Inclusion of spores of Bacillus subtilis var. niger in water-soluble crystals increased the resistance of the spores to dry heat and to a gaseous mixture of methyl bromide and ethylene oxide. Resistance of spores in glycine crystals to dry heat at 125 C was increased 5 to 24 times compared to unprotected spores. There appeared to be a positive correlation between the size of the crystal and the degree of resistance. The resistance to dry heat of spores included in sodium chloride crystals was about six times greater than unprotected spores. A gaseous mixture of methyl bromide (964 mg/liter) and ethylene oxide (642 mg/liter) at 37% relative humidity was ineffective in sterilizing spores enclosed within these water-soluble crystals, as was ethylene oxide alone. However, if the relative humidity was sufficiently high to dissolve the crystals during exposure to the vapor, viable-spore counts were drastically reduced or were negative. The surfaces of crystals grossly contaminated with dry spores were sterilized by exposure to gaseous ethylene oxide. Sterilization of heat-labile or moisture-labile materials with a critical requirement for sterility, as in planetary probes or drugs, may be complicated by the presence of spores in naturally occurring water-soluble crystals. Sodium hypochlorite was an excellent disinfectant at low temperatures. With the addition of ethylene glycol to prevent freezing, hypochlorite solutions at low free available chlorine concentrations, were effective against Bacillus subtilis var. niger spores from 0 to -40 C. The effectiveness of this decontaminant was influenced by temperature, pH, and concentration, with pH 7.2 the optimum for decontamination at all temperatures and concentrations. 相似文献

Gametophytes of Asplenium platyneuron, A. rhizophyllum and A. monlanum were grown from spores under controlled conditions. Factors inhibiting and promoting germination and growth were determined. Leaves cut from sexually produced sporophytes of A. platyneuron, A. rhizophyllum and A. montanum were cultured under controlled conditions on agar slants. The leaves produced neoplastic growths of two kinds: (1) two-dimensional growths, (2) three-dimensional growths. The former developed into normal-appearing, rhizoid- and gametangia-producing, heart-shaped gametophytes having a diploid chromosome complement. The formation of two-dimensional growths was favored by conditions drastically reducing the energy supply of the initiating sporophytic cells and by destroying the correlation of the leaf. Three-dimensional growths most often developed into sporophytes unless conditions were limiting; in the latter case they transformed into two-dimensional growths. Both two- and three-dimensional growths developed from fragmented leaves excised from buds growing near the tips of acuminate fronds of A. ebenoides. The high frequency of two-dimensional adventitious growth leads to the conclusion that apospory may provide a mechanism for the production of fertile allotetraploids, by the fusion of diploid gametes of adventitiously produced diploid gametophytes, in the genus Asplenium and perhaps other fern genera, in contrast to that which has been previously suggested in the literature. 相似文献

Fluorescein isothiocyanate-labeled beta-glucosidase was used as a simple staining reagent with selected gram-positive and gram-negative organisms. Staining in situ appeared to be dependent on the presence of accessible glycosidic-type linkages in the bacterial cell wall. Extensive wall damage or lysis did not occur when stained cells were suspended in washing and mounting solutions. The apparent specificity of labeled enzyme for wall substance was tested by blocking reactions, staining of isolated cell walls, and failure to stain substances lacking appropriate glycosidic linkages. Severe cell wall lesions were produced after prolonged contact with labeled enzyme, and this phenomenon may also be related to staining specificity. Gram-negative organisms and spores were poorly stained unless protected glycopeptide substrate was previously exposed by treatment of cells with thioglycolic acid or dilute alkaline sodium hypochlorite solution. A potential for staining tissues and cell lines may also exist. Some possible applications of labeled enzymes are briefly discussed. 相似文献

A method was developed to evaluate and measure the sporicidal activity of peracetic acid (PAA) and beta-propiolactone (BPL) at subzero temperatures as low as -40 C. Bacillus subtilis var. niger spores were used as the test organism. Both PAA and BPL were sporicidal at low temperatures, with PAA the more active. The temperature coefficients of the two chemicals are generally low over a range of 20 to -20 C, but increase significantly at temperatures below this. Results showed an initial lag in the PAA death rates that was directly dependent on the temperature. BPL did not show this lag time. 相似文献

Four types of hydrosol filters, two reusable (diatomaceous cylinder and fritted-glass funnel) and two disposable (asbestos pad and membrane filter) were challenged with a heavy bacterial suspension to assess their ability to produce sterile filtrates. Two of the four diatomaceous earth filters, the four fritted-glass funnels, and all of the asbestos pads tested generally gave sterile filtrates. However, only one type of filter, one of the membranes in its manufacturer's own holder, consistently gave sterile filtrates. The two other types of membranes usually gave sterile filtrates if tested in one manufacturer's holder, but all types invariably gave contaminated filtrates when tested in another manufacturer's holder. Contaminated filtrates were generally attributed to a poor reusable filter or to a faulty holder used with a disposable filter. If a high degree of certainty is required for sterile heat-labile filtrate, it is suggested that the liquid be passed through two or more filters in a previously tested and proven system. 相似文献

Bacillus cereus phospholipase was characterized as a phospholipase C by the analysis of lecithin degradation products by thin-layer and paper chromatography. Methanol in the growth menstruum inhibited completely the synthesis of phospholipase C, whereas the synthesis of lethal toxin and hemolysin were only partially inhibited. Dialysis of preformed B. cereus products against ethyl alcohol and methanol did not inactivate hemolytic, phospholipase C, or lethal activity. The hemolytic and lethal activities of culture filtrates were completely abolished by trypsin, but phospholipase C activity was resistant to inactivation. Lethal and phospholipase C properties of culture filtrates were resistant to inactivation at 45 C, whereas the hemolytic activity was completely destroyed. Lethal, hemolytic, and phospholipase C activities appeared simultaneously in a complex growth menstruum, but the kinetics of synthesis were different in all cases. Resolution of B. cereus filtrates on columns of Sephadex showed that the phospholipase C, hemolysin, and lethal toxin are distinct proteins. Evidence is also presented which suggests a correlation between the synthesis of B. cereus toxin and the period of transition from vegetative growth to sporulation. The activity of each B. cereus product was cation-independent, as opposed to cation-dependency of the phospholipase C and lethal activities of Clostridium perfringens alpha-toxin. Immunological cross-reactivity between the B. cereus products and C. perfringens alpha-toxin was not apparent; indeed, they were shown to be antigenically distinct. 相似文献
