The aim of this study was to evaluate the stabilisation of the wrist joint and the ad hoc wrist muscles activations during the two principal phases of the freestyle stroke. Seven male international swimmers performed a maximal semi-tethered power test. A swimming ergometer fixed on the start area of the pool was used to collect maximal power. The electromyography signal (EMG) of the right flexor carpi ulnaris (FCU) and extensor carpi ulnaris (ECU) was recorded with surface electrodes and processed using the integrated EMG (IEMG). Frontal and sagittal video views were digitised frame by frame to determine the wrist angle in the sagittal plane and the principal phases of the stroke (insweep, outsweep). Important stabilisation of the wrist and high antagonist muscle activity were observed during the insweep phase due to the great mechanical constraints. In outsweep, less stabilisation and lower antagonist activities were noted. Factors affecting coactivations in elementary movements, e.g. intensity and instability of the load, accuracy and economy of the movement were confirmed in complex aquatic movement. 相似文献

Mycosphaerella species are well-known causal agents of leaf diseases on many economically and ecologically important plant species. In Uruguay, a relatively large number of Mycosphaerellaceae and Teratosphaeriaceae are found on Eucalyptus, but nothing is known of these fungi on native Myrtaceae. The aim of this study was to identify Mycosphaerellaceae and Teratosphaeriaceae species associated with leaf diseases on native Myrtaceae in Uruguay and to consider whether host jumps by the pathogen from introduced Eucalyptus to native Myrtaceae have occurred. Several native forests throughout the country were surveyed with special attention given to those located close to Eucalyptus plantations. Five species belonging to the Mycosphaerellaceae and Teratosphaeriaceae clades were found on native Myrtaceous trees and three of these had previously been reported on Eucalyptus in Uruguay. Those occurring both on Eucalyptus and native Myrtaceae included Pallidocercospora heimii, Pseudocercospora norchiensis, and Teratosphaeria aurantia. In addition, Mycosphaerella yunnanensis, a species known to occur on Eucalyptus but not previously recorded in Uruguay, was found on leaves of two native Myrtaceous hosts. Because most of these species occur on Eucalyptus in countries other than Uruguay, it appears that they were introduced in this country and have adapted to be able to infect native Myrtaceae. Methylation is a post-translational modification that can affect numerous features of proteins, notably cellular localization, turnover, activity, and molecular interactions. Recent genome-wide analyses have considerably extended the list of human genes encoding putative methyltransferases. Studies on protein methyltransferases have revealed that the regulatory function of methylation is not limited to epigenetics, with many non-histone substrates now being discovered. We present here our findings on a novel family of distantly related putative methyltransferases. Affinity purification coupled to mass spectrometry shows a marked preference for these proteins to associate with various chaperones. Based on the spectral data, we were able to identify methylation sites in substrates, notably trimethylation of K135 of KIN/Kin17, K561 of HSPA8/Hsc70 as well as corresponding lysine residues in other Hsp70 isoforms, and K315 of VCP/p97. All modification sites were subsequently confirmed in vitro. In the case of VCP, methylation by METTL21D was stimulated by the addition of the UBX cofactor ASPSCR1, which we show directly interacts with the methyltransferase. This stimulatory effect was lost when we used VCP mutants (R155H, R159G, and R191Q) known to cause Inclusion Body Myopathy with Paget''s disease of bone and Fronto-temporal Dementia (IBMPFD) and/or familial Amyotrophic Lateral Sclerosis (ALS). Lysine 315 falls in proximity to the Walker B motif of VCP''s first ATPase/D1 domain. Our results indicate that methylation of this site negatively impacts its ATPase activity. Overall, this report uncovers a new role for protein methylation as a regulatory pathway for molecular chaperones and defines a novel regulatory mechanism for the chaperone VCP, whose deregulation is causative of degenerative neuromuscular diseases. 相似文献

The adenovirus E4orf4 protein selectively kills human cancer cells independently of p53 and thus represents a potentially promising tool for the development of novel antitumor therapies. Previous studies suggested that E4orf4 induces an arrest or a delay in mitosis and that both this effect and subsequent cell death rely largely on an interaction with the B55 regulatory subunit of protein phosphatase 2A. In the present report, we show that the death of human H1299 lung carcinoma cells induced by expression of E4orf4 is typified not by an accumulation of cells arrested in mitosis but rather by the presence of both tetraploid and diploid cells that are arrested in G1 because they are unable to initiate DNA synthesis. We believe that these E4orf4-expressing cells eventually die by various processes, including those resulting from mitotic catastrophe. 相似文献

