Northeast China Transect (NECT), one of the fifteen International Biosphere-Geosphere Programme (IGBP) terrestrial transects, has been established for 10 years by Prof. Zhang Xin-Shi, through a core project of the IGBP - the Global Change and Terrestrial Ecosystems (GCTE). This transect is located in the mid-latitude semi-arid region, ranging 42-46°N latitude and 110-132癊 longitude. The primary driving force for global change is precipitation and the secondary one is land use intensity. Research progresses have been performed during the past decade in the following aspects: ecological database development, climate and its variability, ecophysiological response of plants to environments, vegetation and landscape changes, biodiversity patterns and their changes, plant functional types and traits with relation to climatic gradient, productivity and carbon dynamics, pollen-vegetation relationship, trace gas emissions, land use and land cover changes, as well as biogeographical and biogeochemical modelling. In order to achieve the higher level of integrated research, the NECT needs the consistent basic data sets within the same framework, further field experiments and observations, integrated simulations of vegetation structure, process and function from patch, landscape to biome scales, intercomparisons of results and simulations within the transect and to other IGBP transects, multidisciplinary research, national and international co-ordinates, and full scientific plan and implementation strategy.  相似文献

本研究采用四维杂交试剂,测定PCR产物的特征熔点温度(temperature of melting point,Tmp),对实时荧光定量PCR所获得阳性信号(特异性的或非特异性的)的结果进行分析和确认,以使检测结论更客观.将荧光探针模式的实时荧光PCR检测后的标本再进行熔解曲线温度扫描,然后在4℃冰箱冷却5min,向反应管加入1μL四维杂交液,再按照温度扫描程序做熔解曲线实验.结果显示,加四维杂交试剂之后的熔解曲线中信号峰值是收敛的,且信噪比增大.相同扩增产物的Tmp的误差是在±1℃之内.实验结果证明,四维杂交试剂对荧光探针模式实时荧光PCR结果可进行更精细的分析和确认.  相似文献

Laminin-2/4 is the major laminin isoform of normal muscle and nerve tissues and plays an important role in tumor invasion and metastasis. Despite the fact that laminin-2/4 has been found in the skin basement membrane, insufficient evidence is available on the effect of laminin-2/4 on the behavior of both normal and transformed skin cells. A comparison of the contribution of alpha2beta1, alpha3beta1, alpha6beta4 integrins and 67 kDa laminin receptor on the surface of the human epidermoid carcinoma cell, A-431, to interaction with laminin-2/4 was carried out. The cell interaction with extracellular matrix component is a multistage process. We employed new methods for studying different stages of the interaction of A-431 cells with laminin-2/4. We demonstrated that integrins alpha2beta1, alpha3beta1, alpha6beta4 and 67 kDa laminin receptor are involved in the interaction of A-431 cells with laminin-2/4. We found that contribution of the same receptors to different stages of the interaction with laminin can be different. alpha2beta1 integrins are involved in EGF-induced A-431 cells' migration on laminin-2/4. We demonstrated the cooperation between alpha2beta1 and alpha3beta1 integrins during adhesion and spreading of A-431 cells on laminin-2/4-coated substrate. These results provide information about laminin-2/4 receptors and their contribution to different stages of the interaction with cells.  相似文献

A comparison of the adhesion of human primary keratinocytes to laminin-1 from murine EHS sarcoma and laminin-2/4 from human placenta was carried out using two methods, cell adhesion to substrates covered with the laminin isoforms, and interaction of keratinocytes from suspension with latex beads coated with the proteins. Laminin-2/4 was considerably more potent as a promoter of attachment of primary human keratinocytes than laminin-1 (and fibronectin), with increased attachment of cells correlating well with the number of latex bead binding sites. Only small cells of diameter of less than 20 microm bound more than 5 beads. Staining of keratinocytes with involucrin antibodies confirmed the existence of an inverse relationship between laminin-2/4-coated bead binding and differentiation.  相似文献

20世纪70年代以前,全世界已知宽鳃鮟鱇属Sladenia Regan( 1908)鱼类仅有3种.1980~1982年期间,东海水产研究所“东方”号和南海水产研究所“南锋”号调查船分别在东海和南海的大陆架渔场调查时,共采集到4尾珍稀的深海鮟鱇鱼类.经研究,认为这4尾标本是1新种,定名为朱氏宽鳃鮟鱇Sladenia zhui sp.nov..  相似文献

应用羟脯氨酸法测定了牛蛙皮胶原蛋白含量,通过正交实验对牛蛙皮胶原蛋白酸法提取条件进行了优化,并结合MTT法测定了胶原蛋白对细胞生长和粘附性的影响。结果显示,牛蛙皮胶原蛋白的含量约为45.1%;温度条件对提取率影响最大,其次依次为酸种、时间和浓度,最优组合为乙酸、1.5mol/L、37℃、24h;在低浓度时,牛蛙皮胶原蛋白对正常人类肝细胞的生长和粘附性无显著影响,当浓度升高到一定值时,对细胞生长和粘附性有显著促进作用。  相似文献

应用EXPASY服务器(http://www.expasy.oh/tools)上的SOPMA法对SLA—DR的α链和口链进行二级结构的预测,并与人的HLA—DR相应的α链和β链的氨基酸和二级结构成分比较,在此基础上同源模建SLA—DRα链、β链及复合体SLA—DR的三级结构。结果显示,SLA—DR的α链各二级结构成分α螺旋、β折叠、转角和无规则卷曲的数目分别为36、61、4和69,β链中分别为42、56、16和74。α链和β链各二级结构成分与复合体SLA—DR具有高度的符合率,分别达到98.7%(α螺旋)、99.1%(β折叠)、83.3%(转角)和97.2%(无规则卷曲)。各功能区分析,SLA—DR的α1和β1区为结合抗原的高变化区。同源模建和三级结构分析表明SLA—DR的α链和口链具有独立的三级结构,并可以组成复合体SLA—DR。  相似文献

ST0838 (designed stRad55B) is one of the four RadA paralogs (or Rad55 homologues) in the genome of the hyperthermophilic crenarchaeon Sulfolobus tokodaii. The gene is induced by UV irradiation, suggesting that it is involved in DNA recombinational repair in this organism. However, this protein could not be expressed normally in vitro. In this study, thermostable and soluble stRad55B was obtained by co-expression with S. tokodaii RadA (stRadA) in E. coli, and the enzymatic properties were examined. It was found that stRad55B bound ssDNA preferentially and had a very weak ATPase activity that was not stimulated by DNA. The recombinant protein inhibited the strand exchange activity promoted by stRadA, indicating that stRad55B might be an inhibitor to the homologous recombination in this archaeon. The results will be helpful for further functional and interaction analysis of RadA paralogs and for the understanding of the mechanism of recombinational repair in archaea.  相似文献
