The somatic chromosomes, obtained from short term leukocyte cultures, were studied of four species of the Hylobatidae: Hylobates lar, H. agilis, H. (Nomascus) concolor and Symphalangus syndactylus. In accordance with earlier observations by others, the diploid chromosome numbers were found to be 44 in both Hylobates lar and H. agilis, 52 in H. concolor and 50 in Symphalangus syndactylus. The chromosome associations observed in metaphase spreads are clearly different in the three types of chromosome complements. In Hylobates lar and H. agilis associations are found between both members of the marked chromosome pair. In Symphalangus syndactylus the only two acrocentric elements of the karyo-type, which are of medium size, associate frequently. In H. concolor finally, the members of three pairs of small acrocentrics are involved in chromosome associations. G-banding patterns (obtained by trypsin treatment) showed that in a male individual of this species also the small acrocentric Y chromosome sometimes participates in these associations. The evolutionary aspects of these observations are briefly discussed.  相似文献

Summary A novel human prostatic stromal cell culture, designated DuK50, has been passedin vitro >12 mo. Tissue cultures were obtained from material harvested within a normal region of a radical prostatectomy specimen. These monolayers exhibited normal fibroblastic characteristics with each cell having a flattened, elongated appearance. Karyotypic analysis revealed a normal, male 46, XY chromosomal content with no numerical or structural abnormalities. DNA analysis using a Cell Analysis Systems Image Analyzer confirmed a euploid DNA content (7.9 pg DNA). Cellular markers for verification of stromal cell type were performed by immunohistochemical techniques. DuK50 stained positive for vimentin and fibronectin. Immunostains for epithelial cytokeratins and prostate-specific antigen were negative, which ruled out contamination with prostatic epithelial cells. Negative immunostaining with desmin monoclonal antibody and light staining with smooth muscle actin alpha is consistent with the staining pattern of myofibroblasts. Response to various androgens, measured by a microculture tetrazolium assay technique, revealed a significant growth stimulation of DuK50. Soft agar invasiveness assays and tumorigenicity studies in nude mice were negative. DuK50 exhibits a rapid doubling time with excellent plating efficiency, thrives in a readily available media supplemented with fetal bovine serum, and passes with routine trypsin protocols. The availability of this prostatic stromal cell culture may facilitate studies on this cell type’s role in growth factor modulation, drug and steroid metabolism, and stromal-epithelial interactions in the prostate. A mannosyltransferase has been detected in suspensiosn of BALB/c fibroblasts incubated with GDP-[14C]mannose. The experimental evidence indicates the cell surface as the most likely site for the enzyme. The transferase synthesizes both glycolipids and glycoproteins. The lipid compounds have properties suggestive of lipid-linked mono- and oligosaccharides which can function as intermediates in glycoprotein synthesis. The formation of these compounds by a cell surface enzyme suggests that lipid-linked intermediates may play an important role in the glycosylation of membrane components.  相似文献

We developed six microsatellite markers for Amphitmetus transversus (Kolbe, 1897) (Coleoptera, Curculionidae), a beetle widely distributed in the guineo‐congolian rainforests of east Africa. The numbers of alleles per locus range from three to eight, with a mean number of 4.6. They will be used for population genetic analysis. The ongoing study concentrates on the genetic differentiation and variability of isolated populations in the Kakamega Forest, a highly degraded and fragmented rainforest remnant in western Kenya.  相似文献

H-2-Linked regulation of serum gp70 production in mice   总被引:3,自引:0,他引:3  
By using many congenic strains of C57BL/10 (B10) mice and NZB mice, we have demonstrated a genetic system that controls the production of serum gp70. Our system has been tentatively designated as Sgp-1 and is composed of three phenotypes, Sgp-1a, 1b, and 1c. This system appears to be closely linked to, but not in the H-2 region on chromosome 17. Sgp-1a, which is associated with H-2d, correlates with relatively large amounts of serum gp70 in B10 congenic lines. Sgp-1b, which appeared in most of the H-2 types tested, corresponds with low serum gp70 output in B10 congenic lines and F1 hybrid offspring of B10 and NZB crosses and with increased gp70 production after lipopolysaccharide (LPS) stimulation. Sgp-1c, which is associated with H-2s, also relates to small amounts of serum gp70 in B10 congenic lines and their F1 hybrids from NZB matings, but shows lack of serum gp70 responsiveness to LPS. This failure to accelerate serum gp70 production after injection of LPS is independent of other acute phase responses and polyclonal activation of B cells.  相似文献

Synthetic salmon calcitonin (sCT) given SC (30 MRC U/kg) or ICV (1.4 U, 300 ng) inhibited 24-hr food consumption in the rat by 50–100%. Furthermore, ICV doses of sCT ranging from 75 ng (0.35 U) to 300 ng (1.4 U) lowered both food and water intake in a dose-dependent manner. Use of various forms of CT give an apparent order of potency of salmon>porcine≥human with sCT being at least 40 fold more potent than the mammalian forms. Measurement of food and water intake over a brief (30–90 min) period showed that ICV sCT was effective in reducing food and water consumption regardless of whether it had been given 1, 12, or 23 hr previously. Daily administration of sCT for 5 days caused marked suppression of food and water intake for 2 days; thereafter, consumption returned toward normal, becoming equivalent to vehicle injected controls by day 3 or 4 despite continued daily injections of sCT. The results show that CT can act centrally to modify certain types of behavior and are of special interest since CT-like peptides have been described in the pituitary and hypothalamus and since CT receptors have been reported in hypothalamic and other brain regions.  相似文献
