A novel generic approach for stress profiling was applied to Listeria monocytogenes strain F2365. This food-borne pathogen was exposed to gradients of five different stresses of increasing intensity, typically ranging from moderate to lethal conditions. The stress factors included heat, acidic pH, a detergent disinfectant, an oxidant, and hyperosmotic conditions. In addition to CFU counts and lag time, five different molecular viability parameters were measured by fluorescence-based assays, including membrane integrity, membrane potential, esterase activity, redox activity, and intracellular pH stability. The last was measured by our recently invented real-time viability assay. Exposure to all stresses resulted in clear dose-response relationships for all viability parameters with the exception of hyperosmotic conditions. A statistical analysis showed strong correlations for (i) the growth parameters plate counts and lag times, (ii) the enzyme-associated functions redox and esterase activity, and (iii) the membrane-associated pH stability and membrane integrity. Results indicated a pronounced difference in the susceptibilities of the measured parameters depending on the stress factor applied. However, at relatively high stress intensities, all of the viability parameters became affected independent of the stress factor. Applications of the approach presented here include studies on the mechanism of action of unknown compounds with biocidal activity and a comparative analysis of the severities of the impact of stress conditions of interest. It appears that a meaningful evaluation of the impact of mild stress conditions can be obtained only through measurement of multiple viability parameters.  相似文献

为了分析比格犬黑素皮质素受体-4基因多态性与犬体重的关系, 根据犬MC4R基因DNA外显子序列, 设计MC4R基因特异PCR引物1对, 犬DNA经PCR扩增, 克隆和测序, 寻找和确定犬MC4R基因的多态性位点, 分析多态性与犬体重的关系。结果在比格犬MC4R基因中发现2处单碱基缺失突变, 1个单碱基颠换变异, 存在Psh AⅠ酶切位点, 并基于PshAⅠ酶切位点建立了PCR-RFLP技术。统计分析显示犬MC4R基因型与体重显著相关, 可以考虑将MC4R基因作为犬体重的候选基因。  相似文献

目的:探讨MR弥散加权成像(DWI)鉴别诊断良恶性椎体压缩性骨折的临床价值。方法:对57 例经临床或病理证实的椎体 良恶性压缩性骨折患者行矢状位T1WI、T2WI、T2WI/FS 及DWI扫描,研究其在常规序列和DWI序列上的表现,将常规MR 序列 和DWI序列检出率进行比较,测量正常椎体及病变椎体的表观弥散系数(ADC)值,并进行统计学分析。结果:(1)MR 常规序列和 DWI序列(b=500s/mm2)表现:良性椎体压缩性骨折呈长T1 长或等T2 改变,T2WI/FS 呈高信号,DWI 可以呈高信号、等信号及低 信号;恶性椎体压缩性骨折呈长T1 长T2 信号,大部分病灶T2WI/FS 及DWI呈高信号,少数变现为低信号;(2)MR 常规序列和 DWI 序列(b=500s/mm2)病灶检出率的比较:T1WI、T2WI/FS 及DWI序列病灶检出率均高于T2WI 序列,其间的差别有显著性意 义(P<0.01),T1WI、T2WI/FS 及DWI序列病灶检出率之间无显著性差异(P>0.01);(3)ADC 值比较:在DWI(b=500 s/mm2)上,良性组 ADC 值为(2.03± 0.83)× 10-3mm2/s,恶性组ADC 值为(1.37 ± 0.75)× 10-3mm2/s,正常组ADC值为(0.36± 0.21)× 10-3mm2/s,成像条 件相同时,良性组高于恶性组,两组间有明显的统计学意义(P<0.05)。结论:DWI可较好的反映椎体的弥散特征,ADC值作为量化 指标可对良恶性椎体压缩性骨折进行可靠鉴别。  相似文献

Scent marks are relatively long-lived signals that can be perceived by conspecifics when the producer is absent. Therefore, it is often not obvious to whom the signal is directed. In daytime roosts of the polygynous greater sac-winged bat, males scent mark territories with facial gland secretions. Territories are a valuable resource for males, as they offer exclusive courtship opportunities, which results in increased male reproductive success and, consequently, increased male–male competition over territories. The information encoded in male scent marks could, therefore, be either directed at females as part of an olfactory courtship display or at male competitors as part of territorial behaviour. We expected territorial males to scent mark in the morning, shortly before females return to the territory and close to female roosting sites, if scent marks are directed at females as part of the courtship display. And we expected harem males to scent mark at the territory boundaries, where male–male encounters are most likely to occur, if scent marks are directed at male competitors. We found that males marked more frequently in the afternoon, at a time when all females have already left the territory, and harem males marked at the territory boundaries and not inside their territory in the area where females roost. At boundaries males fan volatiles from specialised wing sacs towards competitors outside the territory. Scent marking of male Saccopteryx bilineata might therefore be congruent with the assessment-hypothesis, which states that scent marks offer intruders the possibility to make an olfactory assessment of the territory owner without direct physical interaction. Thus, scent marks of male S. bilineata are most likely influenced by male–male competition and not by female choice.  相似文献

