The main characteristic of biosurfactants is their property of reducing the superficial and interfacial tension between two immiscible liquids of different polarities. The main obstacle to the application of biosurfactants is the high production costs, the use of alternative substrates being indicated to solve this problem. This work report the production of biosurfactant by Bacillus subtilis LB5a on a pilot scale using cassava wastewater as the substrate, and the study of the parameters related to its production. The cassava wastewater was heated, centrifuged and poured into a 40-liter batch pilot bioreactor adapted for simultaneous foam collection during the fermentative process. The temperature was maintained at 35 degrees C, agitation at 150 rpm and aeration 0.38 vvm during the first 12 h, and 0.63 vvm for the rest of the process. Samples of liquid fermentate were collected at regular intervals for the analysis of total carbohydrates, reducing sugars, pH, CFU/mL count and superficial tension. The foam was centrifuged and the biosurfactant purified. The kinetic data of the process showed that both the microbial population, which reached a maximum after about 24 h, and the foam production of 10.6 L, peaked between 24 and 36 h, coinciding with the greatest production of biosurfactant. The yield of semi-purified surfactant in the foam was 2.4 g/L. The superficial tension of the medium was reduced from 51 to 27 mN/m and the critical micellar concentration was 11 mg/L, which, in principle, characterizes it as a good tensoactive agent. As a function of its composition and productivity, cassava wastewater was identified as a good substrate for the production of the biosurfactant. 相似文献

In this work, the stabilizing effect of bovine serum albumin (BSA), peptone (PEP), and polyethylene glycol (PEG) during immobilization
of Candida antarctica lipase on activated carbon was investigated. The influence of enzyme concentration and type of additive, added during the
immobilization procedure, was studied using a 22 factorial central composite design. The goal was to maximize the synthetic activity of butyl butyrate, using butyric acid
and butanol as substrate in n-heptane. An increase of 31–58% in the esterification activity was obtained when enzyme concentration on the supernatant was
enhanced from 86.50 U m L−1 to 226.80 U mL−1. An enhancement in esterification activity of 38–68.95% was observed, depending on the initial enzyme concentration, when
PEP was used instead of BSA. No significant increase in the esterification activity was observed when PEP was replaced by

Over a one year period (November 2000-December 2001), clinical specimens from 189 dogs and 38 cats, from the city of Fortaleza, Ceará, Brazil, were examined at the Specialized Medical Mycology Center at the Federal University of Ceará to detect animals with dermatophytoses. The mycological analyses were conducted by direct microscopy and by fungal culture on Sabouraud agar, Sabouraud chloramphenicol agar and Mycosel agar. Dermatophytes were isolated from 27 of the 189 (14.3%) canine specimens and 14 of the 38 (36.8%) feline specimens. The identified dermatophytes were Microsporum canis (95%), M. gypseum (2.5%) and Trichophyton mentagrophytes var. mentagrophytes (2.5%). Microsporum canis was the most common species isolated (92.6% and 100%, for dogs and cats respectively). The percentage of positive direct microscopic examinations of clinical specimens and positive cultures was 61%. There was a high proportion of positive cultures from cats less than 1 year of age, but in dogs no significant differences were detected. There were no significant differences between the sexes. Dermatophytes were more frequently isolated in March, April and May, but no significant differences were detected in the seasonal distribution of canine and feline dermatophytoses. 相似文献

The concentrations of Cu, Zn, Pb, and Cd were determined in the mussel Mytella falcata from the Bacanga River estuary, in order to evaluate the potential role of domestic effluents released "in natura" on the quality of the estuarine environment with consequences to shellfish resources. Cu concentrations ranged from 5.2 mg.g-1 to 13.1 mg.g-1, while Zn concentrations ranged from 49.1 mg.g-1 to 76.3 mg.g-1. Pb and Cd showed concentrations lower than 2 mg.g-1. The results showed low concentrations of Cu, Pb and Cd in the filter-feeding Mytella falcata in the study area in comparison with the maximum values stablished to foods by the Brazilian Government (Decree Law No 55,871). Heparin is an excellent inhibitor of P- and L-selectin binding to the carbohydrate determinant, sialyl Lewis(x). As a consequence of its anti-selectin activity, heparin attenuates metastasis and inflammation. Here we show that fucosylated chondroitin sulfate (FucCS), a polysaccharide isolated from sea cucumber composed of a chondroitin sulfate backbone substituted at the 3-position of the beta-D-glucuronic acid residues with 2,4-disulfated alpha-L-fucopyranosyl branches, is a potent inhibitor of P- and L-selectin binding to immobilized sialyl Lewis(x) and LS180 carcinoma cell attachment to immobilized P- and L-selectins. Inhibition occurs in a concentration-dependent manner. Furthermore, FucCS was 4-8-fold more potent than heparin in the inhibition of the P- and L-selectin-sialyl Lewis(x) interactions. No inhibition of E-selectin was observed. FucCS also inhibited lung colonization by adenocarcinoma MC-38 cells in an experimental metastasis model in mice, as well as neutrophil recruitment in two models of inflammation (thioglycollate-induced peritonitis and lipopolysaccharide-induced lung inflammation). Inhibition occurred at a dose that produces no significant change in plasma activated partial thromboplastin time. Removal of the sulfated fucose branches on the FucCS abolished the inhibitory effect in vitro and in vivo. Overall, the results suggest that invertebrate FucCS may be a potential alternative to heparin for blocking metastasis and inflammatory reactions without the undesirable side effects of anticoagulant heparin. 相似文献

