Aims The coastal Brazilian rainforest on white-sand (restinga) ranks among the most fragmented forest types in the tropics, owing to both the patchy distribution of sandy soils and widespread coastal development activities. Here we study the environmental and evolutionary determinants of a forest tree assemblage at a single restinga forest in Southeastern Brazil. We also explore the ability of competing hypotheses to explain the maintenance of species diversity in this forest type, which includes contrasting extremes of edaphic conditions associated with flooding stress.Methods The study was conducted in a white-sand forest permanent plot of 10.24 ha on the coastal plain of Southeastern Brazil. This plot was divided into 256 quadrats of 20×20 m, which were classified into two main edaphic habitats (flooded and drained). Trees with a diameter ≥1cm at breast height were identified. We assembled DNA sequence data for each of the 116 morphospecies recognized using two chloroplast markers (rbcL and matK). A phylogenetic tree was obtained using the maximum likelihood method, and a phylogenetic distance matrix was produced from an ultrametric tree. We analyzed similarity in floristic composition and structure between habitats and related them to cross-plot distances using permutation procedures. Null model torus shift simulations were performed to obtain a statistical significance level for habitat association for each species. The phylogenetic structure for the two habitats and for each 20×20 m quadrat was calculated using the mean phylogenetic distance weighted by species abundance and checked for significance using the standardized effect size generated by 5000 randomizations of phylogenetic tip labels.Important findings Our results indicate that partitioning among edaphic habitats is important for explaining species distributions and coexistence in restinga forests. Species distributions within the plot were found to be non-random: there was greater floristic similarity within than between habitats, and>40% of the more abundant species were positively or negatively associated with at least one habitat. Patterns of habitat association were not independent of phylogenetic relatedness: the community was overdispersed with respect to space and habitat type. Closely related species tended to occur in different habitats, while neighboring trees tended to belong to more distantly related species. We conclude that habitat specialization is important for the coexistence of species in restinga forests and that habitat heterogeneity is therefore an essential factor in explaining the maintenance of diversity of this unique but fragile and threatened type of forest. 相似文献

Dissolved organic carbon (DOC) photochemical reactions establish important links between DOC and planktonic bacteria. We hypothesize that seasonal changes in DOC quality, related to the flood pulse, drive the effects of light-DOC interactions on uptake by planktonic bacteria uptake in clear-water Amazonian ecosystems. Water samples from two ecosystems (one lake and one stream) were incubated in sunlight during different hydrological periods and were then exposed to bacterial degradation. Photochemical and bacterial degradation were driven by seasonal DOC inputs. Bacterial mineralization was the main degradation pathway of autochthonous DOC in the lake, while allochthonous DOC was more available for photochemical oxidation. We suggest that sunlight enhances the bacterial uptake of refractory DOC but does not alter uptake of labile forms. We also observed a positive relationship between sunlight and bacterial degradation of DOC, instead of competition. We conclude that photochemical reactions and bacteria complementarily degrade the different sources of DOC during the flood pulse in Amazonian clear-water aquatic ecosystems. 相似文献

Monoamine oxidase is a flavoenzyme bound to the mitochondrial outer membranes of the cells, which is responsible for the oxidative deamination of neurotransmitter and dietary amines. It has two distinct isozymic forms, designated MAO-A and MAO-B, each displaying different substrate and inhibitor specificities. They are the well-known targets for antidepressant, Parkinson's disease, and neuroprotective drugs. Elucidation of the x-ray crystallographic structure of MAO-B has opened the way for the molecular modeling studies. In this work we have used molecular modeling, density functional theory with correlation, virtual screening, flexible docking, molecular dynamics, ADMET predictions, and molecular interaction field studies in order to design new molecules with potential higher selectivity and enzymatic inhibitory activity over MAO-B. 相似文献

Cell signaling mediated by P2X7 receptors (P2X7R) has been suggested to be involved in epileptogenesis, via modulation of intracellular calcium levels, excitotoxicity, activation of inflammatory cascades, and cell death, among other mechanisms. These processes have been described to be involved in pilocarpine-induced status epilepticus (SE) and contribute to hyperexcitability, resulting in spontaneous and recurrent seizures. Here, we aimed to investigate the role of P2X7R in epileptogenesis in vivo using RNA interference (RNAi) to inhibit the expression of this receptor. Small interfering RNA (siRNA) targeting P2X7R mRNA was injected into the lateral ventricles (icv) 6 h after SE. Four groups were studied: Saline-Vehicle, Saline-siRNA, Pilo-Vehicle, and Pilo-siRNA. P2X7R was quantified by western blotting and neuronal death assessed by Fluoro-Jade B histochemistry. The hippocampal volume (edema) was determined 48 h following RNAi. Behavioral parameters as latency to the appearance of spontaneous seizures and the number of seizures were determined until 60 days after the SE onset. The Saline-siRNA and Pilo-siRNA groups showed a 43 and 37% reduction, respectively, in P2X7R protein levels compared to respective vehicle groups. Neuroprotection was observed in CA1 and CA3 of the Pilo-siRNA group compared to Pilo-Vehicle. P2X7R silencing in pilocarpine group reversed the increase in the edema detected in the hilus, suprapyramidal dentate gyrus, CA1, and CA3; reduced mortality rate following SE; increased the time to onset of spontaneous seizure; and reduced the number of seizures, when compared to the Pilo-Vehicle group. Therefore, our data highlights the potential of P2X7R as a therapeutic target for the adjunct treatment of epilepsy. 相似文献