Starch granule size is an important parameter for starch applications in industry. Starch granules are formed in amyloplasts, which are, like chloroplasts, derived from proplastids. Division processes and associated machinery are likely to be similar for all plastids. Essential roles for FtsZ proteins in plastid division in land plants have been revealed. FtsZ forms the so-called Z ring which, together with inner and outer plastid division rings, brings about constriction of the plastid. It has been shown that modulation of the expression level of FtsZ may result in altered chloroplast size and number. To test whether FtsZ is also involved in amyloplast division and whether this, in turn, may affect the starch granule size in crop plants, FtsZ protein levels were either reduced or increased in potato. As shown previously in other plant species, decreased StFtsZ1 protein levels in leaves resulted in a decrease in the number of chloroplasts in guard cells. More interestingly, plants with increased StFtsZ1 protein levels in tubers resulted in less, but larger, starch granules. This suggests that the stoichiometry between StFtsZ1 and other components of the plastid division machinery is important for its function. Starch from these tubers also had altered pasting properties and phosphate content. The importance of our results for the starch industry is discussed.  相似文献

Several inhibitors of a series of cis-1(S)2(R)-amino-2-indanol-based compounds were reported to be selective for the aggrecanases, ADAMTS-4 and -5 over other metalloproteases. To understand the nature of this selectivity for aggrecanases, the inhibitors, along with the broad spectrum metalloprotease inhibitor marimastat, were independently bound to the catalytic domain of ADAMTS-5, and the corresponding crystal structures were determined. By comparing the structures, it was determined that the specificity of the relative inhibitors for ADAMTS-5 was not driven by a specific interaction, such as zinc chelation, hydrogen bonding, or charge interactions, but rather by subtle and indirect factors, such as water bridging, ring rigidity, pocket size, and shape, as well as protein conformation flexibility.Osteoarthritis (OA)3 pathology includes degradation of articular cartilage, along with subchondral bone sclerosis and osteophyte formation, all contributing to impaired joint function. Pain, restricted movement, and joint instability accompany these structural changes and often result in the need for total joint replacement. Current therapies alleviate the mild to moderate pain and inflammation associated with OA, but do not protect the cartilage from further damage and have not demonstrated an effect on disease progression (1). Therefore, therapeutics that prevent or slow the alteration of joint structure and function will address a major unmet medical need.Loss of aggrecan, a macromolecular proteoglycan providing cartilage with its properties of compressibility and resilience, is a major phenotype associated with OA and is believed to be a critical event in driving disease progression (2, 3). Both ex vivo and in vivo proof of concept studies support ADAMTS-4 and ADAMTS-5, commonly referred to as aggrecanase-1 and -2, respectively, as the two major enzymes responsible for the proteolytic breakdown of cartilage aggrecan (reviewed in Ref. 4). Blocking their activity may be an attractive strategy to stop or slow down the progression of the disease, as suggested by studies in knock-out mice (5). Given the chronic nature of the disease, long term treatment will be likely, demanding very safe therapeutic interventions only achievable with ADAMTS-4- and ADAMTS-5-specific inhibitors lacking off-target side effects. Designing selectivity has been very challenging and a major source of difficulty is that at least 57 metalloproteases (MP) divided in three major families, 1) matrix metalloproteases (MMP); 2) a disintegrin and metalloproteinase (ADAM); and 3) a disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS), are present in humans. ADAMTS-4 and -5 belong to the ADAMTS family and share common catalytic and structural features with the other MP members. These features include the highly conserved amino acid sequence, HEXXHXXGXXH, harboring a catalytic zinc cation, required for activation of the peptide bond toward hydrolysis. In addition, many MPs share significant structural topology in the active site, such as a flexible S1′ loop. To complicate matters further, only a handful of MP structures, mostly in the MMP family, have been determined, and the functions of most MPs still remain unknown, a fact that has earned them an orphan status denomination. Lack of structural information has hindered the design for inhibitor specificity and without known substrates for many MPs, assays for screening inhibitors are often not available, making determinations of selectivity difficult (6, 7).Yao et al. (8, 9) reported the discovery of a series of (2R)-N4-hydroxy-2-(3-hydroxybenzyl)-N1-[(1S,2R)-2-hydroxy-,3-dihydro-1H-inden-1-yl]butanediamide derivatives as potent and selective inhibitors of aggrecanase activity. Using a homology model of aggrecanase based on the active site of atrolysin C and adamalysin II and docking compound 8, shown in Fig. 1, the authors concluded that the 3-hydroxyl group of inhibitor 8 achieved selectivity through a specific hydrogen-bonding interaction with Thr440 (numerical numbering is based on the human sequence of ADAMTS-5) in the S1′ pocket of aggrecanase.Open in a separate windowFIGURE 1.Aggrecanase inhibitors. Recently, our group has established a protocol for crystallizing the catalytic domain of ADAMTS-5 and determined its three-dimensional structure (10). Thus, experimental validation or invalidation of the hypothesis that inhibitor 8 and related molecules form a specific hydrogen bond with the hydroxyl group of threonine in the S1′ pocket of aggrecanases could now be determined by protease-inhibitor crystallographic analysis.In the current study, we wanted to confirm and extend the selectivity profile of compound 8 and 11 against a wide array of MPs. Moreover, we wanted to elucidate the key molecular interactions responsible for the enhanced selectivity profile of this series of compounds. For this purpose we generated co-crystals and solved the structures of marimastat, compound 8, and compound 11 in complex with the catalytic domain of recombinant human ADAMTS-5.  相似文献

