We previously identified a highly active homodimeric FMN-dependent NADH-preferred azoreductase (AzoA) from Enterococcus faecalis, which cleaves the azo bonds (R-N?N-R) of diverse azo dyes, and determined its crystal structure. The preliminary network-based mutational analysis suggested that the two residues, Arg-21 and Asn-121, have an apparent mutational potential for fine-tuning of AzoA, based on their beneficial pleiotropic feedbacks. However, epistasis between the two promising mutational spots in AzoA has not been obtained in terms of substrate binding and azoreductase activity. In this study, we further quantified, visualized, and described the pleiotropic and/or epistatic behavior of six single or double mutations at the positions, Arg-21 and Asn-121, as a further research endeavor for beneficial fine-tuning of AzoA. Based on this network-based mutational analysis, we showed that pleiotropy and epistasis are common, sensitive, and complex mutational behaviors, depending mainly on the structural and functional responsibility and the physicochemical properties of the residue(s) in AzoA.  相似文献

Background  

Many studies have suggested that variability in the attractiveness of humans to host-seeking mosquitoes is caused by differences in the make-up of body emanations, and olfactory signals in particular. Most investigations have either been laboratory-based, utilising odour obtained from sections of the body, or have been done in the field with sampling methods that do not discriminate between visual, physical and chemical cues of the host. Accordingly, evidence for differential attractiveness based on body emanations remains sparse in spite of the far-reaching epidemiological implications of this phenomenon.  相似文献

Nineteen filamentous fungi, isolated from estuarine sediments in Brazil, were screened for degradation of polycyclic aromatic hydrocarbons (PAH). The fungal isolates were incubated with pyrene. The cultures were extracted and metabolites in the extracts were detected by high performance liquid chromatography (HPLC) and u.v. spectral analyses. Six fungi were selected for further studies using [4,5,9,10-14C]pyrene. Cyclothyrium sp., Penicillium simplicissimum, Psilocybe sp., and a sterile mycelium demonstrated the ability to transform pyrene. Cyclothyrium sp. was the most efficient fungus, transforming 48% of pyrene to pyrene trans-4,5-dihydrodiol, pyrene-1,6-quinone, pyrene-1,8-quinone and 1-hydroxypyrene. This fungus was also evaluated with a synthetic mixture of PAH. After 192 h of incubation, Cyclothyrium sp. was able to degrade simultaneously 70, 74, 59 and 38% of phenanthrene, pyrene, anthracene and benzo[a]pyrene, respectively.  相似文献

Microcosm studies were conducted to evaluate the survival and performance of a recently discovered polycyclic aromatic hydrocarbon (PAH)-degrading Mycobacterium sp. when this organism was added to sediment and water from a pristine ecosystem. Microcosms inoculated with the Mycobacterium sp. showed enhanced mineralization, singly and as components in a mixture, of 2-methylnaphthalene, phenanthrene, pyrene, and benzo[alpha]pyrene. Studies utilizing pyrene as the sole added PAH showed that the Mycobacterium sp. survived in microcosms for 6 weeks both with and without preexposure to PAH and mineralized multiple doses of pyrene. Pyrene mineralization rates for sterilized microcosms inoculated with the Mycobacterium sp. showed that competition with indigenous microorganisms did not adversely affect survival of or pyrene degradation by the Mycobacterium sp. Pyrene mineralization by the Mycobacterium sp. was not enhanced by inorganic nutrient enrichment and was hindered by organic nutrient enrichment, which appeared to result from overgrowth of indigenous bacteria. This study demonstrates the versatility of the PAH-degrading Mycobacterium sp. and expands its potential applications to include the degradation of two-, three-, four-, and five-ringed PAHs in sediments.  相似文献

Microbiological analyses of sediments chronically exposed to petrogenic hydrocarbons resulted in the isolation of a gram-positive, rod-shaped bacterium which mineralized naphthalene (59.5% of the original amount), phenanthrene (50.9%), fluoranthene (89.7%), pyrene (63.0%), 1-nitropyrene (12.3%), 3-methylcholanthrene (1.6%), and 6-nitrochrysene (2.0%) to carbon dioxide when grown for 2 weeks in pure culture with organic nutrients. The bacterium tolerated salt concentrations up to 4% and grew well at 24 to 30 degrees C. The use of this bacterium may be an attractive alternative to existing physicochemical methods for the remediation of polycyclic aromatic hydrocarbons in the environment.  相似文献

The mobility of phospolipids in free-standing and supported membranes was investigated on the level of individual molecules. For the analysis of trajectories a new statistical treatment was developed that permitted us to clearly distinguish different types of diffusional motion. A freely diffusing subfraction of lipids within supported membranes was identified. Its mobility was characterized by a mean lateral diffusion constant of D(supp) = 4.6 &mgr;m(2)/s. In comparison, the mobility of lipids embedded in "free-standing" planar membranes yielded an increase in the mean diffusion constant by a factor of 4.5, D(free) = 20.6 &mgr;m(2)/s. This increase is attributed to the ultrathin (</=1 nm) lubricating water layer between membranes and glass support.  相似文献

