Northern tamarisk beetles (Diorhabda carinulata) were released in the Upper Colorado River Basin in the United States in 2004–2007 to defoliate introduced tamarisk shrubs (Tamarix spp.) in the region's riparian zones. The primary purpose was to control the invasive shrub and reduce evapotranspiration (ET) by tamarisk in an attempt to increase stream flows. We evaluated beetle–tamarisk interactions with MODIS and Landsat imagery on 13 river systems, with vegetation indices used as indicators of the extent of defoliation and ET. Beetles are widespread and exhibit a pattern of colonize–defoliate–emigrate, so that riparian zones contain a mosaic of completely defoliated, partially defoliated, and refoliated tamarisk stands. Based on satellite data and ET algorithms, mean ET before beetle release (2000–2006) was 416 mm/year compared to postrelease (2007–2015) ET of 355 mm/year (p < 0.05) for a net reduction of 61 mm/year. This is lower than initial literature projections that ET would be reduced by 300–460 mm/year. Reasons for the lower‐than‐expected ET reductions are because baseline ET rates are lower than initially projected, and percentage ET reduction is low because tamarisk stands tend to regrow new leaves after defoliation and other plants help maintain canopy cover. Overall reductions in tamarisk green foliage during the study are 21%. However, ET in the Upper Basin has shown a steady decline since 2007 and equilibrium has not yet been reached. Defoliation is now proceeding from the Upper Basin into the Lower Basin at a rate of 40 km/year, much faster than initially projected. 相似文献

As approaches to ecological restoration become increasingly large scale and collaborative, there is a need to better understand social aspects of restoration and how they influence land management. In this article, we examine social perspectives that influence the determination of ecological reference conditions in restoration. Our analysis is based on in‐depth interviews with diverse stakeholders involved in collaborative restoration of fire‐adapted forest landscapes. We conducted interviews with 86 respondents from six forest collaboratives that are part of the U.S. Forest Service's Collaborative Forest Landscape Restoration Program. Collaboratives use a variety of approaches to develop reference conditions, including historic, contemporary, and future scenarios. Historical conditions prior to European settlement (nineteenth century or “pre‐settlement” conditions), or prior to more recent grazing, logging, and exclusion of fire, were the predominant type of reference used in all sites. Stakeholders described benefits and limitations of reference conditions. Primary benefits include (1) providing a science‐based framework for bringing stakeholders together around a common vision; (2) gaining social understanding and acceptance of the underlying need for restoration; and (3) serving to neutralize otherwise value‐laden discussions about multiple, sometimes competing, resource objectives. Limitations stem from (1) concerns over social conflict when reference conditions are perceived to contradict other stakeholder values and interests, (2) differing interpretations of reference condition science, (3) inappropriate application or over‐generalization of reference information, and (4) limited relevance of historical references for current and future conditions in some ecosystems. The intracellular conversion of testosterone to estradiol by the aromatase enzyme complex is an important step in many of the central actions of testosterone. In rats, estrogen given alone, or in combination with dihydrotestosterone, mimics most of the behavioral effects of testosterone, whereas treatment with antiestrogens or aromatase inhibitors block facilitation of copulatory behavior by testosterone. We used a highly sensitive in vitro radiometric assay to analyze the distribution and regulation of brain aromatase activity. Studies using micropunch dissections revealed that the highest levels of aromatase activity are found in an interconnected group of sexually dimorphic nuclei which constitutes a neural circuit important in the control of male sexual behavior. Androgen regulated aromatase activity in many diencephalic nucleic, including the medial preoptic nucleus, but not in the medial and cortical nuclei of the amygdala. Additional genetic evidence for both androgen-dependent and -independent control of brain AA was obtained by studies of androgen-insensitive testicular-feminized rats. These observations suggest that critical differences in enzyme responsiveness are present in different brain areas. Within several nuclei, sex differences in aromatase induction correlated with differences in nuclear androgen receptor concentrations suggesting that neural responsiveness to testosterone is sexually differentiated. Estradiol and dihydrotestosterone acted synergistically to regulate aromatase activity in the preoptic area. In addition, time-course studies showed that estrogen treatment increased the duration of nuclear androgen receptor occupation in the preoptic area of male rats treated with dihydrotestosterone. These results suggest possible ways that estrogens and androgens may interact at the cellular level to regulate neural function and behavior. 相似文献

In six chronic dialyzed uremic patients, an intravenous sodium selenite (Se 50 μg during 5 wk and then 100 μg) and zinc gluconate
(Zn 5 mg) supplementation was performed during 20 wk at each dialysis session three times weekly. Before supplementation,
plasma Se and Zn, plasma and erythrocytes (RBC) antioxidant metalloenzymes glutathione peroxidase (GPX), and superoxide dismutase
(SOD) were significantly decreased, whereas lipid peroxidation (as thiobarbituric acid reactants TBARs) was increased. To
obtain a significative change in plasma selenium, we had to use an Se dose of 100 μg/dialysis session. Then, treatment-increased
plasma Se (from 0.58 ±0.09 to 0.89±0.16 μmol/L) led to a repletion of RBC-GPX (from 29.6±6 to 43±5.8 U/g Hb) and increased
plasma GPX levels (from 62±13 to 151±43 U/L). Plasma Zn and RBC-SOD did not vary significantly. The change of TBARs was not
observed between wk 1 and 4. They decreased significantly between wk 4 (4.80±0.21μmol/L) and wk 20 (4.16±0.26 μmol/L). We
noted a low correlation between TBARs and plasma GPX. A strong correlation was observed between Se and plasma GPX. The reversal

