The suggested location of broadleaved evergreen trees in Europe during the last full-glacial has traditionally favoured a southerly refugial model, which proposes survival in the Mediterranean peninsulas and recolonization of central and northern Europe during the Holocene. This hypothesis is not always substantiated by thorough reviews of original past and modern occurrence data, or considered in the light of plant traits and autoecology. Our approach focuses on the genus Buxus with the aim of exploring (i) the relationship between the location of refugia and post-glacial population dynamics, (ii) past processes determining density, fragmentation and local extinctions of modern populations, and (iii) the vulnerability of Buxus in the context of the undergoing environmental changes. We compiled a database of over 3600 modern occurrences and 676 fossil sites to reconstruct the distribution of Buxus in Europe since 30 ka cal BP. The location of fossil finds and the plant traits of Buxus indicate that it persisted widely across its modern distribution through the last glacial period with modes varying from region to region. The E Pyrenees, W Alps, and Jura Mts hosted dense populations, which expanded exponentially during the whole Holocene, and resulted in a modern continuous distribution area. In contrast, the Mediterranean Peninsulas hosted sparse populations, which increased exponentially only during the first half of the Holocene, clearly decreased in the last 4.5 ka BP and resulted in a highly fragmented modern distribution area, most likely in relation to the climate trends towards dry conditions of the last few millennia. These results challenge the common view that the Mediterranean regions are the exclusive and most important refuge areas for evergreen broadleaved trees and stress the importance of considering long-term population dynamics based on fossil data to evaluate the vulnerability of modern fragmented plant populations in view of conservation actions. 相似文献

The formation of epithelial tissues requires both the generation of apical-basal polarity and the coordination of this polarity between neighbouring cells to form a central lumen. During de novo lumen formation, vectorial membrane transport contributes to the formation of a singular apical membrane, resulting in the contribution of each cell to only a single lumen. Here, from a functional screen for genes required for three-dimensional epithelial architecture, we identify key roles for synaptotagmin-like proteins 2-a and 4-a (Slp2-a/4-a) in the generation of a single apical surface per cell. Slp2-a localizes to the luminal membrane in a PtdIns(4,5)P(2)-dependent manner, where it targets Rab27-loaded vesicles to initiate a single lumen. Vesicle tethering and fusion is controlled by Slp4-a, in conjunction with Rab27/Rab3/Rab8 and the SNARE syntaxin-3. Together, Slp2-a/4-a coordinate the spatiotemporal organization of vectorial apical transport to ensure that only a single apical surface, and thus the formation of a single lumen, occurs per cell. 相似文献

Population structure and landscape genetics in the endangered subterranean rodent Ctenomys porteousi

In order to devise adequate conservation and management strategies for endangered species, it is important to incorporate
a reliable understanding of its spatial population structure, detecting the existence of demographic partitions throughout
its geographical range and characterizing the distribution of its genetic diversity. Moreover, in species that occupy fragmented
habitats it is essential to know how landscape characteristics may affect the genetic connectivity among populations. In this
study we use eight microsatellite markers to analyze population structure and gene flow patterns in the complete geographic
range of the endangered rodent Ctenomys porteousi. Also, we use landscape genetics approaches to evaluate the effects of landscape configuration on the genetic connectivity
among populations. In spite of geographical proximity of the sampling sites (8–27 km between the nearest sites) and the absence
of marked barriers to individual movement, strong population structure and low values of gene flow were observed. Genetic
differentiation among sampling sites was consistent with a simple model of isolation by distance, where peripheral areas showed
higher population differentiation than those sites located in the central area of the species’ distribution. Landscape genetics
analysis suggested that habitat fragmentation at regional level has affected the distribution of genetic variation among populations.
The distance of sampling sites to areas of the landscape having higher habitat connectivity was the environmental factor most
strongly related to population genetic structure. In general, our results indicate strong genetic structure in C. porteousi, even at a small spatial scale, and suggest that habitat fragmentation could increase the population differentiation. 相似文献

Polyphenol oxidase (PPO) is a ubiquitous enzyme important in the food industry. Although PPO activity followed Michaelis?CMenten kinetics at catechol concentrations of up to 1?mM, it slowly decreased at catechol concentrations above 2?mM. This result indicated that in addition to the active site (site A), the enzyme possesses a second catechol-binding site (site B) that exerts an inhibitory effect on PPO activity. Halides inhibit PPO activity in such a way that substrate inhibition is lessened when halide concentration is increased. Furthermore, elevated concentrations of catechol diminished the degree of inhibition by halides. These findings suggest that halides also bind to site B to inhibit PPO activity. A steady-state kinetic analysis demonstrated that the dissociation constant between catechol and PPO depended on the binding of halides to site B. The dissociation constants were greatest when chloride bound to the site. Bromide and iodide yielded lower dissociation constants, in that order. These data indicate that the binding of halide to site B modulated the structure of site A, thereby exerting an inhibitory effect. 相似文献

Background and aimComplications of diabetes comprise the leading cause of death in Mexico. We aimed to describe the characteristics of management and achievement of therapeutic targets in Mexican patients with diabetes mellitus.MethodsWe analyzed data from 2642 Mexican patients with type 1 (T1D, n = 203, 7.7%) and type 2 diabetes (T2D, n = 2439, 92.3%) included in the third wave of the International Diabetes Management Practices Study.ResultsOf T2D patients, 63% were on oral glucose-lowering drugs (OGLD) exclusively (mostly metformin), 11% on insulin, 22% on OGLD plus insulin, and 4% on diet and exercise exclusively. T2D patients on insulin were more likely to be trained on diabetes, but they were older, had worse control, longer disease duration and more chronic complications than patients on OGLD only. Glycated hemoglobin (HbA1c) < 7% was achieved by 21% and 37% of T1D and T2D patients, respectively. Only 5% of T1D and 3% of T2D attained the composite target of HbA1c < 7%, blood pressure < 130/80 mmHg and low-density lipoprotein cholesterol < 100 mg/dl. T1D patients had less macrovascular but more microvascular complications, compared with T2D patients. Late complications increased with disease duration, so that about 80% of patients after 20 years of diagnosis have at least one late complication. Reaching the target HbA1c < 7% was associated with a reduced number of microvascular but not with less macrovascular complications.ConclusionA great proportion of these Mexican patients with diabetes did not reach therapeutic targets. In the last 50 years, obesity has become a global epidemic and is one of the main public health problems in many parts of the world. Adolescence is a critical period regarding weight control. The factors determining obesity include a complex group of interrelated biological, behavioral and environmental factors which reinforce each other. In children and adolescents, obesity is associated with premature cardiovascular diseases, diabetes mellitus type 2, acanthosis nigricans, respiratory and skeletal muscle problems, as well as psychological problems. The clinical manifestations of cardiovascular disease begin in middle age. Nevertheless, studies indicate that the atherosclerotic process begins to develop during childhood. Postprandial hyperlipemia is a physiological process that occurs several times a day after the complete absorption of a diet including lipids and has been suggested as a risk factor for coronary heart disease (CHD). New study areas include the effects of different fatty acids, lipid sources (endogenous and exogenous), and the effect ingesting alcoholic beverages during meals. Given the evidence that postprandial lipidemia is an independent risk factor for CHD, it is vital to establish normative values for children and adolescents such that more effective and efficient preventive and therapeutic measures can be adopted. 相似文献

A newly reorganized HUPO Cardiovascular Initiative was announced at the HUPO 2011 Cardiovascular Initiative Workshop at Geneva. The new initiative is now part of the biology- and disease-driven component of the HUPO Human Proteome Project (B/D-HPP).