Mycobacterium sp. strain PYR-1 degrades high-molecular-weight polycyclic hydrocarbons (PAHs) primarily through the introduction of both atoms of molecular oxygen by a dioxygenase. To clone the dioxygenase genes involved in PAH degradation, two-dimensional (2D) gel electrophoresis of PAH-induced proteins from cultures of Mycobacterium sp. strain PYR-1 was used to detect proteins that increased after phenanthrene, dibenzothiophene, and pyrene exposure. Comparison of proteins from induced and uninduced cultures on 2D gels indicated that at least six major proteins were expressed (105, 81, 52, 50, 43, and 13 kDa). The N-terminal sequence of the 50-kDa protein was similar to those of other dioxygenases. A digoxigenin-labeled oligonucleotide probe designed from this protein sequence was used to screen dioxygenase-positive clones from a genomic library of Mycobacterium sp. strain PYR-1. Three clones, each containing a 5,288-bp DNA insert with three genes of the dioxygenase system, were obtained. The genes in the DNA insert, from the 5' to the 3' direction, were a dehydrogenase, the dioxygenase small (beta)-subunit, and the dioxygenase large (alpha)-subunit genes, arranged in a sequence different from those of genes encoding other bacterial dioxygenase systems. Phylogenetic analysis showed that the large alpha subunit did not cluster with most of the known alpha-subunit sequences but rather with three newly described alpha subunits of dioxygenases from Rhodococcus spp. and Nocardioides spp. The genes from Mycobacterium sp. strain PYR-1 were subcloned and overexpressed in Escherichia coli with the pBAD/ThioFusion system. The functionality of the genes for PAH degradation was confirmed in a phagemid clone containing all three genes, as well as in plasmid subclones containing the two genes encoding the dioxygenase subunits.  相似文献

PCR procedures based on 16S rRNA gene sequences specific for 12 anaerobic bacteria that predominate in the human intestinal tract were developed and used for quantitative detection of these species in human (adult and baby) feces and animal (rat, mouse, cat, dog, monkey, and rabbit) feces. Fusobacterium prausnitzii, Peptostreptococcus productus, and Clostridium clostridiiforme had high PCR titers (the maximum dilutions for positive PCR results ranged from 10(-3) to 10(-8)) in all of the human and animal fecal samples tested. Bacteroides thetaiotaomicron, Bacteroides vulgatus, and Eubacterium limosum also showed higher PCR titers (10(-2) to 10(-6)) in adult human feces. The other bacteria tested, including Escherichia coli, Bifidobacterium adolescentis, Bifidobacterium longum, Lactobacillus acidophilus, Eubacterium biforme, and Bacteroides distasonis, were either at low PCR titers (less than 10(-2)) or not detected by PCR. The reported PCR procedure including the fecal sample preparation method is simplified and rapid and eliminates the DNA isolation steps.  相似文献

Three filamentous fungi were examined for the ability to biotransform phenanthrene to oxidative (phase I) and conjugative (phase II) metabolites. Phenanthrene metabolites were purified by high-performance liquid chromatography (HPLC) and identified by UV/visible absorption, mass, and1H NMR spectra.Aspergillus niger ATCC 6275,Syncephalastrum racemosum UT-70, andCunninghamella elegans ATCC 9245 initially transformed [9-14C]phenanthrene to produce metabolites at the 9,10-, 1,2-, and 3,4- positions. Subsequently, sulfate conjugates of phase I metabolites were formed byA. niger, S. racemosum, andC. elegans. Minor glucuronide conjugates of 9-phenanthrol and phenanthrenetrans-9,10-dihydrodiol were formed byS. racemosum andA. niger, respectively. In addition,C. elegans produced the glucose conjugates 1-phenanthryl -d-glucopyranoside and 2-hydroxy-1-phenanthryl -d-glucopyranoside, a novel metabolite. [9-14C]Phenanthrene metabolites were not detected in organic extracts from biotransformation experiments with the yeasts,Candida lipolytica 37-1,Candida tropicalis ATCC 32113, andCandida maltosa R-42.  相似文献

Bacillus cereus ATCC 14579 transformed naphthalene predominately to 1-naphthol. Experiments with [14C]naphthalene showed that over a 24 h period, B. cereus oxidized 5.2% of the added naphthalene. 1-Naphthol accounted for approximately 80% of the total metabolites. B. cereus incubated with naphthalene under the presence of 18O2 led to the isolation of 1-naphthol that contained 94% 18O. The metabolism of [1-2H]-and [2-2H]-naphthalene by B. cereus yielded 1-naphthol which retained 95% and 94% deuterium, respectively, as determined by mass spectral analysis. NMR spectroscopic analysis of the deuterated 1-naphthol formed from [1-2H]-naphthalene indicated an NIH shift mechanism in which 19% of the deuterium migrated from the C-1 to the C-2 position. The 18O2 and NIH shift experiments implicate naphthalene-1,2-oxide as an intermediate in the formation of 1-naphthol from naphthalene by B. cereus.Abbreviations HPLC High performance liquid chromatography - NMR nuclear magnetic resonance  相似文献