Several studies have reported the gastrointestinal (GI) effects promoted by the physical exercise. Thus, we aimed to evaluate the influence of swimming exercise on the contractile reactivity, lipid peroxidation and morphology of rat ileum. Wistar rats were divided into sedentary (SED) and groups exercised for two (EX2), four (EX4), six (EX6) or eight (EX8) weeks, 5 days/week. Animals were killed; the ileum was removed and suspended in organ baths where the isotonic contractions were recorded. Lipid peroxidation was evaluated by MDA (malondialdehyde) measurement with TBARS (thiobarbituric acid reactive substances) assay and morphology by histological staining. Cumulative concentration-response curves to KCl were attenuated, as the Emax values were changed from 100% (SED) to 63.1±3.9 (EX2), 48.8±3.8 (EX4), 19.4±1.8 (EX6) and 59.4±2.8% (EX8). Similarly, cumulative concentration-response curves to carbamylcholine hydrochloride (CCh) were attenuated, as the Emax values were changed from 100% (SED) to 74.1±5.4 (EX2), 75.9±5.2 (EX4) and 62.9±4.6 (EX6), but not in the EX8 (89.7±3.4%). However, CCh potency was increased in this latter, as the EC50 was altered from 1.0±0.1×10−6 (SED) to 2.1±0.4×10−7 (EX8). MDA concentration was altered only in EX4 (44.3±4.4) compared with SED (20.6±3.6 μmol/l). Circular layer was reduced in SED when compared with the exercised groups. Conversely, longitudinal layer was increased. In conclusion, chronic swimming exercise reduces the ileum contraction, equilibrates the oxidative damage and promotes changes in tissue size to establish an adaptation to the exercise. 相似文献

The human SIRT3 gene contains an intronic VNTR enhancer whose variability is correlated with life span. The SIRT3 5' flanking region encompasses the PSMD13 gene encoding the p40.5 regulator subunit of the 26S proteasome. Proteasome is a multicatalytic proteinase whose function declines with aging. SIRT3 and PSMD13 are linked in a head-to-head configuration (788-bp intergenic region). The molecular configuration of two genes that are both related to aging prompted us to search for shared regulatory mechanisms between them. Transfection experiments carried out in HeLa cells by deletion mutants of the PSMD13-SIRT3 intergenic region showed a complex pathway of coregulation acting in both directions. Furthermore, linkage disequilibrium (LD) analyses carried out in a sample of 710 subjects (18-108 years of age) screened for A21631G (marker of PSMD13), and for G477T and VNTR(intron5) (markers of SIRT3), revealed high LD, with significantly different PSMD13-SIRT3 haplotype pools between samples of centenarians and younger people. 相似文献

Opencast mining causes severe environmental impacts by removing the vegetation cover and depleting the fauna. Reforestation
methods using native species and diverse pre- and post-disturbance approaches aim to recover the original richness and diversity
of species found before the impact. Bioindicators are powerful tools to evaluate the restoration of the original environmental
conditions in disturbed areas. We used species richness, endemism and diversity measurements of Collembola to compare successional
stages in reforested sites of different ages compared with a control undisturbed area. Richness and abundance of Collembola
were subjected to correlation analysis with age of plots and vegetational variables. Areas that were reforested for up to
16 years supported a much lower Collembola species richness than undisturbed areas. Both the age of reforestation plots and
vegetation variables (number of trees, diameter of crowns, depth of leaf litter and tree species richness) were positively
and significantly correlated to collembolan abundance and richness. The results showed that the diversity of the 16-year-old
plot was significantly higher than that of younger areas, but significantly less diverse than that of the control area. Endemic
species were more sensitive to disturbance than non-endemics. Thus, species richness and diversity of soil Collembola can
be only partially restored with appropriate reforestation methods, and although it takes many years, to some extent even endemic
species can be gradually restored. Nevertheless, the maintenance of undisturbed diversity reservoirs linked by ecological
corridors to reforested plots is imperative, as only undisturbed areas can support most of the endemic species able to re-colonize

In tropical dry environments rainfall periodicity may affect demographic parameters, resulting in fluctuations in bird abundance. We used capture–recapture data for the Grey Pileated Finch from a Neotropical dry forest to evaluate the hypothesis that intra- and inter-annual survival, individuals entrance and population abundance, are related to local rainfall. Sampling occurred across 3 years, with individuals captured, tagged and evaluated for age and presence of brood patch every 14 days. Using the POPAN formulation, we generated demographic models to evaluate study population temporal dynamics. Best-fit models indicated a low apparent annual survival in the first year (16%) compared to other years (between 47 and 62%), with this low value associated with an extreme drought. The abundance of juveniles at each capture occasion was significantly dependent on the accumulated precipitation in the previous 14 days, and the juvenile covariate was a strong predictor of the intra-annual entrance probability (natality). Individuals entrance during the reproductive period corresponded to 53, 52 and 75% of total ingress for each year, respectively. The trend in sampled population size indicated positive exponential growth (Ninitial = 50, Nlast = 600), with intra-annual fluctuations becoming progressively more intense. Low survival was relevant during population decline at study onset, while at study end intense Individuals entrance promoted rapid population growth. Thus, the indirect effects of rainfall and the combined effect of two demographic rates operated synergistically on the immediate population abundance of Grey Pileated Finch, an abundant bird in a Neotropical dry forest. 相似文献
