Most terrestrial plants establish symbiotic associations with microorganisms that enable them to overcome abiotic or biotic filters in ecosystems. Here we investigated how aerial mutualisms involving invasive species may affect the recipient community’s structure. We hypothesized that the endophyte Epichloë occultans enhances the ability of Lolium multiflorum to establish and colonize, but that success would depend on the structure and invasion resistance of the recipient grassland community. Seeds of L. multiflorum with high (E+) and low (E?) endophyte incidence were sown in plots located in grasslands with or without recent grazing history. Relative cover of L. multiflorum and floristic groups was determined during the growing season. Whereas we did not detect any endophyte effect in sites with grazing history, L. multiflorum cover was 63 % in E+ and 27 % in E? plots in sites without grazing history. As cover of L. multiflorum increased in these sites, the cover of warm- and cool-season grasses decreased in spring, with that of warm-season grasses continuing to decrease in summer. These decreases corresponded to 1.9, 3.7 and 1.6 %, for every % increase of L. multiflorum cover. Path analysis and posterior modelling predicted a greater impact of the endophyte on L. multiflorum cover than of seed addition when resident L. multiflorum cover was ≤20 %. This effect decreased asymptotically as L. multiflorum cover increased beyond 20 %. Our results suggest that the endophyte may boost the invasion ability of L. multiflorum particularly in natural grassland without grazing history with potential longer-term consequences for community structure and dynamics.  相似文献

Ryanodine receptor (RyR2) is the major Ca2+ channel of the cardiac sarcoplasmic reticulum (SR) and plays a crucial role in the generation of myocardial force. Changes in RyR2 gating properties and resulting increases in its open probability (Po) are associated with Ca2+ leakage from the SR and arrhythmias; however, the effects of RyR2 dysfunction on myocardial contractility are unknown. Here, we investigated the possibility that a RyR2 mutation associated with catecholaminergic polymorphic ventricular tachycardia, R4496C, affects the contractile function of atrial and ventricular myocardium. We measured isometric twitch tension in left ventricular and atrial trabeculae from wild-type mice and heterozygous transgenic mice carrying the R4496C RyR2 mutation and found that twitch force was comparable under baseline conditions (30°C, 2 mM [Ca2+]o, 1 Hz). However, the positive inotropic responses to high stimulation frequency, 0.1 µM isoproterenol, and 5 mM [Ca2+]o were decreased in R4496C trabeculae, as was post-rest potentiation. We investigated the mechanisms underlying inotropic insufficiency in R4496C muscles in single ventricular myocytes. Under baseline conditions, the amplitude of the Ca2+ transient was normal, despite the reduced SR Ca2+ content. Under inotropic challenge, however, R4496C myocytes were unable to boost the amplitude of Ca2+ transients because they are incapable of properly increasing the amount of Ca2+ stored in the SR because of a larger SR Ca2+ leakage. Recovery of force in response to premature stimuli was faster in R4496C myocardium, despite the unchanged rates of recovery of L-type Ca2+ channel current (ICa-L) and SR Ca2+ content in single myocytes. A faster recovery from inactivation of the mutant R4496C channels could explain this behavior. In conclusion, changes in RyR2 channel gating associated with the R4496C mutation could be directly responsible for the alterations in both ventricular and atrial contractility. The increased RyR2 Po and fractional Ca2+ release from the SR induced by the R4496C mutation preserves baseline contractility despite a slight decrease in SR Ca2+ content, but cannot compensate for the inability to increase SR Ca2+ content during inotropic challenge.  相似文献

Niche differentiation among tropical forest plants can generate species turnover along gradients of soil, topography, climate, and land use history. In this study we explore the relative importance of these variables as drivers of floristic composition in Cueva de Los Guacharos National Park. We established twenty 0.1‐ha plots, within which trees, lianas, and shrubs (diameter ≥ 2.5 cm) were censused. We selected plot locations in primary and disturbed forests, and we measured topography and soil variables. Despite their structural similarity, primary and disturbed forests differed floristically, and also differed in environmental variables measured. A NMDS ordination showed that variation in the floristic composition across plots is highly correlated to the exchangeable acidity, elevation, temperature, and magnesium availability. Variance partitioning analysis shows that together spatial and environmental variables explain 24.2 percent of the variation in species composition. ‘Pure environmental’ variables were more important in explaining compositional variability than ‘pure spatial’ processes (9.8% and 1.4%, respectively). Residual variance may be attributed to stochastic process or non‐measured biotic effects.  相似文献

We report the oldest fossil evidence of osteophagia by terrestrial invertebrates on both the Asian and African continents. Bones attributable to the Middle Jurassic dinosaur Chuanjiesaurus (Dinosauria: Sauropoda) were found with post-mortem insect modification in the Chuanjie Formation, Yunnan Province, China. The morphology of the borings closely matches the ichnogenus Cubiculum. Based on the lack of bioglyphs observed in Cubiculum ornatus, a new ichnospecies is proposed here. The new trace fossil, Cubiculum inornatus isp. nov., is interpreted to have been constructed for pupation by an unknown taxon of insect. Additionally, we report even older borings from Early Jurassic dinosaur bones of the Elliott Formation in the Karoo Basin, which represent the second oldest occurrence of insect traces in bone from continental settings. Both trace fossils sites have palaeogeographic implications for the origins and dispersal of osteophagia amongst terrestrial invertebrates during the Mesozoic. These discoveries push back the antiquity of pupation in animal bones by more than 100 million years to the Middle Jurassic, indicating that this behaviour, and osteophagy more generally, originated early in the Mesozoic, roughly comparable with the origination of insect pupation in woody substrates (Late Triassic).  相似文献

HIV infection induces phenotypic and functional changes to CD8+ T cells defined by the coordinated upregulation of a series of negative checkpoint receptors that eventually result in T cell exhaustion and failure to control viral replication. We report that effector CD8+ T cells during HIV infection in blood and SIV infection in lymphoid tissue exhibit higher levels of the negative checkpoint receptor TIGIT. Increased frequencies of TIGIT+ and TIGIT+ PD-1+ CD8+ T cells correlated with parameters of HIV and SIV disease progression. TIGIT remained elevated despite viral suppression in those with either pharmacological antiretroviral control or immunologically in elite controllers. HIV and SIV-specific CD8+ T cells were dysfunctional and expressed high levels of TIGIT and PD-1. Ex-vivo single or combinational antibody blockade of TIGIT and/or PD-L1 restored viral-specific CD8+ T cell effector responses. The frequency of TIGIT+ CD4+ T cells correlated with the CD4+ T cell total HIV DNA. These findings identify TIGIT as a novel marker of dysfunctional HIV-specific T cells and suggest TIGIT along with other checkpoint receptors may be novel curative HIV targets to reverse T cell exhaustion.  相似文献
