toxic effects of metals occur
primarily on the renal proximal tubules, the development of a rat kidney epithelial cell culture has provided a unique system
to study the uptake and mechanism of toxicity of metals and their intracellular binding ligands. In the presence ofd-valine, fibroblast growth was retarded and a primary epithelial monolayer culture was selectively grown from rat kidney cells.
A distinct difference in the uptake of chemically similar divalent metals, such as Pb2+, Hg2+, Cd2+, and Zn2+, was observed in these cells. Both Pb2+ and Hg2+ were more avidly taken up by kidney cells than Cd2+ and Zn2+ salts and they also showed increased toxicity. On the other hand, the cellular uptake of Cd from cadmium-metallothionein
(CdMT) was much less than from CdCl2, but CdMT was about seven times more toxic than CdCl2 when added to the renal cell culture. The cytotoxicity of CdCl2 was decreased significantly with pretreatment of the cells with CdCl2, although this had no effect on the toxicity of CdMT. The cellular toxicity of CdMT occurred probably during the process
of its transport across the plasma membrane whereas that of CdCl2 occurred after it had entered the cell. Thus rat kidney epithelial cells may be a useful tool to study the mechanism of renal
toxicity of environmental chemicals and drugs.
