The objectives of the study, conducted during the 2003/2004 growing season in the National Park of Bou Hedma (South Tunisia), were to quantify the effects of the single-woody species Acacia tortilis subsp. raddiana on grass species composition, on total plant cover, on density of perennial species, on dry matter (DM) yield and on soil nutrients at lightly and heavily grazed sites. In each study site, two subhabitats were distinguished, i.e. under tree canopies and open grasslands. In the lightly grazed site, the nutrient status of soil (organic matter, total N, extractable P, K+, Ca2+, Na+, Mg2+) under Acacia raddiana canopy, was found to be significantly higher (p<0.05) than under the open grassland. In the same way, total plant cover (p<0.05), density of perennial species (p<0.01) and DM yield (p<0.01) were significantly higher under tree canopies in the lightly grazed site. Heavy grazing proved to exert a strong overriding effect over the positive influences of the woody plants. For most studied parameters, a non-significant difference was recorded between canopied and uncanopied subhabitats. Some palatable species were frequently found under trees. In the heavily grazed site, these species are being replaced by less desirable species. This emphasizes the importance of conservation stocking rates and proper pasture management.  相似文献

Seeds of Acacia tortilis (Forsskal) Hayne ssp. raddiana (Save) Brenan, a savannah tree of great potential for forestry and an excellent browse for game and livestock, is heavily infested by the bruchid beetle Bruchidius raddianae Anton & Delobel. The development from egg to beetle, and the various development stages were investigated using Scanning Electron Microscopy (SEM). The adult B. raddianae lays eggs on the green pod in the autumn. The first instar larva hatches from five to seven weeks and develops outside the host seed. From the second instar onwards development took place inside the host seed. Pupation takes about three weeks, late in the summer. The beetle of B. raddianae is univoltine and the newly emerged adult makes an exit hole to leave the seed. During the development from first instar lava to imago, all embryonic tissue is destroyed. This results in a weakness of its soil seed stock, which reduces the possibilities of natural regeneration of the species.  相似文献

In this study, three adherent Staphylococcus strains able to use nitrate as electron acceptor were isolated from textile wastewater activated sludge. Based on the biochemical profiles, bacterial strains were identified as Staphylococcus lentus, Staphylococcus warneri and Staphylococcus epidermidis the PCR amplification of the nir genes reveal that S. lentus and S. epidermidis were nirK positive and that S. epidermidis was nirS positive. The three strains were also icaA/icaD positive. To obtain the optimal formulation of pure cultures of the staphylococci, the influence of the different mixtures of organisms was studied using mixture design. The regression model of microorganism composition and main metabolites was established. The most predictable reduction of nitrate was 87.2 and 12% of COD consumption. The results suggested that the predictable production of nitrite would reach a minimum of 6.1 mg/l. Based on this, the response values that satisfied all expectations were optimized using MINITAB® 14 analysis software. The most optimal proportion combination was 49.75% of S. lentus (curve value), 36.85% of S. warneri (curve value) and 13.39% S. epidermidis (curve value).  相似文献

Thirty-five Staphylococcus aureus strains from auricular infections were isolated. The identification of strains was confirmed by Api ID 32 Staph strips, the antibiotic susceptibility test was performed using ATB Staph kit. PCR assay was used to detect the oxacillin resistance gene (mecA) and the erythromycin genes (ermA, ermB, ermC, msrA and mef). The susceptibility profile of all strains revealed a low resistance level to oxacillin and erythromycin. The PCR results show that 60 % of the strains are mecA positive. The frequency of erythromycin genes was: ermA (+) 22.8 %, ermB (+) 45.7, ermC (+) 17.1, msrA (+) 28.6. The mef gene was not detected in any strain. No correlations between genotypic and phenotypic methods for the determination of oxacillin and erythromycin resistance was found. However, multiplex PCR technique was shown to be a fast, practical and economic technique for the detection of methicillin-and erythromycin-resistant staphylococci.  相似文献

