Many epithelia have a common planar cell polarity (PCP), as exemplified by the consistent orientation of hairs on mammalian skin and insect cuticle. One conserved system of PCP depends on Starry night (Stan, also called Flamingo), an atypical cadherin that forms homodimeric bridges between adjacent cells. Stan acts together with other transmembrane proteins, most notably Frizzled (Fz) and Van Gogh (Vang, also called Strabismus). Here, using an in vivo assay for function, we show that the quintessential core of the Stan system is an asymmetric intercellular bridge between Stan in one cell and Stan acting together with Fz in its neighbour: such bridges are necessary and sufficient to polarise hairs in both cells, even in the absence of Vang. By contrast, Vang cannot polarise cells in the absence of Fz; instead, it appears to help Stan in each cell form effective bridges with Stan plus Fz in its neighbours. Finally, we show that cells containing Stan but lacking both Fz and Vang can be polarised to make hairs that point away from abutting cells that express Fz. We deduce that each cell has a mechanism to estimate and compare the numbers of asymmetric bridges, made between Stan and Stan plus Fz, that link it with its neighbouring cells. We propose that cells normally use this mechanism to read the local slope of tissue-wide gradients of Fz activity, so that all cells come to point in the same direction. 相似文献

The ability to quickly develop germplasm having tolerance to several complex polygenic inherited abiotic and biotic stresses combined is critical to the resilience of cropping systems in the face of climate change.Molecular breeding offers the tools to accelerate cereal breeding;however,suitable phenotyping proto-cols are essential to ensure that the much-anticipated benefits of molecular breeding can be realized.To facilitate the full potential of molecular tools,greater emphasis needs to be given to reducing the within-experimental site variability,application of stress and characterization of the environment and appropriate phenotyping tools.Yield is a function of many processes throughout the plantcycle,and thus integrative traits that encompass crop performance over time or organization level(i.e.canopy level) will provide a better alternative to instantaneous measurements which provide only a snapshot of a given plant process.Many new phenotyping tools based on remote sensing are now available including non-destructive measurements of growth-related parameters based on spectral reflectance and infrared thermometry to estimate plant water status.Here we describe key field phenotyping protocols for maize with emphasis on tolerance to drought and low nitrogen. 相似文献

Background
Human values and folklore of wildlife strongly influence the effectiveness of conservation efforts. These values and folklore may also vary with certain demographic characteristics such as gender, age, or education. Reptiles and amphibians are among the least appreciated of vertebrates and are victims of many negative values and wrong ideas resulting from the direct interpretation of folklore. We try to demonstrate how these values and folklore can affect the way people relate to them and also the possible conservation impacts on these animals.Methods
A questionnaire survey distributed to 514 people in the district of Évora, Portugal, was used to obtain data regarding the hypothesis that the existence of wrong ideas and negative values contributes to the phenomenon of human-associated persecution of these animals. A structural equation model was specified in order to confirm the hypothesis about the possible relationships between the presence of perceptions and negative values about amphibians and reptiles and persecution and anti-conservation attitudes. Sociodemographic variables were also added.Results
The results of the model suggest that the presence of folklore and negative values clearly predicts persecution and anti-conservation attitudes towards amphibians and reptiles. Also, the existence of folklore varies sociodemographically, but negative values concerning these animals are widespread in the population.Conclusions
With the use of structural equation models, this work is a contribution to the study of how certain ideas and values can directly influence human attitudes towards herpetofauna and how they can be a serious conservation issue. 相似文献44.
Carboxylic acid transporters form a heterogeneous group of proteins, presenting diverse mechanisms of action and regulation, and belonging to several different families. Multiple physiological and genetic studies in several organisms, from yeast to mammals, have allowed the identification of various genes coding for carboxylate transporters. Detailed understanding of the metabolism and transport of these nutrients has become more important than ever, both from a fundamental and from an applied point of view. Under a biotechnological perspective, the increasing economic value of these compounds has boosted this field of research considerably. Fire modifies the germination of seeds of numerous species. One of the fire factors that cause these modifications is the
ash. This study analysed the germination of seeds of Pinus sylvestris L., Pinus nigra Arn. Pinus radiata D. Don and Pinus pinaster Aiton, subjected to different ash treatments, and the development of seedlings that grew in these conditions for 14 weeks.
We obtained the ash by completely burning leaves and small twigs from the most abundant woody species in populations of pines.
The ash treatments applied were as follows: Control (without ash), Low (half of the amount registered in a fire), Medium (equal
to the amount registered in a fire) and High (double the amount registered in a fire). Germination took place on paper inside
Petri dishes and in soil, and growth was only analysed for the plants that grew in the soil. For all species, and especially
P. nigra, germination rate decreased as the amount of ash applied increased. The negative effects of the ash were more apparent following
treatments in Petri dishes than in the soil. In the dishes, the average germination time varies little between the four species
of pine. In soil, the average germination time is more prolonged, and only some differences were observed between treatments
in P. sylvestris and P. nigra. In these species the high treatment significantly increased the average germination time. We found a greater sensitivity
of P. sylvestris and P. nigra to the addition of ash, showing a higher seedling mortality rate. However, it appears that seedling development (length and
weight) was not affected in any of the species through the addition of ash. These tests allow us to deduce that, in P. sylvestris, P. nigra, P. radiata and P. pinaster, the ash produced by forest fires has an inhibiting effect on germination and little effect on the development of seedlings
in the first months of life.

