PRMT3, a protein arginine methyltransferase, has been shown to influence ribosomal biosynthesis by catalyzing the dimethylation of the 40S ribosomal protein S2. Although PRMT3 has been reported to be a cytosolic protein, it has been shown to methylate histone H4 peptide (H4 1-24) in?vitro. Here, we?report the identification of a PRMT3 inhibitor (1-(benzo[d][1,2,3]thiadiazol-6-yl)-3-(2-cyclohexenylethyl)urea; compound 1) with IC(50) value of 2.5?μM by screening a library of 16,000 compounds using H4 (1-24) peptide as a substrate. The crystal structure of PRMT3 in complex with compound 1 as well as kinetic analysis reveals an allosteric mechanism of inhibition. Mutating PRMT3 residues within the allosteric site or using compound 1 analogs that disrupt interactions with allosteric site residues both abrogated binding and inhibitory activity. These data demonstrate an allosteric mechanism for inhibition of protein arginine methyltransferases, an emerging class of therapeutic targets.  相似文献

The musk shrew (Suncus murinus) is a small mammalian species belonging to Insectivora. It is widely distributed in Asia. To identify the genetic relationship among wild musk shrew populations and examine its migration route, we investigated the populations of Cambodia and Bhutan by using mitochondrial DNA restriction fragment length polymorphism analysis and compared them with other Asian populations previously described. Four haplotypes were detected in Cambodia and eight in Bhutan. A total of 53 haplotypes were detected in Asia and were classified largely into two groups, the Continental and Island types, based on a minimum spanning network. From the distribution of mtDNA types in wild musk shrews, three major population groups are identified in Asia: South Asia, Southeast Asia, and Malay. It is suggested that the Malay population group was a mix of South and Southeast Asian population groups and that this was a contact area of the two groups. In addition, other contact areas between the South and Southeast Asian groups exist in Myanmar, but unlike the Malay, the Myanmar area was the border of these groups.  相似文献

Coupling of incident light through an air region into an S-shape silver (Ag) plasmonic nanowire waveguide (SSAPNW) is a highly difficult challenge of light guiding on the surface of metal nanowire. In this paper, we numerically analyze the coupling effect of an SSAPNW which is covered by a dielectric medium using a finite element method. The coupling effect can be modulated by adjusting the Ag nanowire diameter and the covering dielectric medium width and wavelength of incident light, and the propagation length of surface plasmon (SP) coupling can be maximized. Simulation results reveal that the field confinement can be significantly improved and the majority of the electric field can be carried on the surface of a bending Ag nanowire. The effect of electric field transport along an SSAPNW due to SP coupling and Fabry-Perot resonance is investigated for different dimensions and lengths. Accordingly, long propagation lengths of about 41.5 μm for 10?×?SSAPNW at an incident wavelength of 810 nm and longer propagation length can be achieved if more sections of an SSAPNW are used. Simulation results offer an efficient method for optimizing SP coupling into bending metal nanowire waveguides and promote the realization of highly integrated plasmonic devices.  相似文献

The paper presents a new approach for medical image segmentation. Exudates are a visible sign of diabetic retinopathy that is the major reason of vision loss in patients with diabetes. If the exudates extend into the macular area, blindness may occur. Automated detection of exudates will assist ophthalmologists in early diagnosis. This segmentation process includes a new mechanism for clustering the elements of high-resolution images in order to improve precision and reduce computation time. The system applies K-means clustering to the image segmentation after getting optimized by Pillar algorithm; pillars are constructed in such a way that they can withstand the pressure. Improved pillar algorithm can optimize the K-means clustering for image segmentation in aspects of precision and computation time. This evaluates the proposed approach for image segmentation by comparing with Kmeans and Fuzzy C-means in a medical image. Using this method, identification of dark spot in the retina becomes easier and the proposed algorithm is applied on diabetic retinal images of all stages to identify hard and soft exudates, where the existing pillar K-means is more appropriate for brain MRI images. This proposed system help the doctors to identify the problem in the early stage and can suggest a better drug for preventing further retinal damage.  相似文献

