Host resistance to “yellow dwarf” or “moonlight” disease cause by any population (Hg type) of Heterodera glycines I., the soybean cyst nematode (SCN), requires a functional allele at rhg1. The host resistance encoded appears to mimic an apoptotic response in the giant cells formed at the nematode feeding site about 24–48 h after nematode feeding commences. Little is known about how the host response to infection is mediated but a linked set of 3 genes has been identified within the rhg1 locus. This study aimed to identify the role of the genes within the locus that includes a receptor-like kinase (RLK), a laccase and an ion antiporter. Used were near isogeneic lines (NILs) that contrasted at their rhg1 alleles, gene-based markers, and a new Hg type 0 and new recombination events. A syntenic gene cluster on Lg B1 was found. The effectiveness of SNP probes from the RLK for distinguishing homolog sequence variants on LgB1 from alleles at the rhg1 locus on LgG was shown. The resistant allele of the rhg1 locus was shown to be dominant in NILs. None of the recombination events were within the cluster of the three candidate genes. Finally, rhg1 was shown to reduce the plant root development. A model for rhg1 as a dominant multi-gene resistance locus based on the developmental control was inferred.  相似文献

Pre-planting irrigation and planting patterns are important factors in weed management that effect on seed bank. Additionally, the nitrogen is the most important factor in plant growth that affects weed-crop competition and ultimately, seed rain into the soil. A field experiment was conducted to study the effect of nitrogen application rates, pre-planting irrigation and maize planting patterns on weed seed bank population. Experimental factors were nitrogen rates at 4 levels (200, 300, 400 and 500 kg per hectare) as main plot; and pre-planting irrigation at 2 levels (irrigation before planting plus weeding emerged seedlings and, irrigation after sowing), and maize planting patterns (one-row and two-row planting of maize with same density per square of row length) that were assigned in a factorial arrangement to the sub plots. Soil samples were taken at the beginning of the season (before planting of maize) and at the end of the season (after harvest) at depth of 0-5 cm in the fixed quadrates (60 cm x 60 cm). The weed seeds were extracted from the soil samples and were identified using standard methods. The majority of weed seed bank populations included 6 weed species: Portulaca oleracea, Chenopodium album, Amaranthus retroflexus, Sorghum halepense, Daturea stramonium, Xanthium strumarium. Results showed that population of weed seed bank increased significantly with increasing nitrogen rate. The increasing rate was different between one-row and two-row planting patterns. The parameters indicated that seed bank population was much higher in a one row planting pattern of maize. With two-row planting, seed bank was decreased by 34, 26, 20 and 5% at 200, 300, 400 and 500 kg N/ha, respectively. Pre-planting irrigation was also found an effective implement to reduce the weed seed bank. When pre-planting irrigation was applied, seed bank was decreased by 57, 43, 34 and 9% at 200, 300, 400 and 500 kg N/ha. Increasing nitrogen because of weed's better growth and higher seed production neutralized the decreasing effect of pre-planting irrigation and two-row planting of maize on weed seed bank population.  相似文献

Linum spp. from section Syllinum are promising for the production of aryltetralin lignans like podophyllotoxin (PTOX) and 6-methoxypodophyllotoxin (MPTOX). MPTOX is a PTOX congener that has cytotoxic activity comparable with PTOX. In this study root cultures of Linum Bungei from section Dasyllinum, L. strictum from section Linastrum, L. album, L. mucronatum ssp. mucronatum and L. nodiflorum from section Syllinum were established and their MPTOX levels were investigated in 1000 ml flasks. Root cultures of L. mucronatum ssp. mucronatum and L. nodiflorum were used to examine cell growth and production of MPTOX during a culture period of 36 days in 250 ml flasks. Considerable amounts of MPTOX in root cultures (1000 ml flasks) of L. album (6 mg/100 g DW), L. mucronatum ssp. mucronatum (770 mg/100 g DW) and L. nodiflorum (91 mg/100 g DW) were detected while it wasn't detected in root cultures of L. Bungei and L. strictum. In time course experiments, the maximum amount of MPTOX in L. nodiflorum root culture was at day 16 with 480 mg/ 100 g DW and the maximum amount of MPTOX in L. mucronatum ssp. mucronatum root culture was at day 12 with 130 mg/100 g DW. The results showed that root cultures of Linum species from section Syllinum are rich sources of MPTOX and since this lignan has remarkable cytotoxic activity, it can be used as a precursor for the production of antitumor agents.  相似文献

In numerous tissues, contractility to certain characteristic agents is associated with cells other than muscle, e.g. actin-containing so-called myofibroblasts. Such cells are present in pulmonary interstitium of several mammalian species but their contractility has only been demonstrated from the proliferation of myofibroblasts in fibrotic lung tissues. This study has been done to evaluate such responses in normal tissues in contrast with fibrotic lungs and it was necessary to take account also of smooth muscle reactivity. Distinctive agonists: mepyramine and sodium tungstate characterised myofibroblast mediated contractility, in contrast with acetylcholine and barium chloride as specific smooth muscle stimulants. Histology and immunohistochemistry were used to evaluate the pharmacological results more precisely.  相似文献

