Conservation and sustainable management of wetlands requires participation of local stakeholders, including communities. The Bigodi Wetland is unusual because it is situated in a common property landscape but the local community has been running a successful community-based natural resource management programme (CBNRM) for the wetland for over a decade. Whilst external visitors to the wetland provide ecotourism revenues we sought to quantify community benefits through the use of wetland goods such as firewood, plant fibres, and the like, and costs associated with wild animals damaging farming activities. We interviewed 68 households living close to the wetland and valued their cash and non-cash incomes from farming and collection of non-timber forest products (NTFPs) and water. The majority of households collected a wide variety of plant and fish resources and water from the wetland for household use and livestock. Overall, 53% of total household cash and non-cash income was from collected products, mostly the wetland, 28% from arable agriculture, 12% from livestock and 7% from employment and cash transfers. Female-headed households had lower incomes than male-headed ones, and with a greater reliance on NTFPs. Annual losses due to wildlife damage were estimated at 4.2% of total gross income. Most respondents felt that the wetland was important for their livelihoods, with more than 80% identifying health, education, craft materials and firewood as key benefits. Ninety-five percent felt that the wetland was in a good condition and that most residents observed the agreed CBNRM rules regarding use of the wetland. We examined variability in yolk hormone levels among songbird species and the role of yolk steroids as a mechanism for enhanced exploitation of hosts by the parasitic brown-headed cowbird Molothrus ater . Within-clutch variation in yolk steroids has been found in several avian species in single species studies, but few comparisons have been made among species. We found a large range of differences in yolk testosterone among the seven passerine species examined, with significant differences between those at the high end (song sparrow Melospiza melodia , red-winged blackbird Agelaius phoeniceus , and house sparrow, Passer domesticus ) and those at the low end (eastern phoebe Sayornis phoebe , and house finch Carpodacus mexicanus ). We also found that the testosterone level in cowbird eggs was intermediate in relation to host species levels and was significantly lower than that in three common cowbird hosts (song sparrow, red-winged blackbird, and house sparrow), but not significantly different from three others. Geographical comparisons of yolk testosterone levels in all cowbird subspecies and populations from several regions showed no significant differences, though a trend that deserves further exploration was the pattern of lowest level in the ancestral population of cowbirds in the central prairies and of highest level in the northwestern population where range invasion occurred approximately 40 years ago. The levels of 17 beta-estradiol were similar in the seven songbird species examined, which is consistent with current hypotheses that this hormone plays a role in embryonic sexual differentiation. Further investigation is needed to determine whether the large differences observed among species in absolute level of yolk testosterone are the relevant focal point or whether target tissue sensitivity differences mediate the effects of this yolk steroid, particularly between parasitic and non-parasitic species. 相似文献

BACKGROUND: Liver slices have been reported to retain histological integrity and metabolic capacity for over 24 hours in flask culture systems, and they have been used for pharmacological and toxicological studies before. However, whether this method is suitable to measure hepatic glucose output is unknown. METHODS: Precision-cut liver slices were prepared from fresh male rat liver. After high-glucose pre-incubation (11.2 mmol/l), medium was changed to low-glucose conditions (0.5 mmol/l). Glucose and lactate levels as well as aspartate aminotransferase activity were monitored for 50 minutes with or without addition of insulin (600 pmol/l) and/or epinephrine (0.5 micromol/l). Slice potassium content and histology were examined to prove liver viability. RESULTS: We observed a stable glucose production from the liver slices of 0.3-0.4 micromol/g liver/min. Epinephrine increased (by 82+/-30%) and insulin decreased (by 80+/-8%) liver slice glucose output. Significant signs of ischemia were not detected. CONCLUSIONS: Hepatic glucose release can be reliably measured in a liver slice culture system, and it is regulated by major hormone systems. This method may be helpful for further characterization of direct insulin action and resistance in a complex tissue as the liver; however, pharmacological applications such as the analysis of drug effects on hepatic glucose metabolism can also be envisioned. 相似文献

Loblolly pine (LP, Pinus taeda) is the primary commercial species in southern forests of the US. Somatic embryogenesis (SE) is an effective technique to
implement clonal tree production of high-value genotypes from breeding and genetic engineering programs. Unlike angiosperm
embryos with attached cotyledons as seed storage organs, the diploid conifer embryo is surrounded by the unattached haploid
female gametophyte (FG). The FG is not present in culture. This presents a dilemma if the FG produces necessary or regulatory
compounds for embryo growth, since in culture these important compounds would be missing and would have to be added as supplements.
We report here the direct evidence that extracts from early-stage FG indeed stimulate early-stage somatic embryo (SME) growth
and multiplication, whereas extracts from late-stage FG inhibit early-stage SME growth. Furthermore, we have now isolated
this stimulatory substance from early-stage FG tissue, and identified this substance as citric acid on the basis of NMR and
mass spectrometry. We then demonstrated that topical application of citric acid to SMEs stimulates embryo colony growth at
P = 0.05. Moreover, we find that there is a good correlation between the amount of citric acid isolated from FG tissue (65
nmoles per stage 2–3 FG) and the amount of citric acid that stimulates colony growth (25–50 nmoles) when applied topically
to SMEs. This approach of isolating and characterizing a molecule from plant tissue, and investigating its role on SE processes