Rigid chromatography resins, such as controlled pore glass based adsorbents, offer the advantage of high permeability and a linear pressure‐flow relationship irrespective of column diameter which improves process time and maximizes productivity. However, the rigidity and irregularly shaped nature of these resins often present challenges in achieving consistent and uniform packed beds as formation of bridges between resin particles can hinder bed consolidation. The standard flow‐pack method when applied to irregularly shaped particles does not yield well‐consolidated packed beds, resulting in formation of a head space and increased band broadening during operation. Vibration packing methods requiring the use of pneumatically driven vibrators are recommended to achieve full packed bed consolidation but limitations in manufacturing facilities and equipment may prevent the implementation of such devices. The stop‐flow packing method was developed as an improvement over the flow‐pack method to overcome these limitations and to improve bed consolidation without the use of vibrating devices. Transition analysis of large‐scale columns packed using the stop‐flow method over multiple cycles has shown a two‐ to three‐fold reduction of change in bed integrity values as compared to a flow‐packed bed demonstrating an improvement in packed bed stability in terms of the height equivalent to a theoretical plate (HETP) and peak asymmetry (As). © 2014 American Institute of Chemical Engineers Biotechnol. Conservation of rare plants can be accomplished by the restoration practice of reintroduction, but subsequent management is often required. In species with narrow habitat requirements, it is especially difficult to predict which management methods will be successful at new locations. Marsilea villosa is an endangered endemic Hawaiian fern with only seven remaining populations in ephemerally flooding drylands. Among its uncommon traits are long‐lived sporocarps, a requirement of flood and drought to complete its sexual life cycle, and the potential for extensive vegetative growth. An experiment was performed to determine which restoration techniques might best facilitate growth of outplanted M. villosa. The following effects were tested in a split‐plot factorial design: flooding (once/none), light (50% shade/full sun), weeding (bi‐monthly/none), and all interactions. We hypothesized that flooding would have the largest single‐factor effect and that there would be interactions among treatments. As hypothesized, flooding had the greatest positive effect on percent cover and sporocarp production. However, shade also increased cover over full sun when the plants began to experience drought. There was an interaction of light × flooding because M. villosa grew best in flooded, shaded plots. Weeding had no significant effect except in combination with flooding. Beyond protected status, current management of M. villosa only includes weed management at some populations. This study shows that if reintroducing new populations, the need for labor‐intensive weed management might be reduced if M. villosa is planted under conditions of flooding and moderate shade.  相似文献

Studies utilizing general learning and memory tasks have suggested the importance of neocortical structural plasticity for memory consolidation. However, these learning tasks typically result in learning of multiple different tasks over several days of training, making it difficult to determine the synaptic time course mediating each learning event. The current study used trace-eyeblink conditioning to determine the time course for neocortical spine modification during learning. With eyeblink conditioning, subjects are presented with a neutral, conditioned stimulus (CS) paired with a salient, unconditioned stimulus (US) to elicit an unconditioned response (UR). With multiple CS-US pairings, subjects learn to associate the CS with the US and exhibit a conditioned response (CR) when presented with the CS. Trace conditioning is when there is a stimulus free interval between the CS and the US. Utilizing trace-eyeblink conditioning with whisker stimulation as the CS (whisker-trace-eyeblink: WTEB), previous findings have shown that primary somatosensory (barrel) cortex is required for both acquisition and retention of the trace-association. Additionally, prior findings demonstrated that WTEB acquisition results in an expansion of the cytochrome oxidase whisker representation and synaptic modification in layer IV of barrel cortex. To further explore these findings and determine the time course for neocortical learning-induced spine modification, the present study utilized WTEB conditioning to examine Golgi-Cox stained neurons in layer IV of barrel cortex. Findings from this study demonstrated a training-dependent spine proliferation in layer IV of barrel cortex during trace associative learning. Furthermore, findings from this study showing that filopodia-like spines exhibited a similar pattern to the overall spine density further suggests that reorganization of synaptic contacts set the foundation for learning-induced neocortical modifications through the different neocortical layers.  相似文献

