Opportunities exist in high Arctic polar semidesert communities for colonisation of unvegetated ground by long-lived clonal plants such as Dryas octopetala. This can be achieved by lateral spread of vegetative ramets, or by sexual reproduction and seedling recruitment. The objectives of this study were (1) to determine whether these two means of proliferation show differential sensitivity to contrasting components of the abiotic environment (temperature, soil nutrient and water availability) and (2) to evaluate the potential for D. octopetala to respond to climate change by an increase in cover and biomass in polar semi-desert communities. Factorial environmental manipulations of growing season temperature, soil nutrient and water status were conducted over 3 years at a polar semi-desert community in Svalbard, Norway (78°56.12N, 11°50.4E) and both clonal and sexual reproductive performance, together with instantaneous net photosynthesis (Pn), were recorded during the third season (1993). D. octopetala capitalised rapidly on an amelioration in the availability of inorganic nutrients (N, P and K) by an expansion in leaf area and biomass supported by increased Pn per unit leaf weight, and by apparent luxury uptake of nutrients (particularly P). Several facets of sexual reproductive development and seed viability were markedly improved by elevated temperatures or soil nutrient availability. Thus although D. octopetala is a long-lived clonal plant, with many traits characteristic of stress resistance syndrome, it showed considerable phenotypic plasticity in response to environmental manipulations. The results support the hypothesis that clonal growth confers survival potential during unfavourable years, together with the ability to capitalise on nutrient flushes and recycle nutrients internally. Continued investment in sexual reproduction ensures that seed setting is successful during favourable years, even if these occur infrequently. Cimate warming in the high Arctic could thus result in changes in the cover, biomass and the relative significance of clonal versus sexual proliferation of D. octopetala (and thus the genetic diversity of the population) but the long-term responses will probably be constrained by lack of available nutrients. 相似文献

The aim of this study was to test the potential of proteinase inhibitors to controlChrysomela tremulae, a beetle that causes severe damage in young plantations and in short-rotation intensive culture (SRIC) of poplar. As a first step, cysteine proteinases were determined to be the major digestive proteinases ofC. tremulae and oryzacystatin OCI, a cysteine proteinase inhibitor, was shown to inhibit this activityin vitro. The gene encoding OCI was introduced into poplar (Populus tremula ×P. tremuloides) and transgenic plants expressing OCI at a high level were selected. Retrovirus morphogenesis involves assembly of structural Gag polyproteins with subsequent budding from the plasma membrane, followed by proteolytic cleavage by the viral proteinase (PR) and extracellular maturation to the infectious virion. Intracisternal A-type particles (IAPs) are defective retroviruses that assemble and bud at the membranes of the endoplasmic reticulum (ER), where they remain as immature particles consisting exclusively of uncleaved polyproteins. To analyze requirements for intracellular polyprotein transport and PR activation, we constructed deletion and substitution mutations in the IAP gag gene, including the putative ER-targeting signal. Mutant polyproteins were transported to various intracellular locations, including the nucleus, the cytoplasm, the ER, and the plasma membrane. Interestingly, assembly of capsid-like particle structures occurred at almost all sites. However, only those polyproteins transported to the plasma membrane were efficiently and specifically cleaved by viral PR, with cleavage occurring predominantly within the virus particle. Thus, at least in the experimental system presented here, retroviral particle assembly can occur at almost any location within the cell, while polyprotein processing and, consequently, virion maturation are confined to a specific cellular site. These results suggest that a factor restricted to the plasma membrane is required to trigger PR activation and maturation of infectious retroviruses. 相似文献

A new species of Neomarica from the Brazilian Atlantic Forest is described and illustrated. Neomarica sergipensis A. Gil & M.C.E. Amaral sp. nov. (Trimezieae, Iridoideae, Iridaceae) grows in restinga forests (forests on the coastal plains dominated by sandy soils) in the state of Sergipe, northeastern Brazil. The new species resembles Neomarica northiana, especially due to the white flowers with the basal third of the tepals yellow and covered by brown maculae, but differs by the inner tepals of with the center of the apical third is tinted violet to purple, yellow stigmatic crests with fimbriate to erose margins, and the endemic occurrence in Sergipe. 相似文献

Mechanisms involved in severe P. vivax malaria remain unclear. Parasite polymorphisms, parasite load and host cytokine profile may influence the course of infection. In this study, we investigated the influence of circumsporozoite protein (CSP) polymorphisms on parasite load and cytokine profile in patients with vivax malaria. A cross-sectional study was carried out in three cities: São Luís, Cedral and Buriticupu, Maranhão state, Brazil, areas of high prevalence of P. vivax. Interleukin (IL)-2, IL-4, IL-10, IL-6, IL-17, tumor necrosis factor alpha (TNF-α, interferon gamma (IFN-γ and transforming growth factor beta (TGF-β were quantified in blood plasma of patients and in supernatants from peripheral blood mononuclear cell (PBMC) cultures. Furthermore, the levels of cytokines and parasite load were correlated with VK210, VK247 and P. vivax-like CSP variants. Patients infected with P. vivax showed increased IL-10 and IL-6 levels, which correlated with the parasite load, however, in multiple comparisons, only IL-10 kept this association. A regulatory cytokine profile prevailed in plasma, while an inflammatory profile prevailed in PBMC culture supernatants and these patterns were related to CSP polymorphisms. VK247 infected patients showed higher parasitaemia and IL-6 concentrations, which were not associated to IL-10 anti-inflammatory effect. By contrast, in VK210 patients, these two cytokines showed a strong positive correlation and the parasite load was lower. Patients with the VK210 variant showed a regulatory cytokine profile in plasma, while those infected with the VK247 variant have a predominantly inflammatory cytokine profile and higher parasite loads, which altogether may result in more complications in infection. In conclusion, we propose that CSP polymorphisms is associated to the increase of non-regulated inflammatory immune responses, which in turn may be associated with the outcome of infection. 相似文献

