1. In situ exclosure experiments in the Mississippi and Ohio Rivers determined the importance of fish predation in regulating zebra mussels (Dreissena polymorpha), an increasingly important constituent of the benthic invertebrate assemblages in both rivers. 2. We evaluated the effects of predatory fish on the density, biomass and size distribution of zebra mussels in a floodplain reach of the upper Mississippi River and in a naturally constrained reach of the Ohio River. Fifty, six-sided, predator-exclusion cages and fifty ‘partial’ cages (mesh at the upstream end only) were deployed, with half the cages containing willow snags and half clay tiles suspended 12–16 cm above the bottom. A single snag or tile sample unit was removed from each cage at approximately monthly intervals from July to October 1994. Types and relative abundances of molluscivorous fish were evaluated by electrofishing near the cages in both rivers. Actual and potential recruitment of young zebra mussels on to the substrata were measured using benthic samples in both rivers and estimated (Ohio River only) from counts of planktonic veligers. 3. Zebra mussels were consumed by at least three fish species in the upper Mississippi River (mostly carp, Cyprinus carpio, and redhorse suckers, Moxostoma sp.) and five species in the Ohio River (primarily smallmouth buffalo, Ictiobus bubalus, and channel catfish, Ictalurus punctatus), but potential recruitment seemed adequate to replace consumed mussels, at least in the Ohio River. The number of juvenile benthic mussels showed no apparent link with the density of veligers soon after initiation of reproduction. Recruitment of juveniles on snags and tiles was not affected by cage type (thus eliminating a potentially confounding ‘cage effect’). 4. Fish significantly influenced mussel populations, but the impact was often greatest among low density populations in the upper Mississippi. Density and biomass differed in both rivers for cage type (higher inside cages), substratum (greater on tiles), and date (increased over time). Presumed size-selective predation was present in the Mississippi (greater on larger size classes) but was not evident in the Ohio. We hypothesize that fish in the Mississippi can more easily select larger prey from the low density populations; whereas size-selective predation on tightly packed zebra mussels in the Ohio would be difficult. 5. Although fish can reduce numbers of Dreissena polymorpha in the two rivers, current levels of fish predation seem insufficient to regulate zebra mussel densities because of its great reproductive capacity. The recent invasion of zebra mussels, however, could lead to larger fish populations while promoting greater carbon retention and overall ecosystem secondary production.  相似文献

Summary In a prior study we combined game theory and inclusive fitness models to examine whether the guarded altruism that can evolve among non-relatives (tit for tat, TFT) might also evolve among close relatives, supplanting unconditional altruism. In most cases, TFT replaced unconditional altruism in family-structured models. Even when TFT is selected at a single locus, however, by withholding altruism from non-reciprocating relatives it may qualify as an outlaw from the standpoint of modifier genes at other loci. Here we examine this possibility with a series of haploid, two-locus models in which a modifier gene transforms TFT into unconditional altruism. The modifier allele spreads to fixation whenever Hamilton's Rule is satisfied, resulting in an unconditional altruist replacing the TFT strategy. As such, TFT may be regarded as an outlaw vulnerable to suppression by alleles at other loci.  相似文献

The selective estrogen receptor modulator, clomiphene citrate (CC), has been the principal drug used for induction of ovulation in women with polycystic ovarian syndrome (PCOS). CC is associated with adverse side effects and low pregnancy rates attributed to long-lasting estrogen receptor depletion. Anastrozole and letrozole are potent, non-steroidal, reversible aromatase inhibitors, developed for postmenopausal breast cancer therapy. We hypothesized that aromatase inhibitors could mimic the action of CC in reducing estrogen negative feedback on follicle stimulating hormone (FSH) secretion, without depleting estrogen receptors. In a series of preliminary studies, we reported the success of aromatase inhibition in inducing ovulation in anovulatory women with PCOS. Moreover, we showed that concomitant use of aromatase inhibitors resulted in a significant reduction of the FSH dose needed for controlled ovarian hyperstimulation. We suggest that aromatase inhibitors act through an increase in endogenous gonadotropin secretion as well as through increased intraovarian androgen levels that may increase ovarian FSH receptors. Recently, we demonstrated the safety of aromatase inhibitors in pregnancy outcome studies examining spontaneous pregnancy loss, multiple pregnancy rates and congenital anomalies compared to a control group of infertility patients treated with CC.  相似文献

