The Drosophila mutation, quartet, affects development at points in the life cycle that require intense mitotic activity. Examination of embryos affected by the maternal effect of quartet has revealed defects that can be attributed to incomplete chromosome separation at mitosis. These defects include uneven spacing of nuclei, strands of DNA creating bridges between nuclei, and abnormal amounts of DNA per nucleus. Nuclei in quartet-affected embryos also have a greater-than-normal number of centrosomes. Immunofluorescent examination of the spindles in quartet-affected embryos has revealed tripolar spindles and adjacent spindles that share a common spindle pole. Finally, chromosome separation distance was measured in anaphase and telophase spindles in quartet-affected embryos and found to be blocked in anaphase. Examination of mitotic figures in quartet larvae revealed a reduced mitotic index and an elevated frequency of abnormal mitotic figures. quartet could encode a function necessary for the disengagement of chromosomes in mitosis, for kinetochore function or for function of a spindle motor. Mutations in quartet prevent the post-translational modification of three abundant proteins. These proteins may be involved in chromosome separation in mitosis.  相似文献

Picornaviruses utilize virally encoded RNA polymerase and a uridylylated protein primer to ensure replication of the entire viral genome. The molecular details of this mechanism are not well understood due to the lack of structural information. We report the crystal structure of human rhinovirus 16 3D RNA-dependent RNA polymerase (HRV16 3Dpol) at a 2.4-A resolution, representing the first complete polymerase structure from the Picornaviridae family. HRV16 3Dpol shares the canonical features of other known polymerase structures and contains an N-terminal region that tethers the fingers and thumb subdomains, forming a completely encircled active site cavity which is accessible through a small tunnel on the backside of the molecule. The small thumb subdomain contributes to the formation of a large cleft on the front face of the polymerase which also leads to the active site. The cleft appears large enough to accommodate a template:primer duplex during RNA elongation or a protein primer during the uridylylation stage of replication initiation. Based on the structural features of HRV16 3Dpo1 and the catalytic mechanism known for all polymerases, a front-loading model for uridylylation is proposed.  相似文献

Acoustic Features of Female Chacma Baboon Barks   总被引:3,自引:0,他引:3  
We studied variation in the loud barks of free-ranging female chacma baboons ( Papio cynocephalus ursinus ) with respect to context, predator type, and individuality over an 18-month period in the Moremi Game Reserve, Botswana. To examine acoustic differences in relation to these variables, we extracted a suite of acoustic parameters from digitized calls and applied discriminant function analyses. The barks constitute a graded continuum, ranging from a tonal, harmonically rich call into a call with a more noisy, harsh structure. Tonal barks are typically given when the signaler is at risk of losing contact with the group or when a mother and infant have become separated (contact barks). The harsher variants are given in response to large predators (alarm barks). However, there are also intermediate forms between the two subtypes which may occur in both situations. This finding is not due to an overlap of individuals' distinct distributions but can be replicated within individuals. Within the alarm bark category, there are significant differences between calls given in response to mammalian carnivores and those given in response to crocodiles. Again, there are intermediate variants. Both alarm call types are equally different from contact barks, indicating that the calls vary along different dimensions. Finally, there are consistent, significant differences among different individuals' calls. However, individual identity in one call type cannot directly be inferred from knowledge of the individuals' call characteristics in the other. In sum, the barks of female baboons potentially provide rich information to the recipients of these signals. The extent to which baboons discriminate between alarm and contact barks, and classify calls according to context and/or acoustic similarity will be described in a subsequent paper.  相似文献

Mutualisms affect the biodiversity, distribution and abundance of biological communities. However, ecological processes that drive mutualism-related shifts in population structure are often unclear and must be examined to elucidate how complex, multi-species mutualistic networks are formed and structured. In this study, we investigated how the presence of key marine mutualistic partners can drive the organisation of local communities on coral reefs. The cleaner wrasse, Labroides dimidiatus, removes ectoparasites and reduces stress hormones for multiple reef fish species, and their presence on coral reefs increases fish abundance and diversity. Such changes in population structure could be driven by increased recruitment of larval fish at settlement, or by post-settlement processes such as modified levels of migration or predation. We conducted a controlled field experiment to examine the effect of cleaners on recruitment processes of a common group of reef fishes, and showed that small patch reefs (61–285 m2) with cleaner wrasse had higher abundances of damselfish recruits than reefs from which cleaner wrasse had been removed over a 12-year period. However, the presence of cleaner wrasse did not affect species diversity of damselfish recruits. Our study provides evidence of the ecological processes that underpin changes in local population structure in the presence of a key mutualistic partner.  相似文献

The exact nature of many interspecific interactions remains unclear, with some evidence suggesting mutualism and other evidence pointing to parasitism for the same pair of interacting species. Here, we show spatial variation in the outcome of the cleaning relationship between Caribbean cleaning gobies (Elacatinus evelynae) and longfin damselfish (Stegastes diencaeus) over the distribution range of these species, and link this variation to the availability of ectoparasites. Cleaning interactions at sites with more ectoparasites were characterized by greater reductions in ectoparasite loads on damselfish clients and lower rates of removal of scales and mucus (i.e. cheating) by cleaning gobies, whereas the opposite was observed at sites where ectoparasite abundance was lower. For damselfish clients, cleaning was therefore clearly mutualistic in some locations, but sometimes neutral or even parasitic in others. Seasonal variability in ectoparasite abundance may ensure that locally low parasite availability, which promotes cleanerfish cheating, may be a transient condition at any given site. Conflicting conclusions about the nature of cleaning symbioses may, therefore, be explained by variation in ectoparasite abundance.  相似文献

The synthesis, SAR, pharmacokinetic profile, and modeling studies of both monocyclic and fused pyrazoles containing substituted N-arylpiperidinyl P4 moieties that are potent and selective factor Xa inhibitors will be discussed. Fused pyrazole analog 16a, with a 2'-methylsulfonylphenyl piperidine P4 group, was shown to be the best compound in this series (FXa Ki = 0.35 nM) based on potency, selectivity, and pharmacokinetic profile.  相似文献

Aggressive mimics are predatory species that resemble a 'model' species to gain access to food, mating opportunities or transportation at the expense of a signal receiver. Costs to the model may be variable, depending on the strength of the interaction between mimics and signal receivers. In the Indopacific, the bluestriped fangblenny Plagiotremus rhinorhynchos mimics juvenile cleaner wrasse Labroides dimidiatus. Instead of removing ectoparasites from larger coral reef fish, fangblennies attack fish to feed on scales and body tissue. In this study, juvenile cleaner wrasse suffered significant costs when associated with P. rhinorhynchos mimics in terms of reduced cleaning activity. Furthermore, the costs incurred by the model increased with heightened aggression by mimics towards signal receivers. This was apparently because of behavioural changes in signal receivers, as cleaning stations with mimics that attacked frequently were visited less. Variation in the costs incurred by the model may influence mimicry accuracy and avoidance learning by the signal receiver and thus affect the overall success and maintenance of the mimicry system.  相似文献