Novel C(3) propenylamide and propenylsulfonamide cephalosporins have been synthesized and tested for their ability to inhibit the penicillin-binding protein 2′ (PBP2′) from Staphylococcus epidermidis and the growth of a panel of clinically relevant bacterial species, including methicillin-resistant Staphylococcus aureus (MRSA). The most potent compounds inhibited the growth of MRSA strains with minimum inhibitory concentrations (MIC) as low as 1 μg/mL. The structure–activity relationship revealed the potential for further optimization of this new cephalosporin class.  相似文献

目的:探究高血压脑出血患者术后重症监护治疗与早期(24 h内)再出血的相关因素。方法:回顾性分析2014年1月至2018年10月于中山大学附属第一医院及中山市人民医院行手术治疗并进行重症监护的高血压脑出血患者的相关资料,记录术后早期发生再出血情况,比较其相关因素,包括年龄、性别、术前格拉斯哥昏迷量表(GCS)评分、出血量、术前收缩压、术后收缩压、镇静时间、插管时间、有无使用止血药、血压波动、血压差、有无镇痛情况,分析术后早期再出血的影响因素。结果:本研究共纳入465例患者,其中术后早期再出血患者44例,未再出血患者421例,再出血发生率为9.46%(44/465)。高血压脑出血术后早期再出血患者的术后收缩压、有无镇痛、血压差、血压波动与未再出血患者比较差异具有统计学意义(P0.05)。术后早期再出血患者的年龄、性别及术前GCS评分、出血量、术前收缩压、镇静时间、插管时间、有无使用止血药与未再出血患者比较差异无统计学意义(P0.05)。多因素Logistic回归分析显示,患者血压波动大是术后早期再出血的危险因素,手术前后血压差大、术后使用镇痛治疗是其保护因素。结论:高血压脑出血患者术后血压波动、手术前后血压差及术后镇痛治疗均是早期再出血的影响因素,合理降压及镇痛治疗可减少脑出血术后早期再出血的发生。  相似文献

Male traits that correlate with fertilization success include testis size and structure, ejaculate size, ejaculation frequency, and sperm motility. Two hypotheses potentially explain interspecific differences in these traits: sperm competition and sperm limitation. We examined variation in six traits associated with fertilization success in three closely‐related species of bitterling fish; the European bitterling (Rhodeus amarus), the Chinese rose bitterling (Rhodeus ocellatus), and the Chinese bitterling (Rhodeus sinensis). Interspecific differences indicated that the three study species have evolved different sperm allocation strategies. Rhodeus amarus displayed the most developed reproductive apparatus with a number of traits associated with both high levels of sperm production and fertilization efficiency. Rhodeus ocellatus and R. sinensis appear to have more comparable sperm allocation strategies, although relative testis size and spermatozoa head : tail ratio were greater in R. sinensis, suggesting that sperm competition risk may be higher in this species. All three species possessed an unusually well developed sperm duct with evidence of mucin production, which greatly extends the longevity of sperm and, consequently, the period over which fertilization can occur. We discuss these findings in the context of differences in the mating systems of the species examined, and relate the results obtained to differences in the temporal and spatial clustering of fertilizations. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 622–632.  相似文献
