Nurseryfish are unique among fishes in that the males carry the fertilized eggs on a supraoccipital hook on their head. In an attempt to learn where, when, and at what salinities spawning occurs, an ichthyoplankton net was towed at 14 stations in the Adelaide River from the mouth (38 ppt) to the most upstream sections of the river (0.1 ppt). Larval nurseryfish (5–27.5 mm SL) were collected, preserved and measured, and water chemistry parameters were recorded with each sample. Larvae were found in the mid-reaches of the river during July–October, most commonly at salinities between 13.6–0.5 ppt. Salinities increased as the dry season progressed. Larvae were not taken at salinities higher than 19 ppt, nor did they occur in the upper-most reaches of the river system during this time of the dry season. What nurseryfish do and where they occur during the massive influx of freshwater runoff during the wet season (November–April) remains a mystery. Electrofishing for nurseryfish proved to be ineffective in the Adelaide River due to its extraordinary turbidity which made recovery of stunned fish difficult and fresh material of males carrying eggs remains elusive. Continuous-wave and pulsed electron paramagnetic resonance have been applied to the study of the Cu(II) site of the copper-resistance protein PcoC from Escherichia coli and certain variant forms. Electron spin echo envelope modulation (ESEEM) experiments confirm the presence of two histidine ligands, His1 and His92, at the Cu(II) site of wild-type PcoC, consistent with the available X-ray crystallographic data for the homolog CopC (67% sequence identity) from Pseudomonas syringae pv. tomato. The variants H1F and H92F each lack one of the histidine residues close to the Cu(II) site. The ESEEM data suggest that the surviving histidine residue remains as a ligand. The nA variant features an extra alanine residue at the N terminus, which demotes the His1 ligand to position 2. At least one of the two histidine residues is bound at the Cu(II) site in this form. Simulation of the (14)N superhyperfine structure in the continuous-wave spectra confirms the presence of at least three nitrogen-based ligands at the Cu(II) sites of the wild-type, H92F and nA forms, while the H1F variant has two nitrogen ligands. The spectra of wild-type form can be fitted adequately with a 3N or a 4N model. The former is consistent with the crystal structure of the CopC homolog, where His1 acts as a bidentate ligand. The latter raises the possibility of an additional unidentified nitrogen ligand. The markedly different spectra of the H1F and nA forms compared with the wild-type and H92F proteins further highlight the integral role of the N-terminal histidine residue in the high-affinity Cu(II) site of PcoC.  相似文献

Use of habitat is a critical component related to structure of small-mammal communities, with partitioning occurring primarily along dimensions of microhabitat, although use of microhabitat often does not explain fully use at a macrohabitat level. Through grid studies of small mammals in coastal Colima, Mexico (during January 2003–2005), we appraised influence of available habitat, species richness, abundance, and cumulative abundance of other small mammals on variation in habitat used by species. We evaluated 14 habitat variables (reflecting ground cover, slope, canopy, and vegetation density on vertical and horizontal axes) and developed a composite variable (principal component 1) reflecting general openness of habitat through which we addressed habitat use. For the four most common mammalian species (Sigmodon mascotensis, Heteromys pictus, Baiomys musculus, and Oryzomys couesi), two measures of variation in habitat used were employed to estimate niche breadth, one of which assessed variation in habitat use relative to variation present on a grid. Sigmodon mascotensis and B. musculus preferred areas that were more open, and H. pictus and O. couesi occupied less-open areas; breadth of habitat use did not differ interspecifically. Habitat use was more variable on grids with more variability in habitat, although not greater than chance expectations. Findings do not lend support to the resource-breadth hypothesis as an explanation for population densities of species at a local level or the habitat-heterogeneity hypothesis as a predictor of species richness. Variation in habitat used by S. mascotensis did not proportionally increase when diverse habitat was available but was greater when the species was more abundant. For H. pictus, when cumulative abundance of other small mammals was greater, breadth of habitat used was greater. Intraspecific density-dependent habitat selection may result in S. mascotensis selecting a greater variety of habitats, while greater interspecific abundance is related to a greater range in use of habitats by H. pictus. Baiomys musculus used a higher proportion of habitat relative to that available when more species were present on a grid. Variation in habitat used by O. couesi was unrelated to any factor examined. Overall, the four species responded in notably different ways with respect to availability of habitat, abundance, and presence of other species.  相似文献