This study was conduced to assess the effects of a one time embryonic exposure to p,p'-DDE (dichlorodiphenyldichloroethylene; DDE) on the reproductive development and function in Japanese quail (Coturnix japonica). Embryos were exposed at day one of incubation to either 20 or 40 microg DDE or a sesame oil vehicle control (injection volume=20 microl). Onset of puberty, gonadal histopathology, sperm motility, cloacal gland size, and male copulatory behavior were assessed in adults. DDE accelerated onset of puberty in females and reduced male reproductive behaviors. Gonadal morphology and sperm motility appeared to be unaffected. Results from this study provide evidence that the neuroendocrine system may be more sensitive and less resilient to embryonic exposure to contaminants than traditional measures of reproductive success following contaminant exposure in adults. This study further supports the inclusion of behavioral assessments in toxicity tests. 相似文献

The relative importance of roots and AM-fungi on soil physical processes was investigated by controlling the presence of roots and AM fungi in pot experiments using a mycorrhiza-defective tomato mutant and a wild-type tomato (Solanum lycopersicum L.). Root-Zone and Bulk Soil sections were established by splitting pots into two lengthwise halves using a nylon mesh that contained roots whilst allowing the free movement of fungal hyphae. Post-incubation microbial populations and fungal biomass were measured and related to soil stability, pore structure and water repellency. Unplanted controls consistently had the least fungal biomass, fatty acids, water-stable aggregates (WSA) and water repellency. Wild-type-planted treatments had significantly more WSA than mycorrhiza-defective treatments (P?<?0.01). Fluctuations in water content induced by transpiration caused significant changes in soil pore structure, measured using high-resolution X-Ray computer tomography. Porosity and mean pore size increased in soil aggregates from planted treatments, which had larger more heterogeneous pores than those in the unplanted soils. AM fungi accentuated soil stability. However, changes were not linked to repellency and fungal biomass. The presence of plants, regardless of AM fungi, appears to have the greatest impact on increasing soil stability. 相似文献

An important predictor of male fitness is the fertilizing efficiency of their ejaculates. Ejaculates are costly to produce and males are predicted to devote greater resources to copulations with reproductively superior females. It is well established that males allocate different numbers of sperm to ejaculates. However, less is known about how males adjust their sperm quality, which has important implications for our understanding of fertilization and the evolution of sexual strategies. Here we test in the fowl, Gallus gallus, whether males adjust their sperm velocity by differentially allocating seminal fluid to copulations with attractive and unattractive females. To disentangle the contributions of sperm and seminal fluid to sperm velocity, we separated and remixed sperm and seminal fluid from ejaculates allocated to females of different attractiveness. We show that dominant males increase the velocity of the sperm they invest in more attractive females by allocating larger ejaculates that contain seminal fluid that increases sperm velocity. Furthermore, we find weak evidence that males also allocate sperm with higher velocity, irrespective of seminal fluid, to more attractive females. 相似文献

The Kaposi's sarcoma-associated herpesvirus (KSHV) latency-associated nuclear antigen 1 (LANA-1) is required for the replication of episomal viral genomes. Regions in its N-terminal and C-terminal domains mediate the interaction with host cell chromatin. Several cellular nuclear proteins, e.g., BRD2/RING3, histones H2A and H2B, MeCP2, DEK, and HP1alpha, have been suggested to mediate this interaction. In this work, we identify the double-bromodomain proteins BRD4 and BRD3/ORFX as additional LANA-1 interaction partners. The carboxy-terminal region of the short variant of BRD4 (BRD4S) containing the highly conserved extraterminal domain directly interacts with an element in the LANA-1 carboxy-terminal domain. We show that ectopically expressed BRD4S and BRD2/RING3 delay progression into the S phase of the cell cycle in epithelial and B-cell lines and increase cyclin E promoter activity. LANA-1 partly releases epithelial and B cells from a BRD4S- and BRD2/RING3-induced G1 cell cycle arrest and also promotes S-phase entry in the presence of BRD4S and BRD2/RING3. This is accompanied by a reduction in BRD4S-mediated cyclin E promoter activity. Our data are in keeping with the notion that the direct interaction of KSHV LANA-1 with BRD4 and other BRD proteins could play a role in the G1/S phase-promoting functions of KSHV LANA-1. Further, our data support a model in which the LANA-1 C terminus contributes to a functional attachment to acetylated histones H3 and H4 via BRD4 and BRD2, in addition to the recently demonstrated direct interaction (A. J. Barbera, J. V. Chodaparambil, B. Kelley-Clarke, V. Joukov, J. C. Walter, K. Luger, and K. M. Kaye, Science 311:856-861, 2006) of the LANA-1 N terminus with histones H2A and H2B. 相似文献
