Pathogenesis-related proteins (PRs) are plant proteins produced in leaves in response to infection by pathogens including viruses, viroids, fungi and bacteria. Information on the presence and/or expression of PRs in monocotyledonous plants is scarce. Here we report the identification of cDNA and genomic clones coding for a basic form of a protein from germinating maize seeds having a high homology with the group of PR-1 from tobacco.A cDNA library enriched in aleurone-specific sequences was prepared from maize seeds two days after germination. One clone was found to contain an open reading frame encoding a protein homologous to PR proteins from tomato (p14) and tobacco (PR-1 group). Sequence analysis of the corresponding genomic clone revealed that it was encoded by a single exon. Besides, DNA blot hybridization indicates that this PR-like protein is encoded by a single-copy gene in maize. The accumulation of its mRNA increases after rehydration of desiccated seeds. Furthermore, a relationship was found between its expression and infection by a natural pathogen of maize, the fungus Fusarium moniliforme. The possible role of this protein as a response mechanism following fungal infection in cereal seeds is discussed.  相似文献

In vivo metabolism of calcitriol in the pregnant rabbit doe   总被引:1,自引:0,他引:1  
The production rate, the disappearance rate and the half life of calcitriol in gravid rabbit does at 24 days of gestation were compared, under unstressed steady state conditions, to those of nonpregnant animals. The contribution of the fetoplacental unit to the circulating levels of fetal calcitriol was also assessed. The calcitriol levels (139.6 +/- 19.9 vs 55.3 +/- 8.8 pmol/l and production rates (113.9 +/- 8.8 vs 59.2 +/- 9.2 pmol/min/Kg) were higher in pregnant than in nonpregnant animals (P less than 0.01). However, clearance rates (1.07 +/- 0.18 vs 1.12 +/- 0.20 ml/min/Kg and circulating half life (442 +/- 49 vs 368 +/- 35 min; NS) were similar in both groups of animals. Fetal levels (62.3 +/- 1.6 pmol/l) and specific activity (11166 +/- 864 dpm/pmol) of calcitriol were lower than those of the respective mothers (P less than 0.005). Taken together these data suggest that, calcitriol is transported through the placenta; and that the fetoplacental unit contributes to the fetal and perhaps to the maternal calcitriol levels.  相似文献

Aim In this study we present a new view on the extinction of Myotragus balearicus, an extinct highly modified dwarf caprine from the Gymnesic Islands (or eastern Balearic Islands), as a methodological case study for interpretation of Late Quaternary extinctions (LQEs). Methods We analyse all available 14C ages obtained from M. balearicus bones from the uppermost part of the Pleistocene and the Holocene, together with the available chronological data of the putative causes of Myotragus extinction. Results It has been possible to define two critical dates that allow us to establish an ‘uncertainty period for the Myotragus extinction’ (UPME) in each analysed island (Mallorca, Menorca and Cabrera). For Mallorca, the UPME corresponds to the interval c. 3700 to 2030 calbc (i.e. c. 1670 years of uncertainty). In the case of Menorca, the UPME spans from 10,000 to 1930 calbc (8070 years of uncertainity). In Cabrera the UPME is placed between 3650 and 300 calbc (3350 years of uncertainty). These periods, together with a review of the available information on the chronology of human arrival and the chronology of Holocene climatic change, shed light on the possible causes of the extinction of this species. Main conclusions Extinction of Myotragus because of climatic change can be definitively rejected. The Myotragus extinction must be attributed to the rapid effects of the first human occupation. The use of uncertainity periods for the disappearance of species represents a useful tool for the analysis of LQEs.  相似文献

The progression of autoimmune responses is associated with an avidity maturation process driven by preferential expansion of high avidity clonotypes at the expense of their low avidity counterparts. Central and peripheral tolerance hinder the contribution of high-avidity clonotypes targeting residues 206-214 of islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP(206-214)) during the earliest stages of autoimmune diabetes. In this study, we probe the molecular determinants and biochemical consequences of IGRP(206-214)/K(d) recognition by high-, intermediate-, and low-avidity autoreactive CD8(+) T cells, and we investigate the effects of genetic IGRP(206-214) silencing on their developmental biology. We find that differences in avidity for IGRP(206-214)/K(d) map to CDR1α and are associated with quantitative differences in CD3ε proline-rich sequence exposure and Nck recruitment. Unexpectedly, we find that tolerance of high-avidity CD8(+) T cells, unlike their activation and recruitment into the pancreas, is dissociated from recognition of IGRP(206-214), particularly in adult mice. This finding challenges the view that tolerance of pathogenic autoreactive T cells is invariably triggered by recognition of the peptide-MHC complex that drives their activation in the periphery, indicating the existence of mechanisms of tolerance that are capable of sensing the avidity, hence pathogenicity of autoreactive T cells without the need to rely on local autoantigen availability.  相似文献
