以江西瑞昌石灰岩山区退耕还林5年的林地为对象,研究了不同退耕还林模式(林苗一体化、药用纯林、多树种混交和竹阔混交)对土壤有机碳含量的影响.结果表明:与对照相比,退耕还林地0~20 cm土层的土壤有机碳、微生物生物量碳和可矿化碳含量分别增加24.4%、29%和18.4%,退耕还林显著提高了土壤有机碳含量,增强了土壤碳储能力;0~10 cm土层的土壤有机碳、微生物生物量碳和可矿化碳含量极显著高于10~20 cm土层,且退耕还林地土层间差异大于对照;4种退耕还林模式中,林苗一体化模式对土壤有机碳的影响较大. 相似文献

The relationship between telomeres, nevi and melanoma is complex. Shorter telomeres have been found to be associated with many cancers and with number of nevi, a known risk factor for melanoma. However, shorter telomeres have also been found to decrease melanoma risk. We performed a systematic analysis of telomere-related genes and tagSNPs within these genes, in relation to the risk of melanoma, dysplastic nevi, and nevus count combining data from four studies conducted in Italy. In addition, we examined whether telomere length measured in peripheral blood leukocytes is related to the risk of melanoma, dysplastic nevi, number of nevi, or telomere-related SNPs. A total of 796 cases and 770 controls were genotyped for 517 SNPs in 39 telomere-related genes genotyped with a custom-made array. Replication of the top SNPs was conducted in two American populations consisting of 488 subjects from 53 melanoma-prone families and 1,086 cases and 1,024 controls from a case-control study. We estimated odds ratios for associations with SNPs and combined SNP P-values to compute gene region-specific, functional group-specific, and overall P-value using an adaptive rank-truncated product algorithm. In the Mediterranean population, we found suggestive evidence that RECQL4, a gene involved in genome stability, RTEL1, a gene regulating telomere elongation, and TERF2, a gene implicated in the protection of telomeres, were associated with melanoma, the presence of dysplastic nevi and number of nevi, respectively. However, these associations were not found in the American samples, suggesting variable melanoma susceptibility for these genes across populations or chance findings in our discovery sample. Bamboo is one of the fastest growing plants in the world, but their shoot buds develop very slowly. Information about the sugar storage and metabolism during the shoot growth is lacking. In the present study, we determined the activity of sucrose and starch metabolizing enzymes during the developmental period of Fargesia yunnanensis from shoot buds to the young culms that have achieved their full height. The soluble sugars and starch contents were also determined and analyzed in shoot buds and shoots at different developmental stages. The results showed that there were higher sucrose contents in shoot buds than shoots, which coincides with the sweeter taste of shoot buds. As the shoot buds sprouted out of the ground, the starch and sucrose were depleted sharply. Coupled with this, the activity of soluble acid invertase (SAI), cell wall-bound invertase (CWI), sucrose synthase at cleavage direction (SUSYC) and starch phosphorylase (STP) increased significantly in the rapidly elongating internodes. These enzymes dominated the rapid elongation of internodes. The activities of SAI, CWI, SUSYC and STP and adenosine diphosphate-glucose pyrophosphorylase were higher as compared to other enzymes in the shoot buds, but were far lower than those in the developing shoots. The slow growth of shoot buds was correlated with the low activity of these enzymes. These results complement our understanding of the physiological differences between shoot buds and elongating shoots and ascertain the physiological mechanism for the rapid growth of bamboo shoots. 相似文献

Rice is an important crop and major model plant for monocot functional genomics studies. With the establishment of various genetic resources for rice genomics, the next challenge is to systematically assign functions to predicted genes in the rice genome. Compared with the robustness of genome sequencing and bioinformatics techniques, progress in understanding the function of rice genes has lagged, hampering the utilization of rice genes for cereal crop improvement. The use of transfer DNA (T‐DNA) insertional mutagenesis offers the advantage of uniform distribution throughout the rice genome, but preferentially in gene‐rich regions, resulting in direct gene knockout or activation of genes within 20–30 kb up‐ and downstream of the T‐DNA insertion site and high gene tagging efficiency. Here, we summarize the recent progress in functional genomics using the T‐DNA‐tagged rice mutant population. We also discuss important features of T‐DNA activation‐ and knockout‐tagging and promoter‐trapping of the rice genome in relation to mutant and candidate gene characterizations and how to more efficiently utilize rice mutant populations and datasets for high‐throughput functional genomics and phenomics studies by forward and reverse genetics approaches. These studies may facilitate the translation of rice functional genomics research to improvements of rice and other cereal crops. 相似文献

[目的]为了研究铜绿假单胞菌全局调控因子RsmA对两个吩嗪(Phenazine)合成基因簇phz1和phz2的调控方式与机制.[方法]采用基因缺失和抗性基因(gentamycin resistance cassette,aacC1)插入相结合的策略构建了rsmA基因缺失突变株PA-RG ;通过构建互补表达载体和过表达载体,进一步确认RsmA对绿脓菌素的调控作用 ;采用电转化方法将构建的翻译融合表达载体pMEZ1(phz1'-'lacZ)和pMEZ2(phz2'-'lacZ)分别导入铜绿假单胞菌突变株PA-RG和野生株PAO1,采用Miller法测定融合β-半乳糖苷酶活性.[结果]在GA培养基中,互补分析和过表达分析表明,RsmA抑制绿脓菌素的合成.此外,pMEZ1在突变株PA-RG中的表达增强,为野生株的2-3倍 ;而pMEZ2在突变株PA-RG中的表达降低,野生株是突变株的2倍.[结论]由此初步判定,铜绿假单胞菌全局调控因子RsmA对两个不同吩嗪合成基因簇的调控作用具有特异性,在一定程度上RsmA负调控phz1,正调控phz2. 相似文献

噪音环境下花臭蛙求偶鸣声特征分析 总被引:1,自引:1,他引:1
2012年7月份,在黄山浮溪地区利用超声录音设备录制并分析了繁殖季节雄性花臭蛙(Odorrana schmackeri)个体的求偶鸣声。观察发现花臭蛙繁殖活动主要集中在7月中旬,繁殖高峰期活动无昼夜规律,全天均可见求偶鸣叫及抱对产卵等行为,并且多在浅水滩处活动。花臭蛙鸣声根据音节数和声谱特征可分为4种类型:即单音节音、婴儿音、双音节音和多音节断奏音,其中,单音节音、双音节音和婴儿音较为常见。利用Selena软件给出4种声音的语图以及各自对应的能谱图,利用Sound Analysis pro v1.2对单音节音、双音节音和婴儿音的鸣声特征参数进行定量分析,分析的声音参数包括鸣叫持续时间、音节数、音节持续时间、音节间隔、主频、脉冲率等。结果表明,花臭蛙鸣声的主频范围为1.8~4.5 k Hz(n=65)。鸣声不包括超声组分,主频峰值(3.1±0.7)k Hz,与前人电生理实验所得花臭蛙听觉敏感峰值一致,说明花臭蛙主要在这一频段进行通讯。对3种常见鸣叫音声音参数的单因素方差分析结果表明,双音节音与单音节音和婴儿音在声音持续时间上存在显著性差异(P0.01),双音节音和单音节音在第二谐波声强上也具有显著性差异(P=0.01)。花臭蛙的双音节音在3种常见鸣叫音中具有最长的持续时间,为(99.5±8.4)ms,故推测,双音节音为花臭蛙繁殖期主要求偶鸣声,并通过其鸣声时长的变化来体现自身品质的好坏。 相似文献