Effects of the intraperitoneal injection of quercetin in streptozocin-induced diabetic and normal rats were investigated and compared. Although quercetin had no effect on plasma glucose level of normal animals, it significantly and dose-dependently decreased the plasma glucose level of streptozocin-induced diabetic rats. Glucose tolerance tests of the diabetic animals approached those of normal rats, their plasma cholesterol and triglycerides were reduced significantly, while their hepatic glucokinase activity was significantly increased upon quercetin treatment. In normal rats, quercetin did not affect the glucose tolerance test, but resulted in an increase of plasma cholesterol and triglycerides and a decrease in hepatic glucokinase activity. No significant pathologic changes were noted in hepatocytes or kidney tubules and glomeruli, while the number of pancreatic islets significantly increased in both treated normal and diabetic groups. It is concluded that quercetin, a flavonoid with antioxidant properties brings about the regeneration of the pancreatic islets and probably increases insulin release in streptozocin-induced diabetic rats; thus exerting its beneficial antidiabetic effects. However, it may be of little value in normoglycemic animals.  相似文献

Biomolecules are increasingly attractive templates for the synthesis of functional nanomaterials. Chief among them is the plant tobacco mosaic virus (TMV) due to its high aspect ratio, narrow size distribution, diverse biochemical functionalities presented on the surface, and compatibility with a number of chemical conjugations. These properties are also easily manipulated by genetic modification to enable the synthesis of a range of metallic and non-metallic nanomaterials for diverse applications. This article reviews the characteristics of TMV and related viruses, and their virus-like particle (VLP) derivatives, and how these may be manipulated to extend their use and function. A focus of recent efforts has been on greater understanding and control of the self-assembly processes that drive biotemplate formation. How these features have been exploited in engineering applications such as, sensing, catalysis, and energy storage are briefly outlined. While control of VLP surface features is well-established, fewer tools exist to control VLP self-assembly, which limits efforts to control template uniformity and synthesis of certain templated nanomaterials. However, emerging advances in synthetic biology, machine learning, and other fields promise to accelerate efforts to control template uniformity and nanomaterial synthesis enabling more widescale industrial use of VLP-based biotemplates.  相似文献

B cell lymphoma 2 (Bcl-2) homology domain 3 (BH3)–only proteins of the Bcl-2 family are important functional adaptors that link cell death signals to the activation of Bax and/or Bak. The BH3-only protein Nbk/Bik induces cell death via an entirely Bax-dependent/Bak-independent mechanism. In contrast, cell death induced by the short splice variant of Bcl-x depends on Bak but not Bax. This indicates that Bak is functional but fails to become activated by Nbk. Here, we show that binding of myeloid cell leukemia 1 (Mcl-1) to Bak persists after Nbk expression and inhibits Nbk-induced apoptosis in Bax-deficient cells. In contrast, the BH3-only protein Puma disrupts Mcl-1–Bak interaction and triggers cell death via both Bax and Bak. Targeted knockdown of Mcl-1 overcomes inhibition of Bak and allows for Bak activation by Nbk. Thus, Nbk is held in check by Mcl-1 that interferes with activation of Bak. The finding that different BH3-only proteins rely specifically on Bax, Bak, or both has important implications for the design of anticancer drugs targeting Bcl-2.  相似文献

In this study, the possibility of keratin extraction from wool and feather by an enzymatic treatment along with a reducing agent has been investigated. The effects of different parameters, that is, enzyme loading, type of substrate and surfactant, hydrolysis time, and reducing agent concentration, have been examined in order to optimize the enzymatic hydrolysis. The optimal condition for maximum keratin extraction was attained by making use of 1 g/L sodium dodecyl sulfate (an anionic surfactant) and 2.6% (v/v) protease (Savinase), along with 8.6 and 6.4 g/L sodium hydrogen sulfite (a reducing agent) for wool and feathers, respectively, at liquor to fiber ratio of 25 mL/g for 4 hr. The obtained results indicated higher degradation of wool fiber in comparison with feathers, which might be due to the higher hydrophilic nature of the former. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) patterns revealed that the molecular weights of the extracted proteins from wool and feather were lower than those for the untreated fibers. Scanning electron micrographs showed fibers fibrillation and degradation upon enzymatic treatment. Besides, Fourier-transform infrared (FTIR) spectra indicated no evident changes in the chemical structure of the hydrolyzed fibers. However, wool and feather remainders were mostly composed of α-helix and β-sheets conformations, respectively.  相似文献
