A zero erucic acid (C22:1) line of Brassica juncea (VH486), adapted to the agronomic conditions of Northern India, has been modified for its fatty acid composition in the seed oil with antisense constructs using the sequence of fad2 gene of B. rapa. The full-length B. rapa fad2 cDNA sequence was determined by 5 and 3 RACE of a partial sequence available in the EST database. Construct pASfad2.1 contained 315 to 1251 bp and construct pASfad2.2 contained 1 to 1251 bp fragment of the fad2 gene, both in antisense orientation, driven by a truncated napin promoter. Analysis of the levels of linoleic acid (C18:2) in the BC1 seeds of single-copy transgenics showed that the construct pASfad2.2 gave better suppression of the fad2 gene as compared to the construct pASfad2.1. The BC1 transgenic seeds containing the pASfad2.2 construct segregated into two distinct classes of C18:2>20% (putative null homozygotes) and C18:2<20% (putative heterozygotes) in a 1:1 ratio, while the T1 seeds segregated into three classes, C18:2>20%, C18:2 between 12% and 20%) and C18:2<12% (putative homozygotes) in a 1:2:1 ratio. Putative homozygous T1 seeds (C18:2<12% analyzed by the half-seed method) of four of the transgenic lines were grown to establish T2 homozygous lines. These had ca. 73% C18:1 and 8 to 9% each of C18:2 and C18:3 (-linolenic acid) fractions in comparison to ca. 53% C18:1, 24% C18:2 and 16% C18:3 in the parental line VH486. 相似文献

E. coli nucleoside diphosphate kinase (EcoNDK) is an important cellular enzyme required to maintain balanced nucleotide pools in the cells. Recently, it was reported that EcoNDK is also a multifunctional base excision repair enzyme, possessing uracil-DNA glycosylase (UDG) and AP-DNA processing activities. We investigated for the presence of such activities in M. tuberculosis NDK (MtuNDK), which shares 45.2% identity, and 52.6% similarity with EcoNDK. In contrast to the robust uracil excision activity reported for EcoNDK, MtuNDK preparation exhibited very poor excision of uracil from DNA. However, this activity was undetectable when MtuNDK was purified from an ung(-) strain of E. coli, or when the assays were performed in the presence of extremely low amounts of a highly specific proteinaceous inhibitor, Ugi which forms an extremely tight complex with the host Ung (UDG), showing that MtuNDK preparation was contaminated with UDG. Reinvestigation of uracil processing activity of EcoNDK, showed that even this protein lacked UDG activity. All preparations of NDK were shown to be active by their autophosphorylation activity. Ugi neither displayed a physical interaction with EcoNDK nor did it affect autophosphorylation of NDKs. Further, neither of the NDK preparations processed the AP-DNA generated by UDG treatment of the uracil containing DNA duplexes. The eccentric connectivity index, which has recently been employed successfully for the development of numerous mathematical models for the prediction of biological activities of diverse nature, has been reformed to overcome its limitations caused by degeneracy and insensitivity towards heteroatoms. The reformed eccentric connectivity index, termed the eccentric connectivity topochemical index, overcomes the limitations of the eccentric connectivity index by exhibiting very low degeneracy and displaying sensitivity to both the presence and relative position of heteroatoms without compromizing the discriminating power of the eccentric connectivity index. The relationship of the eccentric connectivity topochemical index, eccentric connectivity index and Wieners index with regard to the anti-HIV activity of 2, 3-diaryl-1, 3-thiazolidin-4-one derivatives was subsequently investigated. The values of the eccentric connectivity topochemical index, the eccentric connectivity index and Wieners index of each of 31 analogues comprizing the data set were computed using in-house computer program. Resultant data was analyzed and suitable models developed after identification of active ranges. Subsequently, each derivative was assigned a biological activity using these models, which was then compared with the reported anti-HIV activity. The accuracy of prediction using these models was found to vary from 81 to 90%. The present studies were initiated to determine the protein expression patterns of fibroblast-like synovial (FLS) cells derived
from the synovia of rheumatoid arthritis patients. The cellular proteins were separated by two-dimensional polyacrylamide
gel electrophoresis and the in-gel digested proteins were analyzed by matrix-assisted laser desorption ionization mass spectrometry.
A total of 368 spots were examined and 254 identifications were made. The studies identified a number of proteins that have
been implicated in the normal or pathological FLS function (e.g. uridine diphosphoglucose dehydrogenase, galectin 1 and galectin
3) or that have been characterized as potential autoantigens in rheumatoid arthritis (e.g. BiP, colligin, HC gp-39). A novel
uncharacterized protein product of chromosome 19 open reading frame 10 was also detected as an apparently major component
of FLS cells. These results demonstrate the utility of high-content proteomic approaches in the analysis of FLS composition. 相似文献