A method is described for evaluation of fat-soluble vitamin in human adipose tissue with the aim to obtain, accurately and within the shortest analysis time, a time-integrated measure of exposure to vitamins from the diet. Fat tissue was deproteinized with ethanol and extracted with n-hexane. Normal-phase HPLC was performed in a Lichrosorb Si60 column with a gradient of n-hexane–2-propanol at 1 ml/min. Detection was accomplished using a diode-array system (for retinol and β-carotene) in series with a fluorescence detector (α-tocopherol). The method was validated and applied to human adipose tissue in a total of 140 subjects. The mean contents found were 0.43, 0.84, 240.3 μg/g for retinol, β-carotene and α-tocopherol, respectively. The method is sensitive enough for detecting the compounds in 1.6 mg of adipose tissue considering the lowest concentration found. 相似文献

Abstract. Arthrocnemum macrostachyum is a perennial species acting as a primary colonizer of salt pans in Mediterranean high salt marshes. Salicornia ramosissima, an annual, occurs in salt pans under Arthrocnemum canopies and in open areas. The aim of this study was to analyse, in wild populations and a transplant experiment, how S. ramosissima population dynamics and growth are affected by A. macrostachyum. The environmental conditions within the patches of Arthrocnemum were less stressful than in the open areas, with lower radiation levels and salinity concentrations. In the inner areas of A. macrostachyum patches, density‐dependent mortality processes of S. ramosissima seedlings led to low densities of adult individuals with greater morphological development and reproductive success than in open areas. However, at the edges of Arthrocnemum patches facilitation of seedling survival favoured high densities. Environmental stress hindered development, decreased reproduction and premature death. These results are in agreement with the general theory of factors controlling vegetation distribution that biotic interactions dominate in low stress environments, while abiotic interactions dominate under harsher environmental conditions. A. macrostachyum plays an essential role in the succession in these salt pans, facilitating seed production and stimulating nucleation processes in S. ramosissima. 相似文献

The major fungal pathogen Candida albicans has the metabolic flexibility to assimilate a wide range of nutrients in its human host. Previous studies have suggested that C. albicans can encounter glucose‐poor microenvironments during infection and that the ability to use alternative non‐fermentable carbon sources contributes to its virulence. JEN1 encodes a monocarboxylate transporter in C. albicans and we show that its paralogue, JEN2, encodes a novel dicarboxylate plasma membrane transporter, subjected to glucose repression. A strain deleted in both genes lost the ability to transport lactic, malic and succinic acids by a mediated mechanism and it displayed a growth defect on these substrates. Although no significant morphogenetic or virulence defects were found in the double mutant strain, both JEN1 and JEN2 were strongly induced during infection. Jen1‐GFP (green fluorescent protein) and Jen2‐GFP were upregulated following the phagocytosis of C. albicans cells by neutrophils and macrophages, displaying similar behaviour to an Icl1‐GFP fusion. In the murine model of systemic candidiasis approximately 20–25% of C. albicans cells infecting the kidney expressed Jen1‐GFP and Jen2‐GFP. Our data suggest that Jen1 and Jen2 are expressed in glucose‐poor niches within the host, and that these short‐chain carboxylic acid transporters may be important in the early stages of infection. 相似文献

Glycophorin from the human erythrocyte membrane has been isolated in pure form and reconstituted into large unilamellar vesicles comprised of binary mixtures of 1,2-dipalmitoyl-3-sn-phosphatidylcholine (DPPC) and chain perdeuterated 1,2-dimyristoyl-3-sn-phosphatidylcholine (DMPC-d54). The effect of temperature and protein on lipid structure and mixing was monitored by using Fourier transform infrared spectroscopy; deuteration of one of the components of the mixture permits observation of the protein interaction with each lipid species. The melting curves were analyzed by assuming that each lipid chain can exist in one of two physical states (i.e., gel or liquid crystalline), characterized by a temperature-dependent Lorentzian distribution for the line shape of the C-H or C-D stretching vibrations. The fraction of each lipid component melted at temperatures within the two-phase region of the phase diagram was calculated and approximate phase diagrams were constructed. Addition of protein lowers the liquidus line of the phase diagram while leaving the solidus line essentially unchanged. No lipid phase separation is observed. The effect of protein is more pronounced on the DPPC component than on the DMPC-d54. The former is significantly more disordered and/or fluidized at all lipid mole fractions in the ternary system than in the binary phospholipid mixture. 相似文献
