At relatively high currents, fused anion-cation membranes give rise to rectifying and reactive effects. The rectification becomes less pronounced with increasing frequency. This effect results from changes in the concentration profiles of the ions during the positive and negative phases of the AC cycle. With reduction of the current, the voltage-current response becomes linear. The reactive effect can then be separated from the rectifying effect. The former effect can be attributed essentially to two factors: (a) the presence of transition regions of fixed charge and (b) the diffusion mechanism of the ions in an AC field. The first factor is largely frequency-independent and the second, frequency-dependent. A first approximation equivalent circuit is described. This circuit involves frequency-dependent elements. 相似文献

The mutation site in hemoglobin Rothschild (37 beta Trp----Arg) is located in the "hinge region" of the alpha 1 beta 2 interface, a region that is critical for normal hemoglobin function. The mutation results in greatly reduced cooperativity and an oxygen affinity similar to that of hemoglobin A [Gacon, G., Belkhodja, O., Wajcman, H., & Labie, D. (1977) FEBS Lett. 82, 243-246]. Crystal were grown under "low-salt" conditions [100 mM Cl- in 10 mM phosphate buffer at pH 7.0 with poly(ethylene glycol) as a precipitating agent]. The crystal structure of deoxyhemoglobin Rothschild and the isomorphous crystal structure of deoxyhemoglobin A were refined at resolutions of 2.0 and 1.9 A, respectively. The mutation-induced structural changes were partitioned into components of (1) tetramer rotation, (2) quaternary structure rearrangement, and (3) deformations of tertiary structure. The quaternary change involves a 1 degree rotation of the alpha subunit about the "switch region" of the alpha 1 beta 2 interface. The tertiary changes are confined to residues at the alpha 1 beta 2 interface, with the largest shifts (approximately 0.4 A) located across the interface from the mutation site at the alpha subunit FG corner-G helix boundary. Most surprising was the identification of a mutation-generated anion-binding site in the alpha 1 beta 2 interface. Chloride binds at this site as a counterion for Arg 37 beta. The requirement of a counterion implies that the solution properties of hemoglobin Rothschild, in particular the dimer-tetramer equilibrium, should be very dependent upon the concentration and type of anions present. 相似文献

We report differences in the thermal biology, elevational, temporal and geographic distributions of sympatric clones of the widespread asexual house gecko, Lepidodactylus lugubris. The two most common L. lugubris clones in Fiji, clones 2NA and 2NB, differ significantly in preferred temperature as measured in a laboratory heat gradient, but were similar in critical thermal maximum and minimum. Significant differences were found in the relative frequency of clones 2NA, 2NB, and a third Fijian clone, clone 3NB, at seven sites along an elevational gradient in Fiji. Clone 2NB was not collected at sites above 235 m, consistent with its higher preferred temperature, whereas clone 2NA was captured as high as 835 m. Clone 3NB was extremely rare at sealevel (1% of all individuals at three sites below 100 m), but predominated at the two highest-elevation sites (42% and 100%). Clones 2NA and 2NB did not differ significantly in their activity time or ambient activity temperature at low-elevation sites. Clone 3NB however, was active on significantly cooler nights at two of those sites. These significant inter-clonal differences in spatial and temporal distribution should allow a more complete utilization of resources by the assemblage of clones than by any single clonal genotype, and may promote coexistence of clones at a within-island and within-site scale. Clone 2NA, which is the most common clone in Fiji and has the broadest elevational distribution, also has the widest geographic distribution. It was the predominant clone at 27 of 34 sites surveyed in nine Pacific archipelagoes. This suggests that the ecological attributes that favor this clone in Fiji also favor it elsewhere in the Pacific despite differing environmental conditions and clonal composition in those areas. 相似文献

The region of the clock gene period (per) that encodes a repetitive tract
of threonine-glycine (Thr-Gly) pairs has been compared between Dipteran
species both within and outside the Drosophilidae. All the non-
Drosophilidae sequences in this region are short and present a remarkably
stable picture compared to the Drosophilidae, in which the region is much
larger and extremely variable, both in size and composition. The
accelerated evolution in the repetitive region of the Drosophilidae appears
to be mainly due to an expansion of two ancestral repeats, one encoding a
Thr-Gly dipeptide and the other a pentapeptide rich in serine, glycine, and
asparagine or threonine. In some drosophilids the expansion involves a
duplication of the pentapeptide sequence, but in Drosophila pseudoobscura
both the dipeptide and the pentapeptide repeats are present in larger
numbers. In the nondrosophilids, however, the pentapeptide sequence is
represented by one copy and the dipeptide by two copies. These observations
fulfill some of the predictions of recent theoretical models that have
simulated the evolution of repetitive sequences.