Acute intermittent porphyria (AIP) is an autosomal dominant disease characterized by a deficiency of porphobilinogen deaminase (PBGD). Up to now 14 different mutations have been described. In an effort to investigate the molecular epidemiology of AIP we have undertaken a systematic study of different exons of the PBGD gene from a large number of unrelated patients. Here, exon 8 from 82 unrelated Dutch and French AIP patients was examined using single strand confirmation polymorphism analysis (SSCP) after polymerase chain reaction (PCR) amplification. A single base mutation, C to T, at position 346 of the sequence coding for PBGD was observed in 15 Dutch families but in only 1 French family. A simple PCR assay is described to facilitate the diagnosis of this common mutation at the DNA level. 相似文献

The Eastern Arc Mountains are one of the most important ecosystems that conserve biodiversity in the world. These ecosystems are threatened by the increasing occurrence of wildfires. Nevertheless, there is inadequate information useful for the development of effective strategies to prevent or respond to future fires. This paper analyses the current extent of dry season fires, underlying causes and the effectiveness of the fire management strategy being implemented in and around the Uluguru Nature Forest Reserve (UNFR) between 2016 and 2021. Differenced Normalised Burn Ratio derived from Landsat satellite images was applied to determine the extent of burned areas, and focus group discussions were held to determine the underlying causes of fires and the extent of implementation of fire management strategies. About 2% (472 ha) of reserved UNFR and 5% (2,854 ha) of unreserved forests were burned in 2017. Some of the fires impacted on 60% (370 ha) of the grassy Lukwangule plateau, which is home to a fire‐sensitive endemic species. The underlying causes of fires varied spatially across the mountains but generally, fire escaping from farm preparation and hunting activities were found to be the most prevalent. On average, survey participants perceived that fire management strategy objectives were achieved by only 29% mainly constrained by a shortage of financial and human resources. Our findings suggest that ignitions and fire spread in UNFR could be prevented or controlled through sustainable funding of fire management activities and the effective engagement of local communities in the management of the reserve. 相似文献

- Most of the world's large rivers are dammed for the purposes of water storage, flood control, and power production. Damming rivers fundamentally alters water temperature and flows in tailwater ecosystems, which in turn affects the presence and abundance of downstream biota.
- We collaborated with more than 200 citizen scientists to collect 2,194 light trap samples across 2 years and more than 2,000 river km. Samples contained 16,222 net‐spinning caddisfly (Hydropsyche) individuals across six species. We used these data to model the distribution of Hydropsyche throughout the Colorado River Basin in the western U.S.A. to identify the roles of water temperature, flows, and species‐specific morphology in determining aquatic species distributions throughout a large arid watershed that has been heavily altered by damming.
- We predicted that water temperatures would determine Hydropsyche presence and abundance to a greater extent than diel variation in river stage associated with hydropower production. Among many species, adult female Hydropsychids are morphologically adapted to swim to deep‐water oviposition sites. We predicted that the presence of this ability would negate the otherwise deleterious effects of high stage change on caddisfly egg mortality.
- We found that distributions of the two most widespread species, Hydropsyche occidentalis and Hydropsyche oslari (92% of total Hydropsyche captured), were both predicted by water temperatures. However, we also found that the abundance of H. oslari decreased by as much as 10‐fold as diel stage change increased, despite the presence of female morphological adaptations for deep‐water oviposition. We found sexual dimorphism and evidence for deep‐water swimming adaptations in 5/6 species.
- Our results show that net‐spinning caddisflies have species‐specific responses to environmental variation and suggest that environmental flows designed to reduce diel stage change and destabilise water temperatures may improve habitat quality for these ubiquitous and important aquatic insects.

Inorganic cofactors (Mn, Ca2+ and Cl-) are essential for oxidation of H2O to O2 by Photosystem II. The Mn reductants NH2OH and its N-methyl derivatives have been employed as probes to further examine the interactions between these species and Mn at the active site of H2O oxidation. Results of these studies show that the size of a hydroxylamine derivative regulates its ability to inactivate O2 evolution activity, and that this size-dependent inhibition behavior arises from the protein structure of Photosystem II. A set of anions (Cl-, F- and SO4
2-) is able to slow NH2OH and CH3NHOH inactivation of intact Photosystem II membranes by exerting a stabilizing influence on the extrinsic 23 and 17 kDa polypeptides. In contrast to this non-specific anion effect, only Cl- is capable of attenuating CH3NHOH and (CH3)2NOH inhibition in salt-washed preparations lacking the 23 and 17 kDa polypeptides. However, Cl- fails to protect against NH2OH inhibition in salt-washed membranes. These results indicate that the attack by NH2OH and its N-methyl derivatives on Mn occurs at different sites in the O2-evolving complex. The small reductant NH2OH acts at a Cl--insensitive site whereas the inhibitions by CH3NHOH and (CH3)2NOH involve a site that is Cl- sensitive. These findings are consistent with earlier studies showing that the size of primary amines controls the Cl- sensitivity of their binding to Mn in the O2-evolving complex.Abbreviation MES

Human cholesteryl ester transfer protein (CETP) mediates the net transfer of cholesteryl ester mass from atheroprotective high-density lipoproteins to atherogenic low-density lipoproteins by an unknown mechanism. Delineating this mechanism would be an important step toward the rational design of new CETP inhibitors for treating cardiovascular diseases. Using EM, single-particle image processing and molecular dynamics simulation, we discovered that CETP bridges a ternary complex with its N-terminal β-barrel domain penetrating into high-density lipoproteins and its C-terminal domain interacting with low-density lipoprotein or very-low-density lipoprotein. In our mechanistic model, the CETP lipoprotein-interacting regions, which are highly mobile, form pores that connect to a hydrophobic central cavity, thereby forming a tunnel for transfer of neutral lipids from donor to acceptor lipoproteins. These new insights into CETP transfer provide a molecular basis for analyzing mechanisms for CETP inhibition. 相似文献