Our understanding of nicotinamide adenine dinucleotide mitochondrial transporter 1 (Ndt1A) in Aspergillus fumigatus remains poor. Thus, we investigated whether Ndt1A could alter fungi survival. To this end, we engineered the expression of an Ndt1A-encoding region in a Δndt1Δndt2 yeast strain. The resulting cloned Ndt1A protein promoted the mitochondrial uptake of nicotinamide adenine dinucleotide (NAD+), generating a large mitochondrial membrane potential. The NAD+ carrier utilized the electrochemical proton gradient to drive NAD+ entrance into mitochondria when the mitochondrial membrane potential was sustained by succinate. Its uptake has no impact on oxidative stress, and Ndt1A expression improved growth and survival of the Δndt1Δndt2 Saccharomyces cerevisiae strain. 相似文献

The aim of this study was to evaluate the influence of Campusurus notatus Eaton 1868 (Ephemeroptera: Polimitarciydae) and the impact of bauxite tailings on ammonium (NH4+) and dissolved organic carbon (DOC) fluxes, oxygen uptake and bacterioplankton production in the sediment‐water interface of Lake Batata, a shallow Amazonian floodplain lake. Mesocosms were constructed from natural and impacted areas of Lake Batata, to reproduce the sediment‐water interface. The cores were incubated with 0 to 2,388 ind m–2 of Campsurus notatus nymphs, and the changes in NH4+, DOC, O2 concentration and bacterioplankton production in the overlying water column were measured. Ammonium efflux (F = 9.8, p < 0.05, multiple regression) and oxygen uptake (F = 11.8, p < 0.05) showed a significant correlation with the density of C. notatus in the cores with natural sediment. No differences on DOC release were observed in cores with natural or impacted sediment. In the cores incubated with natural sediment and nymphs of C. notatus, a significant increase (Two‐way ANOVA, p < 0.05) in bacterial production (0.44 μg C l–1 h–1) was observed after 3 hours of incubation. In cores incubated with sediment impacted by bauxite tailings, there was no difference in bacterial production with and without C. notatus. We conclude that C. notatus is an important bioturbator in Lake Batata, increasing the turnover rate of nitrogen (NH4+) at the sediment‐water interface and bacterial production in cores incubated with natural sediment. It is also clear that bauxite tailings reduce the nutrients turnover rates in impacted regions of Lake Batata and influence bacterial production. 相似文献

The parasitic plant Striga (Striga hermonthica) invades the host root through the formation of a haustorium and has detrimental impacts on cereal crops. The haustorium results from the prehaustorium, which is derived directly from the differentiation of the Striga radicle. The molecular mechanisms leading to radicle differentiation shortly after germination remain unclear. In this study, we determined the developmental programs that regulate terminal prehaustorium formation in S. hermonthica at cellular resolution. We showed that shortly after germination, cells in the root meristem undergo multiplanar divisions. During growth, the meristematic activity declines and associates with reduced expression of the stem cell regulator PLETHORA1 and the cell cycle genes CYCLINB1 and HISTONE H4. We also observed a basal localization of the PIN-FORMED (PIN) proteins and a decrease in auxin levels in the meristem. Using the structural layout of the root meristem and the polarity of outer-membrane PIN proteins, we constructed a mathematical model of auxin transport that explains the auxin distribution patterns observed during S. hermonthica root growth. Our results reveal a fundamental molecular and cellular framework governing the switch of S. hermonthica roots to form the invasive prehaustoria.The parasitic plant Striga hermonthica forms its invasive organ, the prehaustorium, by inducing differentiation of the radicle by arresting cell division. 相似文献

The egg parasitoid Anaphes nitens (Hymenoptera: Mymaridae) parasitizes the Eucalyptus snout-beetle Gonipterus platensis (Coleoptera: Curculionidae), one of the main defoliating beetles of Eucalyptus. Outbreaks of this pest are being recorded in areas with low parasitism rates by A. nitens. Endosymbiont bacteria can affect the reproductive characteristics of host insects increasing or decreasing the parasitism rate. The objectives of this study were to identify the presence, phylogeny and transmission modes of endosymbiont bacteria in A. nitens and G. platensis. Six populations of A. nitens and one of G. platensis were evaluated. Genomic DNA from these populations was extracted and nine genera of cell endosymbionts were searched by PCR. Three species of the Enterobacteriaceae family, Erwinia amylovora, Serratia grimesii, Yersinia massiliensis and the cell endosymbiont Rickettsia belli were identified in all A. nitens populations. Only Serratia grimesii was found in G. platensis. The presence of E. amylovora, Y. massiliensis and R. belli in the F1 and F2 generations indicates vertical transmission in A. nitens, while S. grimesii is vertically transmitted in G. platensis. 相似文献

In April 2022, Aristolochia plants with symptoms of mosaic were observed in a garden at Jardim Botânico Plantarum, Nova Odessa, São Paulo State, Brazil. Potyviridae-like particles were observed by transmission electron microscopy in leaf extracts. Total RNA extracted from symptomatic plants used in RT-PCR with universal and BCMV-specific primers detected the potyvirus bean common mosaic virus (BCMV). The cucumovirus cucumber mosaic virus (CMV) was identified only in Aristolochia littoralis plants that tested negative by RT-PCR for BCMV. Phylogenetic analysis grouped samples of Aristolochia in a different clade among samples of Phaseolus vulgaris. Phylogenetic analysis indicated that the CMV isolate from Aristolochia belongs to the CMV group IA. BCMV was mechanically transmitted to healthy plants of A. fimbriata, Chenopodium quinoa, P. vulgaris cv. Jalo and Macroptilium lathyroides. CMV was mechanically transmitted to plants of A. fimbriata and C. quinoa. The BCMV and CMV were aphid transmitted only by Aphis gossypii to Aristolochia plants. This is the first report of BCMV and CMV infecting Aristolochia plants in Brazil. 相似文献