Assessing differences in plant functional traits (PFTs) along climatic gradients is potentially useful for understanding variation within and across populations, and for predicting their responses to climate change. This study investigates the intraspecific variability of several PFTs in Stipa tenacissima (Alpha grass) seedlings from different populations distributed across a climatic gradient. Seven populations from Tunisia to Spain within a 100–600 mm/year rainfall range were selected. Seedlings from each population were grown in a common garden. We expected the functional characteristics to differ among seedling populations according to their climatic gradient. The response patterns were helpful to predict acclimation and fitness under future climatic conditions in these populations. The seedling development analysis showed differences in PFTs among S. tenacissima populations. The biomass traits analysis revealed that higher above-ground biomass was related to higher below-ground development. The leaf traits proved that seedlings with longer leaf length would have less sclerophyllous leaves, a trade-off between productivity and drought resistance. The root traits analysis reflects seedling strategies to maximize resource uptake efficiency. PFTs showed several significant relationships with climatic conditions. The less rainfall, the higher plant allocation to root systems exploring soil. Higher mean temperatures were related to reduced root/plant development. The PFT analysis proves that species followed the ‘optimal partitioning theory’, in that plants preferentially allocate biomass to acquire the resource that most limits their development. However, both the environmental conditions and genetic diversity in S. tenacissima populations influenced seedling growth and behaviour to face ongoing climate change.

An experimental study has been carried out on the germination properties of seeds of this strictly Saharan species. The regeneration ability of a Tunisian barley accession originated from Kerkena islands was monitored through somatic embryogenesis and organogenesis. To prevent or to reduce normal germination, longitudinally bisected as well as base-wounded mature caryopses were cultured on a modified Chée and Pool-based medium (CP) enriched with different phytohormonal combinations. The greatest embryogenesis response was obtained when base-wounded caryopses were cultured on CP enriched with 2 mg/l chlorophenoxyacetic acid + 2.5 mg/l kinetin (76.85%). The same combination coupled to longitudinally bisected caryopses led to the embryogenic induction at the hypocotyl base of the germinated caryopses (61.9%). Embryogenic calluses differentiated into globular, heart-shaped, torpedo, and fully differentiated stages of somatic embryos on hormone-free Murashige and Skoog-based medium. Rooted plantlets were successfully transferred to soil and grown to maturity in the greenhouse and produced fertile seeds within 3 mo. On the other hand, organogenesis was achieved on CP enriched with 2 mg/l 2,4-dichlorophenoxyacetic acid + 2.5 mg/l kinetin. Histological aspects and scanning electron microscopy of both regeneration methods confirmed further the embryogenic and organogenic nature of the established processes. This efficient plant regeneration system provides a foundation for generating transgenic plants and germplasm preservation of “Kerkena” barley accession.  相似文献

In the last decades, there has been a growing interest on evaluating the effects of biological soil crusts (hereafter, biocrusts) on vascular plants. The purpose of this paper was to investigate the influence of biocrusts on Stipa tenacissima, key species in arid and semi-arid environments of the Mediterranean Basin. In fact, we explored the impact of biocrusts removal underneath S. tenacissima on soil chemical properties and leaf mineral uptake. Indeed, we studied the effect of mixed biocrusts on the seed germination of S. tenacissima under laboratory conditions. The research showed a greater availability of soil resources correlated with enhanced leaf nutrient levels of bioessential elements in the presence of intact biocrusts compared to fragmented biocrusts underneath S. tenacissima tussocks. We suggest that this is related to the loss of soil moisture as a result of biocrusts removal. In addition, we found that mixed biocrusts favored seed germination and viability of S. tenacissima. By improving soil chemical properties and leaf nutrient uptake, biocrusts can provide enhanced growth conditions for Stipa tenacissima. These findings with the positive effect on seed germination suggest that biocrusts can be used in the restoration of S. tenacissima.  相似文献