Some odontocetes possess unique features of the hyolingual apparatus that are involved in suction feeding. The hyoid bone and associated musculature generates rapid, piston‐like retraction, and depression of the hyoid and tongue. “Capture” suction feeders (e.g., Globicephala) use suction for capturing and swallowing prey. “Combination” feeders (i.e., Lagenorhynchus) use both raptorial feeding (to capture prey) and suction (to ingest prey). In “capture” suction feeders, features of the hyoid and skull have been attributed to creating suction (i.e., large surface area and mandibular bluntness). In addition to odontocetes, a mysticete, the gray whale (Eschrichtius robustus), is considered a benthic suction feeder. However, anatomical studies of purported suction‐feeding structures of the gray whale are lacking. In addition, few studies have utilized evolutionary approaches to understand the history of suction feeding in cetaceans. This study incorporates quantitative and qualitative hyoid and cranial data from 35 extant and 14 extinct cetacean species into a multivariate principal component analysis and comparative phylogenetic analyses. Conclusions from these analyses are that some commonly attributed features (i.e., ventral throat grooves and mandibular bluntness) and one principal component are significantly correlated with suction feeding. Finally, ancestral state reconstructions indicate that suction feeding likely evolved once, early in cetacean evolutionary history.  相似文献

Redox thermodynamic data provide a detailed insight into control of the reduction potential E degrees' of the [Fe(S-Cys)4] site in rubredoxin. Mutant forms were studied in which specific structural changes were made in both the primary and secondary coordination spheres. Those changes have been probed by resonance Raman spectroscopy. The decrease of approximately 200 mV in E degrees' observed for the [Fe(S-Cys)3(O-Ser)]-/2- couples in the surface ligand mutants C9S and C42S is essentially enthalpic in origin and associated with the substitution of ligand thiolate by ligand olate. However, the pH dependence of the potentials below characteristic pKa(red) approximately equals 7 is an entropic contribution, plausibly associated with increased conformational flexibility induced by a longer Fe(II)-O(H)-Ser bond in the reduced form. The presence of a second surface Ser ligand in the new double mutant protein C9S/C42S affects the enthalpic term primarily for pH>pKa(red) > or = 9.3, but for pHpKa approximately 9: [Fe(III)(S-Cys)3(OH)]- + e- --> [Fe(II)(S-Cys)3(OH)]2-. pH [Fe(II)(S-Cys)3(OH2)]-.  相似文献

The intensity of exercise above which oxygen uptake (Vo2) does not account for all of the required energy to perform work has been associated with lactate accumulation in the blood (lactate threshold, LT) and elevated carbon dioxide output (gas exchange threshold). An increase in hydrogen ion concentration ([H+]) is approximately concurrent with elevation of blood lactate and CO2 output during exercise. Near-infrared spectra (NIRS) and invasive interstitial fluid pH (pHm) were measured in the flexor digitorum profundus during handgrip exercise to produce a mathematical model relating the two measures with an estimated error of 0.035 pH units. This NIRS pHm model was subsequently applied to spectra collected from the vastus lateralis of 10 subjects performing an incremental-intensity cycle protocol. Muscle oxygen saturation (SmO2) was also calculated from spectra. We hypothesized that a H+ threshold could be identified for these subjects and that it would be different from but correlated with the LT. Lactate, gas exchange, SmO2, and H+ thresholds were determined as a function of Vo2 using bilinear regression. LT was significantly different from both the gas exchange threshold (Delta = 0.27 +/- 0.29 l/min) and H+ threshold (Delta = 0.29 +/- 0.23 l/min), but the gas exchange threshold was not significantly different from the H+ threshold (Delta = 0.00 +/- 0.38 l/min). The H+ threshold was strongly correlated with LT (R2 = 0.95) and the gas exchange threshold (R2 = 0.85). This initial study demonstrates the feasibility of noninvasive pHm estimations, the determination of H+ threshold, and the relationship between H+ and classical metabolic thresholds during incremental exercise.  相似文献

The central California coast is visited by millions of people every year, many of whom collect intertidal gastropods for food, bait or recreation in near-shore marine environments. The harvesting of these animals has caused a decline in body size because humans preferentially take the largest individuals. Marine protected areas (MPAs) may serve to protect gastropods from this impact. The average body size and frequency of four gastropod species from eight study locations along the central California coast were determined. Five study locations were in MPAs and three study locations were in non-MPA zones. Gastropods in four California museums were measured and compared to specimens collected in the field. Three of the four species studied were larger and/or more frequent in MPAs than outside MPAs, including both harvested species and one unharvested species. For three of the four species, the average size of individuals from MPA sites was significantly smaller than museum specimens, but this may be due, in part, to our sampling procedure. These findings indicate that MPAs can be effective at reducing human impacts for some species, but that further study is needed to make MPAs as effective as possible for more species.  相似文献