UDP-GlcNAc: Man1-6R (1-2)-N-acetylglucosaminyltransferase II (GlcNAc-T II; EC 2.4.1.143) is a key enzyme in the synthesis of complexN-glycans. We have tested a series of synthetic analogues of the substrate Man1-6(GlcNAc1-2Man1-3)Man-O-octyl as substrates and inhibitors for rat liver GlcNAc-T II. The enzyme attachesN-acetylglucosamine in 1-2 linkage to the 2-OH of the Man1-6 residue. The 2-deoxy analogue is a competitive inhibitor (K i=0.13mm). The 2-O-methyl compound does not bind to the enzyme presumably due to steric hindrance. The 3-, 4- and 6-OH groups are not essential for binding or catalysis since the 3-, 4- and 6-deoxy and -O-methyl derivatives are all good substrates. Increasing the size of the substituent at the 3-position to pentyl and substituted pentyl groups causes competitive inhibition (K i=1.0–2.5mm). We have taken advantage of this effect to synthesize two potentially irreversible GlcNAc-T II inhibitors containing a photolabile 3-O-(4,4-azo)pentyl group and a 3-O-(5-iodoacetamido)pentyl group respectively. The data indicate that none of the hydroxyls of the Man1-6 residue are essential for binding although the 2- and 3-OH face the catalytic site of the enzyme. The 4-OH group of the Man-O-octyl residue is not essential for binding or catalysis since the 4-deoxy derivative is a good substrate; the 4-O-methyl derivative does not bind. This contrasts with GlcNAc-T I which cannot bind to the 4-deoxy-Man- substrate analogue. The data are compatible with our previous observations that a bisectingN-acetylglucosamine at the 4-OH position prevents both GlcNAc-T I and GlcNAc-T II catalysis. However, in the case of GlcNAc-T II, the bisectingN-acetylglucosamine prevents binding due to steric hindrance rather than to removal of an essential OH group. The 3-OH of the Man1-3 is an essential group for GlcNAc-T II since the 3-deoxy derivative does not bind to the enzyme. The trisaccharide GlcNAc1-2Man1-3Man-O-octyl is a good inhibitor (K i=0.9mm). The above data together with previous studies indicate that binding of the GlcNAc1-2Man1-3Man- arm of the branched substrate to the enzyme is essential for catalysis. Global climate change is projected to increase the incidence of heat waves, their magnitude and duration resulting in insects experiencing increasing environmental stress in both natural and managed ecosystems. While studies on insect thermal tolerance are rapidly increasing, variation across developmental or juvenile stress cross-stage effects within and across generations remain largely unexplored. Yet in holometabolous insects, heat stress at an early developmental stage may influence performance and survival during later stages. Here, we investigated the effects of pupal mild heat stress on the performance of laboratory-reared adult Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) measured as longevity, critical thermal maximum (CTmax), critical thermal minima (CTmin), heat knockdown time (HKDT) and chill coma recovery time (CCRT). Pupal heat stress significantly influenced performance of B. dorsalis adults resulting in impaired longevity and heat tolerance (CTmax and HKDT) in both sexes with improved and compromised cold tolerance (CTmin and CCRT) in females and males, respectively. These findings highlight the role of juvenile stages in mediating stress responses at adult stages. For B. dorsalis, pupal heat stress largely compromised thermal tolerance implying that the species has limited potential to shift its geographic range in heat prone areas. Significant benefits in cold tolerance in females following heat stress may help in improving survival in the cold in the short-term despite restricted activity to the same traits in males. This study suggests that basal heat tolerance and not short-term compensatory thermal plasticity following heat stress may have aided the recent invasion of B. dorsalis in African landscapes.  相似文献

Many efforts to restore disturbed landscapes seek to meet ecological goals over timescales from decades to centuries. It is thus crucial to know how different actions available to restoration practitioners may affect ecosystems in the long term, yet few such data exist. Here, we test the effects of seed and compost applications on plant community composition 9 years after their application, by taking advantage of a well‐controlled restoration experiment on a mountainside severely degraded by over 80 years of zinc smelting emissions. We asked whether plots have converged on similar plant communities regardless of initial seed and compost treatments, or if these initial treatments have given rise to lasting differences in whole plant communities or in the richness and abundance of native, exotic, and planted species. We found that compost types significantly affected plant communities 9 years later, but seed mix species composition did not. Observed differences in species richness and vegetative cover were negatively correlated, and both were related to the differences in plant communities associated with different compost types. These observed differences are due primarily to the number and abundance of species not in original seed mixes, of which notably many are native. Our results underscore the importance of soils in shaping the aboveground composition of ecosystems. Differences in soil characteristics can affect plant diversity and cover, which are both common restoration targets. Even in highly polluted and devegetated sites, compost and seed application can reinstate high vegetative cover and allow continued colonization of native species.  相似文献

Common fragile sites are loci that form chromosome gaps or breaks when DNA synthesis is partially inhibited. Fragile sites are prone to deletions, translocations, and other rearrangements that can cause the inactivation of associated tumor suppressor genes in cancer cells. It was previously shown that ATR is critical to fragile-site stability and that ATR-deficient cells have greatly elevated fragile-site expression (A. M. Casper, P. Nghiem, M. F. Arlt, and T. W. Glover, Cell 111:779-789, 2002). Here we demonstrate that mouse and human cells deficient for BRCA1, due to mutation or knockdown by RNA interference, also have elevated fragile-site expression. We further show that BRCA1 functions in the induction of the G(2)/M checkpoint after aphidicolin-induced replication stalling and that this checkpoint function is involved in fragile-site stability. These data indicate that BRCA1 is important in fragile-site stability and that fragile sites are recognized by the G(2)/M checkpoint pathway, in which BRCA1 plays a key role. Furthermore, they suggest that mutations in BRCA1 or interacting proteins could lead to rearrangements at fragile sites in cancer cells.  相似文献
