Using CD11b knockout mice as a source of macrophages (Mphi;), we show that complement receptor 3 (CR3) mediates approximately 40-50% of nonopsonic binding and 50-60% of serum-mediated binding of Mycobacterium tuberculosis to resident Mphi;. We demonstrate that opsonic binding of M. tuberculosis to Mphi; is mediated by an immunoglobulin-independent, heat-labile component of serum, in both the presence and the absence of CD11b. The survival and replication of M. tuberculosis in an in vitro Mphi; model and an in vivo mouse model of infection were not significantly affected by the absence of CD11b, indicating that CR3-mediated uptake of M. tuberculosis is not a major factor in controlling the subsequent intracellular survival of the mycobacteria. However, whether a mycobacterium will gain access to the intracellular environment, and the type of Mφ that the bacterium enters, is significantly affected by the presence or absence of CR3.  相似文献

A possible method of control for the management of wild populations consists of continual introgression of an inducible transgene by releasing transgenic individuals, with periodic exposure of the population to a trigger. Exposure to the trigger causes death or sterility in carriers of the transgene, but is otherwise benign. We investigate the effectiveness of various strategies for control. We show that suppression of the population density below any pre-specified level is possible using this technique. At the same time we show that too frequent or too efficient exposure to the trigger can select for non-transgenic genotypes at an intensity such that the population density will be largely unaffected by the trigger. Choices for management parameters can ensure that the latter scenario is avoided. We show that releasing individuals carrying the transgene at more than one locus facilitates density control.  相似文献

Although plasmids play an important role in biological evolution, the number of plasmid families well-characterized in terms of geographical distribution and evolution remains limited, especially in archaea. Here, we describe the first systematic study of an archaeal plasmid family, the pT26-2 plasmid family. The in-depth analysis of the distribution, biogeography and host–plasmid co-evolution patterns of 26 integrated and 3 extrachromosomal plasmids of this plasmid family shows that they are widespread in Thermococcales and Methanococcales isolated from around the globe but are restricted to these two orders. All members of the family share seven core genes but employ different integration and replication strategies. Phylogenetic analysis of the core genes and CRISPR spacer distribution suggests that plasmids of the pT26-2 family evolved with their hosts independently in Thermococcales and Methanococcales, despite these hosts exhibiting similar geographic distribution. Remarkably, core genes are conserved even in integrated plasmids that have lost replication genes and/or replication origins suggesting that they may be beneficial for their hosts. We hypothesize that the core proteins encode for a novel type of DNA/protein transfer mechanism, explaining the widespread oceanic distribution of the pT26-2 plasmid family.  相似文献

Inflammatory bowel disease (IBD) is considered to be the most common cause of vomiting and diarrhoea in dogs, and the German shepherd dog (GSD) is particularly susceptible. The exact aetiology of IBD is unknown, however associations have been identified between specific single-nucleotide polymorphisms (SNPs) in Toll-like receptors (TLRs) and human IBD. However, to date, no genetic studies have been undertaken in canine IBD. The aim of this study was to investigate whether polymorphisms in canine TLR 2, 4 and 5 genes are associated with IBD in GSDs. Mutational analysis of TLR2, TLR4 and TLR5 was performed in 10 unrelated GSDs with IBD. Four non-synonymous SNPs (T23C, G1039A, A1571T and G1807A) were identified in the TLR4 gene, and three non-synonymous SNPs (G22A, C100T and T1844C) were identified in the TLR5 gene. The non-synonymous SNPs identified in TLR4 and TLR5 were evaluated further in a case-control study using a SNaPSHOT multiplex reaction. Sequencing information from 55 unrelated GSDs with IBD were compared to a control group consisting of 61 unrelated GSDs. The G22A SNP in TLR5 was significantly associated with IBD in GSDs, whereas the remaining two SNPs were found to be significantly protective for IBD. Furthermore, the two SNPs in TLR4 (A1571T and G1807A) were in complete linkage disequilibrium, and were also significantly associated with IBD. The TLR5 risk haplotype (ACC) without the two associated TLR4 SNP alleles was significantly associated with IBD, however the presence of the two TLR4 SNP risk alleles without the TLR5 risk haplotype was not statistically associated with IBD. Our study suggests that the three TLR5 SNPs and two TLR4 SNPs; A1571T and G1807A could play a role in the pathogenesis of IBD in GSDs. Further studies are required to confirm the functional importance of these polymorphisms in the pathogenesis of this disease.  相似文献

