The use of low doses of dopamine in intensive care medicine

The dopamine alpha- and beta-adrenoceptor dose-response curves are investigated in four patients who are exempt from cardiovascular disease. A dose-related increase in CO, HR and SV is observed with infusion rates of up to 3 micrograms kg-1 min-1. With concentrations greater than 10 micrograms kg-1 min-1, both BP and SVR increase. Low-dose dopamine infusion less than 3 micrograms kg-1 min-1 is investigated in ten other patients. With this infusion rate, a selective renal vasodilation is induced without peripheral or cardiac beta-adrenoceptor activation. Dopamine is responsible for an increase in diuresis FENa, GFR and RBF. These properties are indicated in renal failure, and when haemodynamic support is required in cardiac failure, if an infusion rate of up to 10 micrograms kg-1 min-1 is able to reverse cardiac insufficiency.     

Presence of pH2-sensitive circulating interferon among Callithrix jacchus marmosets

Significant and relatively stable levels of serum-interferon were demonstrated in a Callithrix jacchus population. This circulating interferon was acid-sensitive in all cases, classifying it as immune or "gamma-type" interferon. Our results in these hematopoietic chimeras suggest that the presence of immune-type or at least pH 2-sensitive interferon could be related to the presence of two allogenic lymphocyte populations in each marmoset.     

What's new in lysozyme research?

The present review is focused on the main achievements realized in the lysozyme research field since the meeting held in 1972 to commemorate the fiftieth anniversary of the discovery of this enzyme. Despite of extensive structural, physico-chemical, crystallographic, genetic, immunological and evolutionary studies devoted to lysozymes, their biological role is still not exactly known.     

Substrate slope and orientation in a land hermit crab,Coenobita clypeatus (Decapoda,Coenobitidae)

Summary Coenobita clypeatus were tested on an underwater inclined plane in order to determine their behavioral threshold for perceiving slope, and to investigate the relative importance of input from the statocyst organ and from the weight of the body and gastropod shell on upward orienting ability. The shell's weight was compensated for by a neutral float and increased by weights. The crabs perceive an inclination of less than 5°, and do so with increasing certainty as the slope increases. Perception of the slope seems to be both by statocyst and proprioceptive input, but the latter is not essential for upward orientation.     

Characterization of the amino acids of bovine fibrinogen involved in the fibrinogen-thrombin interaction of the blood clotting process. Comparison with the milk clotting process

Summary Bovine fibrinogen and the A and B chains of bovine fibrinogen have been subjected to chemical modification by a number of reagents and the effects of these procedures on the susceptibility of the proteins to thrombin hydrolysis is described. The reagents used were rose bengal (for photo-oxidation), 2-hydroxy-5-nitrobenzyl bromide, N-acetylimidazole, iodoacetic acid and diethyl pyrocarbonate. Evidence is presented which indicates that the tryptophan and tyrosine residues of fibrinogen are not involved to any great extent in the interaction of this protein with thrombin. Modification with iodoacetic acid suggests that methionine residues play a major role in such interactions, but the fibrinogen chains on which the important residues reside remain uncertain. The use of diethyl pyrocarbonate indicates the participation also of histidine in fibrinogen-thrombin interactions and that, whereas the histidine residues of the B chain are involved to a great extent, it appears that those of the Aa chain are not. Étude d'un extrait de Sporothrix schenckii (forme levure). Analyse électrophorétique et immunoélectrophorétique; caracterisation des activités enzymatiques

Résumé Un extrait de S. schenckii a été préparé à partir de levures vivantes provenant d'une culture agitée pendant 3 jours à 35° C et à l'obscurité dans du milieu BHI (brain heart infusion). Il a été obtenu par broyage au broyeur cellulaire MSK et amélioré par une congélation-décongélation. Ses constituants, séparés par électrophorèse, ont été révélés par leur activité enzymatique; 30 bandes ont ainsi pu être caractérisées. Ces activités enzymatiques ont été recherchées au niveau des fractions de l'extrait révélées par un hyperimmunsérum de lapin: 16 sur les 22 précipités sont identifiés par leur pouvoir catalytique. L'ordre d'apparition des premiers anticorps chez le lapin immunisé et leur identification complètent cette étude. Les conditions de culture et de préparation permettant d'améliorer la qualité de l'extrait sont également précisées.

An extract from living yeast forms of S. schenckii was prepared. The yeasts originated from a shake culture in B.H.I. broth (Difco) incubated for 3 days at 35°C in darkness; they were harvested, washed and disrupted with glass beads in a model MSK Braun mechanical cell homogenizer; a freezing-thawing was added to improve the extract.After electrophoretic separation in agarose gel, the extract's components were characterized by their enzymic activity; with this technique, 30 bands were revealed. These enzymic activities were also investigated on the antigenic fractions of the extract revealed by a rabbit hyperimmunserum: 16 among 22 immunoprecipitates are identified by their catalytic properties. Study of the earliest precipitating antibodies (appearing-order and enzymic caracterization) in rabbits just immunized completes this work. How to ameliorate the quality of the extract by culture and extraction conditions is also specified.

     

Time resolved spectroscopy of the tryptophyl fluorescence of the E. coli LAC repressor.

A new crystal form of a mitogenic lectin from pea seeds ($\text{Pisum sativum}$) has been obtained which is suitable for high resolution structural work. The crystals are orthorhombic, space group P2₁2₁2₁, with unit cell dimensions: $\text{a}$ = 64.2Å, $\text{b}$ = 72. 7Å, $\text{c}$ = 108. 3Å. The asymmetric unit contains one protein molecule.