Island varanids seem to be an exception to the rule that territorial vertebrate taxa often become gigantic relative to mainland relatives when on islands, whereas non-territorial species become dwarfed (Case 1978). However, no systematic island/mainland studies have examined the empirical size trends in this group of carnivorous lizards. We perform such an analysis for the Australian region and critically evaluate various selective agents that might be responsible for size changes in several island populations. Insular gigantism occurs at least four times among the island populations examined. The magnitude of size change is positively correlated to prey abundance on the islands (as indirectly measured through a condition index of the lizards, essentially a measure of how fat they arc) and the size of prey: islands with large prey have large varanids and vice versa. Since the island population with the largest size change, the Reevesby Varanus rosenbergi, was introduced less than 100 years ago, these size changes can be quite rapid. This might indicate that selective coefficients are strong; however, we can not exclude the possibility that these size differences have no genetic component and simply reflect environmental differences in growth rate and shifts in age structure between island and mainland locations. 相似文献

The effects of vanadate on calcium homeostasis and enzyme secretion have been assessed in the incubated pancreas of young rats. Vanadate causes an acceleration of 45Ca efflux from pre-loaded uncinate glands; amylase release is reversibly increased for the duration of exposure to vanadate. Alkaline orthovanadate is most effective in eliciting these responses; its effects are greatly reduced at pH 7.4. However, changes in pH alone do not mimic these effects. Other vanadium oxides (metavanadate, vanadium pentoxide and vanadyl sulphate) are poor secretagogues. Alkaline ortho-, or meta-vanadate also causes an increased calcium uptake although this does not seem to be responsible for the observed secretory response. Intercellular gap junctions occur between the ciliated cells that make up the comb plates of the ctenophore Pleurobrachia. Similar junctions are found within the ciliated grooves which run from the apical organ to the first plate of each comb row, as well as throughout the endoderm of the meridional canals. Gap junctions were not found in the ectodermal tissue between the comb rows. The distribution of junctions suggests that excitation conduction within the ciliated grooves, comb plates and meridional canal endoderm may be epithelial. 相似文献

To understand the role of the renin-angiotensin-aldosterone system in the pathogenesis of human hypertension, in serial studies we have blocked the system using three different pharmacologic probes: 1) reduction of renin secretion by administration of the beta receptor blocker, propranolol; 2) blockade of the action of angiotensin II by infusion of saralasin, a competitive antagonist of angiotensin II; and 3) blockade of the enzymatic conversion of angiotensin I to angiotensin II by infusing a nonapeptide competitive inhibitor. The depressor responses induced by either propranolol or the nonapeptide expose a significant to major involvement of excess renin--angiotensin in maintaining the hypertension of some 50 to 70% of common forms of hypertension including "essential" hypertension. This subgroup includes nearly all patients with high or "normal" renin--sodium profiles. The considerably lower estimates for a renin factor in essential hypertension suggested by saralasin testing now appear due to the partial agonism of this drug. Further studies are required to determine whether this relative or absolute excess of renin secretion is primarily involved in the hypertension and if not why it fails to shut itself off. Similar studies of normal subjects are also needed to determine whether renin support of blood pressure is proportionately greater or less than in hypertensive subjects. Meanwhile the validation provided by these three different pharmacologic probes portends a burgeoning clinical role for renin--sodium profiling not only in screening for renal and adrenal cortical hypertensions but also for characterizing the vasoconstrictor and volume elements involved in various individual patients and thus enabling more specific treatments of the various subtypes of essential hypertension. 相似文献