The coupling reaction of acetoxymethoxy ribofuranoside 4 with nucleic acid bases 5a-f to synthesize novel (ribofuranosyloxy)methyl uracil, thymine, cytosine, adenine, guanine derivatives 6a-g respectively in preference to the expected formation of natural nucleosides 2',3',5'-tri-O-benzoyl uridine, methyluridine, cytidine, adenosine and guanosine 7a-g is described. Detailed study of these reactions catalysed by Lewis acids TMSOTf and SnCl4 is described. TMSOTf exhibited selectivity for the formation of ribofuranosyloxy methyl derivatives 6a-g rather than 7a-g. The ability to control conformational properties of polypeptides in their films is of considerable interest for many possible applications of these materials. By rational choice of the solvent system for film fabrication, control over the conformation of the main chain, the intermolecular hydrogen bonding in the side chain is easily achieved in poly(alpha-L-glutamic acid) (PLGA) thin films. The spectral data from circular dichromism (CD), FT-IR, and solid state (13)C NMR spectroscopies suggest that the beta-sheet conformation is dominant in PLGA films cast from trifluoroacetic acid (TFA) solution, whereas the right-handed alpha-helix is dominant in those cast from pyridine or DMF solution. In comparison with films cast from TFA solutions, the films fabricated from pyridine or DMF solutions exhibit strong intermolecular hydrogen bondings between -COOH groups and have a more ordered arrangement of side chains. Moreover, the extent of alpha-helix conformation of the PLGA backbone in films cast from pyridine or DMF solution is several times higher than that observed in the PLGA powder precipitated from aqueous solution at pH 4. All spectroscopic studies indicate clearly that the solvents (used for casting these films) play a crucial role in directing the organization of PLGA in these thin films. 相似文献

The mucoadhesive properties of chitosan microspheres prepared by different method were evaluated by studying the interaction
between mucin and microspheres in aqueous solution. The interaction was determined by the measurement of mucin adsorbed on
the microspheres. A strong interaction between chitosan microspheres and mucin was detected. The intensity of the interaction
was dependent upon the method of preparation of chitosan microspheres and the amount of mucin added. The extent of mucus adsorption
was proportional to the absolute values of the positive zeta potential of chitosan microspheres. The zeta potential in turn
was found to be dependent upon the method of preparation of microspheres. The adsorption of type III mucin (1% sialic acid
content) was interpreted using Freundlich or Langmuir adsorption isotherms. The values ofr
2 were greater for Langmuir isotherm as compared with Freundlich isotherm. The adsorption of a suspension of chitosan microspheres
in the rat small intestine indicated that chitosan microspheres prepared by tripolyphosphate cross-linking and emulsification
ionotropic gelation can be used as an excellent mucoadhesive delivery system. The microspheres prepared by glutaraldehyde
and thermal cross-linking showed good stability in HC1 as compared with microspheres prepared by tripolyphosphate and emulsification

This study aims to investigate the effect of the ice nucleation temperature on the primary drying process using an ice fog
technique for temperature-controlled nucleation. In order to facilitate scale up of the freeze-drying process, this research
seeks to find a correlation of the product resistance and the degree of supercooling with the specific surface area of the
product. Freeze-drying experiments were performed using 5% wt/vol solutions of sucrose, dextran, hydroxyethyl starch (HES),
and mannitol. Temperature-controlled nucleation was achieved using the ice fog technique where cold nitrogen gas was introduced
into the chamber to form an “ice fog”, there-by facilitating nucleation of samples at the temperature of interest. Manometric
temperature measurement (MTM) was used during primary drying to evaluate the product resistance as a function of cake thickness.
Specific surface areas (SSA) of the freeze-dried cakes were determined. The ice fog technique was refined to successfully
control the ice nucleation temperature of solutions within 1°C. A significant increase in product resistance was produced
by a decrease in nucleation temperature. The SSA was found to increase with decreasing nucleation temperature, and the product
resistance increased with increasing SSA. The ice fog technique can be refined into a viable method for nucleation temperature
control. The SSA of the product correlates well with the degree of supercooling and with the resistance of the product to
mass transfer (ie, flow of water vapor through the dry layer). Using this correlation and SSA measurements, one could predict
scaleup drying differences and accordingly alter the freeze-drying process so as to bring about equivalence of product temperature
