The African swine fever virus (ASFV) g5R gene encodes a protein containing a Nudix hydrolase motif which in terms of sequence appears most closely related to the mammalian diadenosine tetraphosphate (Ap4A) hydrolases. However, purified recombinant g5R protein (g5Rp) showed a much wider range of nucleotide substrate specificity compared to eukaryotic Ap4A hydrolases, having highest activity with GTP, followed by adenosine 5'-pentaphosphate (p5A) and dGTP. Diadenosine and diguanosine nucleotides were substrates, but the enzyme showed no activity with cap analogues such as 7mGp3A. In common with eukaryotic diadenosine hexaphosphate (Ap6A) hydrolases, which prefer higher-order polyphosphates as substrates, g5Rp also hydrolyzes the diphosphoinositol polyphosphates PP-InsP5 and [PP]2-InsP4. A comparison of the kinetics of substrate utilization showed that the k(cat)/K(m) ratio for PP-InsP5 is 60-fold higher than that for GTP, which allows classification of g5R as a novel diphosphoinositol polyphosphate phosphohydrolase (DIPP). Unlike mammalian DIPP, g5Rp appeared to preferentially remove the 5-beta-phosphate from both PP-InsP5 and [PP]2-InsP4. ASFV infection led to a reduction in the levels of PP-InsP5, ATP and GTP by ca. 50% at late times postinfection. The measured intracellular concentrations of these compounds were comparable to the respective K(m) values of g5Rp, suggesting that one or all of these may be substrates for g5Rp during ASFV infection. Transfection of ASFV-infected Vero cells with a plasmid encoding epitope-tagged g5Rp suggested localization of this protein in the rough endoplasmic reticulum. These results suggest a possible role for g5Rp in regulating a stage of viral morphogenesis involving diphosphoinositol polyphosphate-mediated membrane trafficking. 相似文献

Numerous transposed sequences of mitochondrial cytochrome oxidase I-II in aphids of the genus Sitobion (Hemiptera: Aphididae) 总被引:4,自引:1,他引:3
Polymerase chain reaction (PCR) products corresponding to 803 bp of the
cytochrome oxidase subunits I and II region of mitochondrial DNA (mtDNA
COI-II) were deduced to consist of multiple haplotypes in three Sitobion
species. We investigated the molecular basis of these observations. PCR
products were cloned, and six clones from one individual per species were
sequenced. In each individual, one sequence was found commonly, but also
two or three divergent sequences were seen. The divergent sequences were
shown to be nonmitochondrial by sequencing from purified mtDNA and Southern
blotting experiments. All seven nonmitochondrial clones sequenced to
completion were unique. Nonmitochondrial sequences have a high proportion
of unique sites, and very few characters are shared between
nonmitochondrial clones to the exclusion of mtDNA. From these data, we
infer that fragments of mtDNA have been transposed separately (probably
into aphid chromosomes), at a frequency only known to be equalled in
humans. The transposition phenomenon appears to occur infrequently or not
at all in closely related genera and other aphids investigated. Patterns of
nucleotide substitution in mtDNA inferred over a parsimony tree are very
different from those in transposed sequences. Compared with mtDNA,
nonmitochondrial sequences have less codon position bias, more even
exchanges between A, G, C and T, and a higher proportion of nonsynonymous
replacements. Although these data are consistent with the transposed
sequences being under less constraint than mtDNA, changes in the
nonmitochondrial sequences are not random: there remains significant
position bias, and probable excesses of synonymous replacements and of
conservative inferred amino acid replacements. We conclude that a
proportion of the inferred change in the nonmitochondrial sequences
occurred before transposition. We believe that Sitobion aphids (and other
species exhibiting mtDNA transposition) may be important for studying the
molecular evolution of mtDNA and pseudogenes. However, our data highlight
the need to establish the true evolutionary relationships between sequences
in comparative investigations.

Aim The aim of this paper is to analyse fossil charcoal deposits, largely identified to the species level and spanning a sequence from the late Holocene to < 40,000 BP , in order to reconstruct Late Quaternary vegetation and climatic patterns in the western (winter-rainfall) fynbos biome of South Africa. Location The charcoals were excavated from the Elands Bay Cave (32°19S, 18°20E) on the semiarid (200–250 mmyr?1), winter-rainfall coastline of the western fynbos biome. Methods Patterns in the charcoal data set over time were sought by manual sorting of the charcoal×sample matrix, as well as by subjecting the data to multivariate analysis. Palaeoclimatic reconstruction was attempted by comparing the climatic controls on contemporary vegetation communities that resembled the fossil assemblages. Charcoal diversity was modelled using sample age and number of charcoal fragments as explanatory variables. Results The fossil assemblages ranged from xeric communities (similar to those presently occurring at the site) during the Holocene, to more mesic thicket and fynbos vegetation in the terminal Pleistocene, to Afromontane forest and riverine woodland communities after about 18,000 BP . Diversity of the charcoal samples increased monotonically with increasing sample age. Main conclusions The results suggest that, unlike the eastern fynbos biome, which is under fundamentally different climatic controls, soil moisture conditions in the western part of the biome were higher in the Last Glacial than during the Holocene. This scenario may help to explain the higher regional richness and associated diversification in the western than eastern part of the biome. 相似文献

Dietary fats are efficiently absorbed in the small intestine and transported into the blood via the lymph as chylomicrons, despite enormous variations in the amount and composition of the dietary lipid. The aim of the present study was to investigate how enterocytes respond to increased dietary fats of different composition. Rabbits were fed a low fat chow diet, and chow supplemented with sunflower oil (high n-6 polyunsaturated fatty acids), fish oil (high n-3 polyunsaturated fatty acids), or an oil mixture of a composition similar to that of the typical western diet. Feeding fat for 2 weeks markedly stimulated the ability of the isolated enterocytes to synthesize and secrete apolipoprotein B48, triacylglycerol, and cholesteryl ester (up to 18-, 50-, and 80-fold, respectively) in particles of chylomicron density. The magnitude of stimulation was sunflower oil > western diet lipid > fish oil. Single doses of lipid given 18 h prior to isolation of enterocytes stimulated chylomicron secretion by only 10% of that observed after 2 weeks of dietary supplementation. Enterocytes are replaced rapidly (half-life 1-2 days) by cells which move from the crypts to the tips of the villi, where absorption of nutrients takes place.Our observations suggest that dietary lipids modulate the function of enterocytes as they move from the crypts, so that the cells are 'turned-on' to lipid absorption. The results also show that diets of different fatty acid composition vary in their effects. 相似文献
