The pH for the optimum activity of amylase from the midgut of the third-instar larvae and adults ofSarcophaga ruficornis andMusca domestica ranged from pH 6.5 to 7.0 and 7.0 to 7.5, respectively. Maximum activity of the enzyme was observed at 50°C and the activity increased with increasing time period and enzyme concentration. Amylase from the midgut of the adult flies was more active than that from the larvae.

Summary The ultrastructure of corpora lutea obtained during the preimplantation, implantation and early postimplantation periods has been studied in 20 western spotted skunks. Fine structure of granulosa lutein cells was correlated with progesterone levels. The corpus luteum of the prolonged (7 month) preimplantation period contained undifferentiated small granulosa cells and differentiated large granulosa lutein cells. The former ranged in size between 12 and 20 and the latter between 20 and 45 . The ratio of small and large cells was about equal in an animal 2 days prior to nidation whereas only few small cells and numerous large cells were observed in an animal estimated to be 8 to 12 hours from nidation. Occasionally small cells were observed amidst large ones during the 24 hour nidation period, i.e. adhesion of trophoblast with the luminal uterine epithelium, but small cells were absent in animals after this period. Small cells had some smooth and rough endoplasmic reticulum, rod-shaped mitochondria with platelike cristae, small Golgi complex, and relatively smooth plasma membranes. Large lutein cells had abundant smooth endoplasmic reticulum, membranous whorls of smooth endoplasmic reticulum, usually round mitochondria with tubular and lamellar cristae, a well developed Golgi complex, variable amounts of lipid droplets, and highly plicated and ruffled plasma membranes. Peripheral plasma progesterone levels during the prolonged preimplantation period ranged between 1.1 and 7.9 ng/ml, but during implantation it was between 8 and 16.6 ng/ml. It is suggested that plasma progesterone levels fluctuate during the time of implantation and should not be regarded as a basis to predict actual nidation in the western spotted skunk.This research was supported in part by Grant Number HD06556 from the National Institute of Child Health and Human Development. 相似文献

Summary Despite the fact that HeLa and HEp-2 cell lines have been established respectively from a female and a male patient, the frequent loss of the Y chromosome makes it difficult to discriminate the two cell types. The technique of centromeric localization of heterochromatin, however, facilitates more definite identification of the two cell types. Giemsa positive centromeric constitutive heterochromatin blocks were induced within the intact metaphase chromosome preparations of Hela and HEp-2 cells. The Hela cell population had a single heterochromatin marker chromosome, whereas the HEp-2 cell line was found to possess two types of heterochromatin markers. The distinctive patterns of distribution of constitutive heterochromatin of these three chromosomes served as the guides in tracing the presumptive cytological pathways for their orgin.

Der Nachweis des zentromernahen Heterochromatins jedoch ermöglicht eine zuverlässigere Erkennung dieser beiden Zelltypen. Die Hela-Zellpopulation hat ein einzelnes, durch das Heterochromation gekennzeichnetes Marker-Chromosom, während die HEp-2-Zellinie deren 2 besitzt. Das distinkte Verteilungsmuster des konstitutiven Heterochromatons dieser 3 Chromosomen dient dazu, die vermutliche cytologische Herkunft zu interpretieren.

Summary 2 male patients with G-trisomic Down's syndrome, were studied for constitutive heterochromatin pattern by the technique of Arrighi and Hsu (1971). 2 G-chromosomes were found to have heterochromatin distributed around the centromeric areas and the remaining 3 were free of such localized heterochromatin blocks; the extra G-chromosome belonging to the non-localized heterochromatin members.

The sex chromosomes of the opossum, Didelphys virginiana, are the only elements that exhibit C-banding. In contrast, the sex chromosomes as well as the autosomes bear specific G-Bands. However, unlike other mammalian species different types of G-banding are observed if the chromosomes are pretreated with trypsin and SSC solution The SSC-pretreated chromosomes show discrete bands only when stained with Giemsa at certain pH values. An asynchronous pattern of terminal DNA replication is observed among the three C-banding regions of the X-chromosome. The inter- and intrapositive G-banding areas of the chromosomes are not always late in DNA replication in comparison to those negatively stained G-banding areas. 相似文献

Abstract— The characteristics of a rapidly labelled and rapidly transported neuronal perikaryal protein fraction (Rose & Sinha . 1974a) were investigated in three experiments. (1) The kinetics of labelling of neuronal cell body and neuropil fractions from [3H]fucose were followed and shown to be similar to those from [3H]lysine, the label first appearing in the neuronal fraction and then being exported. The neuronal/neuropil incorporation ratio fell from 1.37 at 1 h to 0.77 at 4 h. (2) When cycloheximide (5 mg/kg) was injected intraperitoneally 15 min after [3H]lysine, incorporation into neuronal protein was inhibited to a greater extent (85%) than into neuropil (60%). (3) Colchicine was injected at a dose (40 μg/kg) sufficient to prevent accumulation of radioactively labelled protein into synaptosomes but insufficient to affect total incorporation of precursor into protein. [3H]Lysine was injected 1 h after colchicine and neurons and neuropil fractions made 1 h and 4 h later; colchicine inhibited the export of labelled protein from the neuronal perikaryon and its accumulation in the neuropil. We conclude that the rapidly labelled neuronal protein is partially glycoprotein in character and may be normally transported from the cell body by way of the axonal/(dendritic?) flow mechanism. 相似文献

Adenosine 3',5'-cyclic monophosphate (cyclic AMP) phsophodiesterase activity in mouse neuroblastoma cells in culture markedly increased during exponential growth and reached a maximal level at confluency; whereas guanosine 3'5'-cyclic monophosphate (cyclic GMP) phosphodiesterase activity only slightly but significantly increased under a similar experimental condition. The increase in cyclic AMP phosphodiesterase activity was blocked by both cycloheximide and dactinomycin, whereas the increase in cyclic GMP phosphodiesterase was blocked by only cycloheximide. When the confluent cells were replated at low density, the cyclic nucleotide phosphodiesterase activity decreased; however, when they were plated at high cell density which equaled confluency, the enzyme activity did not decrease. Unlike cyclic AMP phosphodiesterase activity, cyclic GMP phosphodiesterase activity did not change significantly in prostaglandin E1-treated cells, but decreased in cells treated with the inhibitor of phosphodiesterase. Like cyclic AMP phosphodiesterase activity, cyclic GMP phosphodiesterase activity also did not change in cells treated with serum-free medium, X-irradiation, sodium butyrate and 6-thioguanine. 相似文献

Summary Post-incubation fractionation of soils incubated with C14-tagged oat roots under aerobic and anaerobic conditions and chromatographic separation of hydrolysates of different organic matter fractions indicated the incorporation of C14-labelled amino acids in soil organic matter. Anaerobic incubation leads to the formation of hydrolysable heavily C14-labelled organic substances in greater quantity. The amino acid composition of the different fractions revealed not a very significant qualitative difference. The significance and causes of stabilization of amino acids in soil organic matter are discussed. 相似文献