Life history theory predicts that females should vary their investment in offspring according to the quality of their mate. In birds, several studies have now shown that females do vary investment according to perceived male quality, by producing larger eggs, investing more in parental care or by manipulating the sex of their offspring. In a captive breeding colony of canaries, we first show that under normal conditions larger eggs in a clutch are more likely to hatch male offspring. In canaries, male song functions in female attraction and females respond more to complex structures in male song called sexy syllables. In a series of experiments, we go on to show first, that females exposed to playback of male song produce larger eggs than those who heard no song. Next, using synthetic songs, we show that females exposed to playback of more attractive songs containing sexy syllables, produced larger eggs than those exposed to simpler songs containing no sexy syllables. However, in a final analysis, we found no evidence from our experiments that females exposed to playback of more attractive songs also produced more male offspring.  相似文献

Nets with a small mesh size are required to catch Nephrops norvegicus, consequently large quantities of small whitefish are also caught, and much of this bycatch is undersized and is discarded dead. The main bycatch species are whiting (Merlangius merlangus), haddock (Melanogrammus aeglefinus) and cod (Gadus morhua). Here we summarize the known behavioural reactions of these species towards conventional trawls and review the results of using different trawl modifications to increase selectivity of Nephrops trawls. The trawl modifications are categorised as separator grids, separator and guiding panels, square-mesh panels, capture avoidance designs and codend modification. Finally, the extent to which these developments have been legislated for is discussed including the conditions under which new gear regulations have been introduced. Haddock and whiting rise during the trawling process facilitating their separation from Nephrops and escape, however the behaviour of small fish of these species is less consistent. Cod and Nephrops remain on bottom of the trawl, so to separate these species requires some physical filtering process. Overall, there is currently sufficient technical ability to improve selectivity in Nephrops trawls. The design of choice is dependent on the objectives of managers; for reducing discards but retaining marketable fish, square-mesh panels offer the most useful tool; to eliminate all bycatch and create a single-species fishery, grids and traditional Nephrops trawls show most potential. Whatever the objectives of the new measures, it is likely that a short-term economic impact will follow, and some form of incentive may be required to implement effective measures. A voluntary uptake of new measures by industry is preferable, however, to date, restrictions on fishing opportunities have been necessary to introduce innovative gear designs.  相似文献

Eight species of coccidia were recognized in 596 faeces samples from 3 commercial rabbitries in South East England. The level of infection was related to management methods and at one site it was reduced by an outbreak of mucoid enteritis and/or its treatment with oxytetracycline. In samples from rabbits managed conventionally in the wire cages over droppings-pits, 96% contained oocysts and of these, 60% had 1000-10000 oocysts/g. Mixed infections were common, 67% of the animals carrying 2-4 different species. Eimeria, media, E. magna and E. perforans occurred most frequently; E. coecicola, E. irresidua and E. flavescens were less common and E. intestinalis and E. piriformis were relatively rare. E. stiedai was not recorded.  相似文献

Age-related macular degeneration (AMD) is a leading cause of legal blindness in the Western world. There are effective treatments for the vascular complications of neo-vascular AMD, but no effective therapies are available for the dry/atrophic form of the disease. A previously described transgenic CFH-gene deficient mouse model, (cfh−/−), shows hallmarks of early AMD. The ocular phenotype has been further analysed to demonstrate amyloid beta (Aβ) rich basement membrane deposits associated with activated complement C3. Cfh−/− mice were treated systemically in both prophylactic and therapeutic regimes with an anti-Aβ monoclonal antibody (mAb), 6F6, to determine the effect on the cfh−/− retinal phenotype. Prophylactic treatment with 6F6 demonstrated a dose dependent reduction in the accumulation of both Aβ and activated C3 deposition. A similar reduction in the retinal endpoints could be seen after therapeutic treatment. Serum Aβ levels after systemic administration of 6F6 show accumulation of Aβ in the periphery suggestive of a peripheral sink mechanism. In summary, anti-Aβ mAb treatment can partially prevent or reverse ocular phenotypes of the cfh−/− mouse. The data support this therapeutic approach in humans potentially modulating two key elements in the pathogenesis of AMD – Aβ and activated, complement C3.  相似文献
