The HflX‐family is a widely distributed but poorly characterized family of translation factor‐related guanosine triphosphatases (GTPases) that interact with the large ribosomal subunit. This study describes the crystal structure of HflX from Sulfolobus solfataricus solved to 2.0‐Å resolution in apo‐ and GDP‐bound forms. The enzyme displays a two‐domain architecture with a novel “HflX domain” at the N‐terminus, and a classical G‐domain at the C‐terminus. The HflX domain is composed of a four‐stranded parallel β‐sheet flanked by two α‐helices on either side, and an anti‐parallel coiled coil of two long α‐helices that lead to the G‐domain. The cleft between the two domains accommodates the nucleotide binding site as well as the switch II region, which mediates interactions between the two domains. Conformational changes of the switch regions are therefore anticipated to reposition the HflX‐domain upon GTP‐binding. Slow GTPase activity has been confirmed, with an HflX domain deletion mutant exhibiting a 24‐fold enhanced turnover rate, suggesting a regulatory role for the HflX domain. The conserved positively charged surface patches of the HflX‐domain may mediate interaction with the large ribosomal subunit. 以耶氏解脂酵母菌(Yarrowialipolytica)MYA2613为底盘菌,表达来源于卷枝毛霉菌(Mucorcircinelloides)的基因car B,carRP后能有效生产β-胡萝卜素。启动子作为调控基因的重要顺式元件影响着基因的表达。为了优化基因GGS1,car B,carRP的表达强度组合,将上述3个基因、终止子与两组不同的启动子通过DNA assemble方法拼接后,得到了组合TEF1p-GGS1-xpr2t-GPD1pcar B-mig1t-EXP1p-carRP-lip2t和YAT1p-GGS1-xpr2t-GPD1p-car B-mig1t-FBAin1p-carRP-lip2t。经发酵表型验证,两株菌都产β-胡萝卜素,其中前者的产量是后者的4.18倍,具有明显优势。  相似文献

应用石蜡切片法对紫茎泽兰(Eupatorium adenophorum)从花芽分化到胚胎形成的全过程进行系统观察,结果表明:紫茎泽兰从刚孕花序至头状花序的直径(d)大小为1.6~1.8 mm时为花芽分化期;d在1.8~2.0和3.8~4.0 mm范围内为大小孢子及雌雄配子体发育期,其中d在2.3~3.5 mm之间时为减数分裂的主要过程;药壁分化过程为双子叶型,胞质分裂为同时型,绒毡层发育属腺质绒毡层,成熟花粉粒为3-细胞型;胚囊发育属蓼形,胚珠倒生,单珠被,薄珠心;胚胎发育过程中有双胚现象.  相似文献

去甲斑蝥素是我国自行研制的抗肿瘤药物,在临床上主要用于消化道肿瘤的治疗.实验表明,去甲斑蝥素可引起人胃癌BGC-823细胞发生 M期阻滞及细胞凋亡.进一步利用双向电泳和质谱技术,筛选出了去甲斑蝥素抑癌作用相关蛋白.研究显示,线粒体热休克蛋白CH60、线粒体ATP合酶d亚单位、内质网葡萄糖调节蛋白GRP78、线粒体Hsp70的辅助因子GRPE1、SH3L3以及染色质组装因子1小亚基RBBP4参与了去甲斑蝥素的抑癌作用.研究提示,去甲斑蝥素可能通过促进线粒体热休克蛋白及p53的表达进而激活caspase-3依赖的凋亡通路,并且去甲斑蝥素在引发内质网协迫之后,可通过抑制胞外信号调节激酶(extracellular signal regulated kinase, ERK)的活性促进肿瘤细胞的凋亡.进一步分析了去甲斑蝥素与线粒体ATP合酶抑制剂寡霉素A的联合用药对人胃癌细胞生长的影响,结果表明,联合用药的抑瘤效果比单独用药的抑瘤效果显著,提示去甲斑蝥素可能通过抑制线粒体ATP合酶功能抑制BGC-823生长.上述结果为优化去甲斑蝥素的联合用药方案提供了新线索.  相似文献