Nanolipoprotein particles (NLPs) are nanometer‐scale discoidal particles that feature a phospholipid bilayer confined within an apolipoprotein “scaffold,” which are useful for solubilizing hydrophobic molecules such as drugs and membrane proteins. NLPs are synthesized either by mixing the purified apolipoprotein with phospholipids and other cofactors or by cell‐free protein synthesis followed by self‐assembly of the nanoparticles in the reaction mixture. Either method can be problematic regarding the production of homogeneous and monodispersed populations of NLPs, which also currently requires multiple synthesis and purification steps. Telodendrimers (TD) are branched polymers made up of a dendritic oligo‐lysine core that is conjugated to linear polyethylene glycol (PEG) on one end, and the lysine “branches” are terminated with cholic acid moieties that enable the formation of nanomicelles in aqueous solution. We report herein that the addition of TD during cell‐free synthesis of NLPs produces unique hybrid nanoparticles that have drastically reduced polydispersity as compared to NLPs made in the absence of TD. This finding was supported by dynamic light scattering, fluorescence correlation spectroscopy, and cryo transmission electron microscopy (Cryo‐EM). These techniques demonstrate the ability of TDs to modulate both the NLP size (6–30 nm) and polydispersity. The telodendrimer NLPs (TD‐NLPs) also showed 80% less aggregation as compared to NLPs alone. Furthermore, the versatility of these novel nanoparticles was shown through direct conjugation of small molecules such as fluorescent dyes directly to the TD as well as the insertion of a functional membrane protein. 相似文献

Splicing factor 1 (SF1) binds to the branch point sequence (BPS) of mammalian introns and is believed to be important for the splicing of some, but not all, introns. To help identify BPSs, particularly those that depend on SF1, we generated a BPS profile model in which SF1 binding affinity data, validated by branch point mapping, were iteratively incorporated into computational models. We searched a data set of 117,499 human introns for best matches to the SF1 Affinity Model above a threshold, and counted the number of matches at each intronic position. After subtracting a background value, we found that 87.9% of remaining high-scoring matches identified were located in a region upstream of 3'-splice sites where BPSs are typically found. Since U2AF65 recognizes the polypyrimidine tract (PPT) and forms a cooperative RNA complex with SF1, we combined the SF1 model with a PPT model computed from high affinity binding sequences for U2AF65. The combined model, together with binding site location constraints, accurately identified introns bound by SF1 that are candidates for SF1-dependent splicing. 相似文献

Probabilistic approaches for sequence alignment are usually based on pair Hidden Markov Models (HMMs) or Stochastic Context Free Grammars (SCFGs). Recent studies have shown a significant correlation between the content of short indels and their flanking regions, which by definition cannot be modelled by the above two approaches. In this work, we present a context-sensitive indel model based on a pair Tree-Adjoining Grammar (TAG), along with accompanying algorithms for efficient alignment and parameter estimation. The increased precision and statistical power of this model is shown on simulated and real genomic data. As the cost of sequencing plummets, the usefulness of comparative analysis is becoming limited by alignment accuracy rather than data availability. Our results will therefore have an impact on any type of downstream comparative genomics analyses that rely on alignments. Fine-grained studies of small functional regions or disease markers, for example, could be significantly improved by our method. Eucalyptus or guava rust caused by Puccinia psidii is a serious disease of Eucalyptus and other Myrtaceae. In Uruguay, it has been previously found on Eucalyptus globulus and Psidium brasiliensis. Almost nothing is known regarding the occurrence of this pathogen on other Eucalyptus species or native Myrtaceae in that country. In this study, we determined the presence of P. psidii on Eucalyptus species and native Myrtaceae trees in Uruguay and evaluated the pathogenicity of specimens from native myrtaceous hosts on
E. globulus and E. grandis. Phylogenetic analyses based on the internal transcribed spacer (ITS) region of the nuclear ribosomal DNA operon were used
to confirm pathogen identity. Comparisons of ITS sequences confirmed the identity of P. psidii on Eucalyptus globulus, E. grandis, Myrcianthes pungens, and Myrrhinium atropurpureum var. octandrum. This is the first report of P. psidii on M. atropurpureum var. octandrum. Pathogenicity tests showed that isolates from native Myrtaceae could infect both Eucalyptus species tested, indicating a strong biological relationship between both introduced and native Myrtaceae. relevant field data, morphological information, molecular phylogenetic analyses and infection studies that contribute to a