Phage TP-8 lysates of Bacillus stearothermophilus 4S or 4S(8) contain lytic activity exhibiting two pH optima, one at pH 6.5 and the other at pH 7.5. Using a variety of fractionation procedures, the two lytic activities could not be separated. At pH 7.5 the lytic enzyme is an endopeptidase which hydrolyzes the l-alanyl-d-glutamyl linkage in the peptide subunits of the cell wall peptidoglycan and at pH 6.5 it exhibits N-acetylmuramidase activity. Endopeptidase activity is inhibited by NaCl and neither lytic activity was significantly affected by divalent cations or ethylenediaminetetraacetic acid. Crude lysates contain 2.5 to 3.0 times more endopeptidase activity than N-acetylmuramidase activity. The ratio of the two lytic activities (endopeptidase/N-acetylmuramidase) changes to 1.3 to 1.7 during the course of purification, to 1.0 after isoelectric focusing, and 3.9 and 6.00 after exposure for 2 h at 60 and 65 C, respectively. We conclude that the two lytic activities may be associated with a single protein or a lytic enzyme complex composed of two enzymes. Lytic activity at pH 7.5 is more effective in solubilizing cells or cell walls than the lytic activity at pH 6.5. LiCl extracts of 4S and 4S(8) cells contain lytic activity exhibiting endopeptidase activity at pH 7.5 and N-acetylmuramidase activity at pH 6.5. Lytic activity in these LiCl extracts also has a number of other properties in common with those in lysates of phage TP-8. Summary Nitrogen partitioning among three generations of tillers within the bunchgrass Schizachyrium scoparium var. frequens was investigated in a controlled environment as a potential mechanism of herbivory tolerance. Nitrogen-15 was transported from the labelled primary tiller generation to both shoots and roots of nondefoliated secondary and tertiary tiller generations within 24 h. Partial defoliation increased shoot nitrogen concentration of secondary and tertiary generation tillers by 110 and 120%, respectively, 24 h following defoliation. Shoot nitrogen concentration was preferentially increased by partial defoliation of tertiary generation tillers throughout the 120 h experimental period, but diminished to concentrations comparable to nondefoliated tillers within shoots of the secondary generation at 72 h. In contrast to nitrogen concentration, the total amount of nitrogen imported by secondary and tertiary generation tillers decreased 62 and 73%, respectively, 24 h following partial defoliation and did not attain values comparable to respective nondefoliated tillers. Consequently, preferential nitrogen concentration occurred in response to partial tiller defoliation without an increase in total nitrogen import based on the reduction in the total nitrogen requirement per tiller generation associated with defoliation. Estimates of both the total amount of nitrogen import and nitrogen concentration are necessary to accurately interpret the dynamics of intertiller nitrogen allocation. 相似文献

Assimilatory nitrate reductase has been purified with 55% recovery from a Neurospora crassa nmr-1 nit-6 mutant, using a modification of a published procedure. It possesses one heme per 240 000 g, and subunits of mol. wt. 68 000. Upon digestion with chymotrypsin, a heme-binding domain was isolated by gel filtration; its visible spectrum was highly similar to that of cytochrome b5. On SDS gels, the fraction showed two heme-containing bands of ˜10 000 and 12 5000 daltons; their amino acid composition was not very different, suggesting that they originated from the same region of the polypeptide chain. After S-carboxymethylation, the mixture of bands was submitted to cyanogen bromide cleavage, and the fragments were separated by h.p.l.c. The two largest fragments yielded an identical sequence upon automated degradation. This sequence (39 residues with some gaps) could be easily aligned with that of cytochrome b5 starting close to the N terminus. These results are discussed in terms of the possible quaternary structure of N. crassa nitrate reductase, whose heme-binding domain proves to be another member of the family of b5-like cytochromes. 相似文献

Temperate and virulent bacteriophages isolated from soil were shown to carry out generalized transduction of Bacillus stearothermophilus NUB36. A transducing frequency of 1 X 10(-5) to 7 X 10(-4) was obtained for temperate phages TP-42 and TP-56. The transducing frequency for virulent phage TP-68 was two to three orders of magnitude lower. Cotransfer analysis with the three phages showed that hom-1 is linked to thr-1 and that gly-1 is linked to his-1. 相似文献