Nanomedicine is an emerging field that integrates nanotechnology, biomolecular engineering, life sciences and medicine; it is expected to produce major breakthroughs in medical diagnostics and therapeutics. Due to the size-compatibility of nano-scale structures and devices with proteins and nucleic acids, the design, synthesis and application of nanoprobes, nanocarriers and nanomachines provide unprecedented opportunities for achieving a better control of biological processes, and drastic improvements in disease detection, therapy, and prevention. Recent advances in nanomedicine include the development of functional nanoparticle based molecular imaging probes, nano-structured materials as drug/gene carriers for in vivo delivery, and engineered molecular machines for treating single-gene disorders. This review focuses on the development of molecular imaging probes and engineered nucleases for nanomedicine, including quantum dot bioconjugates, quantum dot-fluorescent protein FRET probes, molecular beacons, magnetic and gold nanoparticle based imaging contrast agents, and the design and validation of zinc finger nucleases (ZFNs) and TAL effector nucleases (TALENs) for gene targeting. The challenges in translating nanomedicine approaches to clinical applications are discussed.  相似文献

为探讨胰岛素对神经细胞中神经型一氧化氮合酶(nNOS)的表达及活性的影响,应用流式细胞术、原位杂交、电子自旋共振等技术方法研究胰岛素对PC12细胞中神经型一氧化氮合酶的影响.胰岛素作用PC12细胞9 h 后,神经型一氧化氮合酶的免疫荧光强度显著升高,且呈浓度依赖关系,其最大效应为对照的(155±13)%(P<0.01, n=3, t-test).加入胰岛素(16 mU/L, 6 h)也能够显著上调nNOS mRNA的表达,为对照的(182±13)%(P<0.01, n=3, t-test).另外加入胰岛素(16 mU/L)作用9 h后,神经型一氧化氮合酶的活性也显著升高,为对照的(167±15)%(P<0.01, n=4, t-test).由上述结果可知,胰岛素对PC12细胞的神经型一氧化氮合酶的表达及活性有上调作用.  相似文献

Insulin resistance, hyperglycemia, and type 2 diabetes are among the sequelae of metabolic syndromes that occur in 60-80% of human immunodeficiency virus (HIV)-positive patients treated with HIV-protease inhibitors (PIs). Studies to elucidate the molecular mechanism(s) contributing to these changes, however, have mainly focused on acute, in vitro actions of PIs. Here, we examined the chronic (7 wk) in vivo effects of the PI indinavir (IDV) in male Zucker diabetic fatty (fa/fa) (ZDF) rats. IDV exposure accelerated the diabetic state and dramatically exacerbated hyperglycemia and oral glucose intolerance in the ZDF rats, compared with vehicle-treated ZDF rats. Oligonucleotide gene array analyses revealed upregulation of suppressor of cytokine signaling-1 (SOCS-1) expression in insulin-sensitive tissues of IDV rats. SOCS-1 is a known inducer of insulin resistance and diabetes, and immunoblotting analyses revealed increases in SOCS-1 protein expression in adipose, skeletal muscle, and liver tissues of IDV-administered ZDF rats. This was associated with increases in the upstream regulator TNF-alpha and downstream effector sterol regulatory element-binding protein-1 and a decrease in IRS-2. IDV and other PIs currently in clinical use induced the SOCS-1 signaling cascade also in L6 myotubes and 3T3-L1 adipocytes exposed acutely to PIs under normal culturing conditions and in tissues from Zucker wild-type lean control rats administered PIs for 3 wk, suggesting an effect of these drugs even in the absence of background hyperglycemia/hyperlipidemia. Our findings therefore indicate that induction of the SOCS-1 signaling cascade by PIs could be an important contributing factor in the development of metabolic dysregulation associated with long-term exposures to HIV-PIs.  相似文献

通过开顶箱(OTCs)模拟,以环境臭氧(O3)浓度约40 nmol·mol-1为对照,研究大气O3浓度升高(80和160 nmol·mol-1O3)对冷季型草坪草高羊茅生长、亚细胞结构及其活性氧代谢的影响.结果表明: 14 d的80 nmol·mol-1O3熏蒸使高羊茅株高和叶宽降低,总生物量降低43.7%,老叶变黄,而160 nmol·mol-1O3处理高羊茅叶出现大量枯死褐斑,叶尖坏死,新叶卷曲,总生物量降低46.2%,叶肉细胞膜卷曲,叶绿体和线粒体受损严重.与对照相比,80和160 nmol·mol-1O3熏蒸下高羊茅叶片超氧阴离子(O2)产生速率、过氧化氢(H2O2)和丙二醛(MDA)含量显著增加,抗氧化酶活性显著升高,但叶片总酚含量和抗氧化能力随O3浓度升高而先升高后降低.在明显O3伤害症状出现之前,O3已对高羊茅的生长和抗氧化代谢产生不利影响;高羊茅抗氧化系统虽对O3浓度的升高存在一定的适应性反应,但其不能抵御过高浓度的长期胁迫和伤害.  相似文献
