A novel fluorophore was isolated from human lenses using high performance liquid chromatography (HPLC). The new fluorophore was well separated from 3-hydroxykynurenine glucoside (3-OHKG) and its deaminated isoform, 4-(2-amino-3-hydroxyphenyl)-4-oxobutanoic acid O-glucoside, which are known UV filter compounds. The new compound exhibited UV absorbance maxima at 260 and 365 nm, was fluorescent (Ex(360 nm)/Em(500 nm)), and increased in concentration with age. Further analysis of the purified compound by microbore HPLC with in-line electrospray ionization mass spectrometry revealed a molecular mass of 676 Da. This mass corresponds to that of an adduct of GSH with a deaminated form of 3-OHKG. This adduct was synthesized using 3-OHKG and GSH as starting materials. The synthetic glutathionyl-3-hydroxykynurenine glucoside (GSH-3-OHKG) adduct had the same HPLC elution time, thin-layer chromatography R(F) value, UV absorbance maxima, fluorescence characteristics, and mass spectrum as the lens-derived fluorophore. Furthermore, the (1)H and (13)C NMR spectra of the synthetic adduct were entirely consistent with the proposed structure of GSH-3-OHKG. These data indicate that GSH-3-OHKG is present as a novel fluorophore in aged human lenses. The GSH-3-OHKG adduct was found to be less reactive with beta-glucosidase compared with 3-OHKG, and this could be due to a folded conformation of the adduct that was suggested by molecular modeling. 相似文献

The serological response in burros and horses to the viable LVS strain of Pasteurella tularensis was studied. High-titered agglutinating antisera and fluorescent-antibody conjugates were obtained in both groups of animals. Maximum titers were obtained in horses 14 to 21 days after the start of vaccination and in burros 21 to 28 days after the start of vaccination. In laboratory trials to investigate the parasite/host spectra of certain aphid pests and hymenopterous parasites, the aphidAcyrthosiphon kondoi Shinji encapsulated the egg of the aphelinid parasiteAphelinus asychis Walker. The resultant brown, sclerotic capsule was formed within 24 h of exposure of the aphid to parasitization and as far as is known prevented the development of the parasite to the larval stage. The capsule remained throughout the life of the aphid, whose longevity and fecundity were apparently not seriously impaired. A small number ofAphelinus escaped encapsulation, especially in aphids already containing capsule(s), and developed into normal, reproductive adults.A. kondoi did not encapsulate, andA. asychis was not encapsulated by any other species. Male and female Tenebrio molitor act as intermediate hosts for metacestodes of the rat tapeworm, Hymenolepis diminuta. It is known that the bean-shaped accessory glands of infected males exhibit an extended growth period and are significantly larger than those from noninfected males by day 10 after emergence. We wished to determine whether more material is transferred from these glands into the spermatophores. Here we report that the protein content and trehalase activity of spermatophores produced by bean-shaped accessory glands from infected males is elevated. However, protein transferred to the female spermatheca during mating was not affected by the infection status of the male. No evidence of transfer of trehalase to the spermatheca was detected but spermatheca from virgin, infected females contained significantly greater trehalase activity than those from noninfected females. 相似文献

Summary The use of reticulated polyurethane foam as a support material for the immobilization of methanogenic associations and its application to the anaerobic treatment of fine particulate solid wastes was investigated. The colonization of polyurethane support particles in a continuous upflow reactor fed on a mixture of acetate, propionate and butyrate, was both rapid and dense. To understand how environmental change will modify community assembly and the distribution of organisms it is valuable to
understand mechanisms that drive the occurrence of organisms across the landscape. Salinisation of agricultural land in southwest
Western Australia, as a result of land clearing, is a widespread environmental change, which threatens numerous taxa, but
provides an opportunity to elucidate such mechanisms. Although salinisation affects terrestrial fauna and flora, the greatest
impacts are seen in wetlands and waterways. Many aquatic insect taxa colonise ephemeral water bodies directly as adults or
by oviposition. Few empirical studies, however, evaluate the influence of abiotic factors, such as water body salinity, on
the colonisation behaviour of aquatic fauna. We conducted a manipulative experiment using mesocosms to test whether colonising
insect fauna select aquatic habitats based upon salinity. We found that halosensitive fauna selected less saline mesocosms
for oviposition and colonisation, demonstrating that behaviour can influence the distribution of aquatic organisms. Additionally,
we utilised field surveys of insects from ephemeral water bodies across a broad region of southwest Western Australia to determine
if mesocosm results reflected field observation. The abundance of the same insect taxa and taxonomic groups in the field were
highly variable and, with the exceptions of Culex australicus Dobrotworksy and Drummond and Anopheles annulipes Giles (Diptera: Culicidae), did not show similar patterns of distribution to those observed in the mesocosm experiment. Both
mesocosm and field assemblages exhibited similar and significant trajectories associated with the salinity gradient, even
though there were differences in assemblage structure between the two. Our findings give empirical support to the importance
of behaviour in the spatial distribution and assembly of some aquatic insects.