We suggest a new mechanism for the avoidance of self-superparasitism by a solitary endoparasitoid of free-living hosts. Meteorus pulchricornis (Wesmael) (Hymenoptera: Braconidae) is a solitary larval endoparasitoid of a wide range of Lepidoptera. First, we tested the tendency to avoid multiple oviposition on Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) larvae, examining the distribution pattern of eggs deposited in 20 hosts in 24 h. The standardized Morisita index of dispersion (Ip) for the number of eggs in a host larva was almost always below zero, and the 95% confidence limits of the means were lower than zero for both experienced and inexperienced females, indicating that parasitoids tend to avoid multiple oviposition, although not completely. Second, we observed changes in host movements (head shaking and forward movement) after oviposition and the occurrence of additional ovipositions in pre-oviposited hosts, because a recent study indicated the necessity of visual cues provided by host movements for oviposition by this species. The incidence of host movements decreased markedly after oviposition and remained at a low level for approximately 1 h, during which time additional ovipositions were rarely observed. Temporary host paralysis after oviposition would be a new mechanism for the avoidance of self-superparasitism in a solitary endoparasitoid using visual cues from moving hosts for oviposition.  相似文献

BACKGROUND: Allergic asthma strongly correlates with airway inflammation caused by cytokines secreted by allergen-specific type-2 T helper (Th2) cells, but the immunologic regulation of cell function is yet to be acquired. Further, IL-10 has been found to exert both antiinflammatory and immunoregulatory activities. This study aimed to elucidate the therapeutic effects of IL-10 administration via adenovirus-mediated gene delivery on airway inflammation in the ovalbumin (OVA)-induced murine model of asthma. METHODS: BALB/c mice were sensitized by intraperitoneal injections with OVA and challenged by nebulized OVA. The sensitized mice were given an intratracheal delivery of adenoviral vector expressing the murine IL-10 gene (AdIL-10), or mock adenoviral vector 4 days before the inhalation challenge of the OVA. Inflammatory parameters, such as the development of airway hyper-responsiveness (AHR), bronchial lavage fluid eosinophils, and chemokines were assayed. RESULTS: Intratracheal administration of AdIL-10 could efficiently inhibit antigen-induced AHR and significantly decrease the number of eosinophils and neutrophils in the bronchoalveolar lavage fluid of OVA-sensitized and challenged mice during the effector phase. CONCLUSIONS: Our data showed that the intratracheal transfer of the IL-10 gene could affect the recruitment of inflammatory cells during the challenge phase in a way that would result in the inhibition of airway inflammation. These findings suggest that the development of an immunoregulatory strategy based on IL-10 might shed light on more effective treatment.  相似文献

Within cells, dermatan sulfate (DS) and heparan sulfate (HS) are degraded in two steps. The initial endohydrolysis of these polysaccharides is followed by the sequential action of lysosomal exoenzymes to reduce the resulting oligosaccharides to monosaccharides and inorganic sulfate. Mucopolysaccharidosis (MPS) type II is a lysosomal storage disorder caused by a deficiency of the exoenzyme iduronate-2-sulfatase (I2S). Consequently, partially degraded fragments of DS and HS have been shown to accumulate in the lysosomes of affected cells and are excreted in the urine. Di- to hexadecasaccharides, isolated from the urine of a MPS II patient using anion exchange and gel filtration chromatography, were identified using electrospray ionization-tandem mass spectrometry (ESI-MS/MS). These oligosaccharides were shown to have non-reducing terminal iduronate-2-sulfate residues by digestion with recombinant I2S. A pattern of growing oligosaccharide chains composed of alternating uronic acid and N-acetylhexosamine residues was identified and suggested to originate from DS. A series of oligosaccharides consisting of hexosamine/N-acetylhexosamine alternating with uronic acid residues was also identified and on the basis of the presence of unacetylated hexosamine; these oligosaccharides are proposed to derive from HS. The presence of both odd and even-length oligosaccharides suggests both endo-beta-glucuronidase and endo-N-acetylhexosaminidase activities toward both glycosaminoglycans. Furthermore, the putative HS oligosaccharide structures identified indicate that heparanase activities are directed toward regions of both low and high sulfation, while the N-acetylhexosaminidase activity acted only in regions of low sulfation in this polysaccharide.  相似文献
