The present study compares four different sampling methods potentially applicable to detection of Salmonella in broiler flocks, based on collection of faecal samples (i) by hand, 300 fresh faecal samples (ii) absorbed on five sheets of paper (iii) absorbed on five pairs of socks (elastic cotton tubes pulled over the boots and termed 'socks') and (iv) by using only one pair of socks. Twenty-three broiler flocks were included in the investigation and 18 of these were found to be positive by at least one method. Seven serotypes of Salmonella with different patterns of transmission (mainly horizontal or vertical) were found in the investigation. The results showed that the sock method (five pairs of socks) had a sensitivity comparable with the hand collection method (60 pools of five faecal samples); the paper collection method was inferior, as was the use of only one pair of socks. Estimation of the effective pool sizes for the different methods was also carried out.  相似文献

Automatic click detectors and full-bandwidth sound recorders are widely used in passive acoustic monitoring of small cetaceans. Detection of these signals depends on a variety of factors, including signal to noise ratio. Passive acoustic monitoring is often used to study impact of underwater noise on small cetaceans, but as detection probability is affected by changes in signal to noise ratio, variable noise levels may affect conclusions drawn from these experiments. Therefore, we examine how different detectors and filters perform in varying ocean noise conditions. C-PODs and full-bandwidth recorders (Wildlife Acoustics, SM2M+) were deployed at two stations in an environment with fluctuating ambient noise for 42 days. Noise level and harbour porpoise (Phocoena phocoena) click trains simultaneously recorded on both loggers were compared. Overall, we found that porpoise click detections by the algorithm used to analyse full-band recorder data (Pamguard) paralleled detections by the C-POD. However, Pamguard detected significantly more clicks than the C-POD. A decrease in detections was seen for both loggers with increasing noise in the band 20 –160 kHz, in particular for levels above 100 dB re 1μPa rms. We also found that the Pamguard detection function changed the least over varying noise conditions when compared to the C-POD detectors. This study sheds light on the fact that inference of animal presence/absence or density that are based on echolocation cues (here, Porpoise Positive Minutes) shall account for the acoustic environments where probability of detecting signals may be affected by variability in ambient noise levels.  相似文献

Background

Expression of soluble CD163 (sCD163), a macrophage/microglia biomarker, is increased in inflammatory conditions, and sCD163 levels in the cerebrospinal fluid (CSF) have recently been shown to be elevated in patients with multiple sclerosis (MS): the sCD163 CSF/serum ratio was elevated in patients with relapsing-remitting MS (RRMS), primary progressive MS (PPMS), and clinically isolated syndrome (CIS) compared with symptomatic controls.

Objective

To investigate the contributions of the sCD163 CSF/serum ratio to a biomarker panel focusing on inflammation and axonal degeneration in newly diagnosed MS; thus optimising a diagnostic biomarker panel for MS.

Methods

After a full MS diagnostic work-up, including collection of paired samples of CSF and serum, 125 patients were included in this study. Patients were divided into groups based on their diagnosis, and patients with normal clinical and paraclinical findings were defined as symptomatic controls. Serum and CSF levels, ratios, and indices of sCD163, CXCL13, osteopontin, neopterin, and CSF levels of neurofilament light polypeptide were determined by enzyme-linked immunosorbent assays (ELISAs). For sCD163 the results constitute a post-hoc analysis of already published data.

Results

All tested biomarkers, notably the sCD163 ratio, the CXCL13 ratio, the NEO ratio, the CSF level of NfL, the IgG index, and the serum level of OPN, were significantly correlated to RRMS, PPMS, and/or CIS. The individual biomarkers in single tests had a lower performance than the IgG index, however, their combined receiver operating characteristic (ROC) curve demonstrated excellent diagnostic discriminatory power.

Conclusion

The biomarker panel showed distinct profiles for each patient group and could be a valuable tool for clinical differentiation of MS subgroups. The combined ROC analysis showed that sCD163 contributes positively as a diagnostic marker to a panel of established MS biomarkers. Patients with PPMS were demonstrated to have significantly elevated levels of both inflammatory and degenerative markers.  相似文献

Aim In order to advance our understanding of the assembly of communities on islands and to elucidate the function of different islands in creating regional and subregional distribution patterns, we identify island biogeographical roles on the basis of the distribution of the islands’ biota within the archipelago. We explore which island characteristics determine island biogeographical roles. Furthermore, we identify biogeographical subregions, termed modules. Location Wallacea in Indonesia, and the West Indies in the Caribbean Sea. Methods We use a network approach to detect island biogeographical roles and avian biogeographical modules. To designate the biogeographical role of an island, each island is assigned two coordinates, l and r. The position of an island in lr space characterizes its role, namely as peripheral, connector, module hub, or network hub. Island characteristics are tested as predictors of l and r. Results Both Wallacea and the West Indies were found to be significantly modular and divided into four biogeographical modules. The four modules identified within Wallacea each contain all existing island roles, whereas no module in the West Indies represents all possible roles. Island area and elevation appeared to be the most important determinants of an island’s l score, while measurements of isolation essentially determined the r score. Main conclusions In both Wallacea and the West Indies, the geographic structuring into biogeographical modules corresponds well with our knowledge of past connections and contemporary factors. In both archipelagos, large, mountainous islands are identified as hubs and are thus responsible for faunal coherence within modules (module hubs) and across the entire archipelago (network hubs). We thus interpret these as source islands for the surrounding islands in their module (module hubs) or for the entire archipelago (network hubs). Islands positioned marginally in their module and distant from the mainland are identified as connectors or network hubs, behaving as sinks and stepping stones for dispersing species. Modularity and predictors of biogeographical roles are similar for Wallacea and the West Indies, whereas the build‐up of biogeographical modules and the assortment of roles depend on the spatial constellation of islands in each archipelago.  相似文献

Biogeographical systems can be analyzed as networks of species and geographical units. Within such a biogeographical network, individual species may differ fundamentally in their linkage pattern, and therefore hold different topological roles. To advance our understanding of the relationship between species traits and large‐scale species distribution patterns in archipelagos, we use a network approach to classify birds as one of four biogeographical species roles: peripherals, connectors, module hubs, and network hubs. These roles are based upon the position of species within the modular network of islands and species in Wallacea and the West Indies. We test whether species traits – including habitat requirements, altitudinal range‐span, feeding guild, trophic level, and body length – correlate with species roles. In both archipelagos, habitat requirements, altitudinal range‐span and body length show strong relations to species roles. In particular, species that occupy coastal‐ and open habitats, as well as habitat generalists, show higher proportions of connectors and network hubs and thus tend to span several biogeographical modules (i.e. subregions). Likewise, large body size and a wide altitudinal range‐span are related to a wide distribution on many islands and across several biogeographical modules. On the other hand, species restricted to interior forest are mainly characterized as peripherals and, thus, have narrow and localized distributions within biogeographical modules rather than across the archipelago‐wide network. These results suggest that the ecological amplitude of a species is highly related to its geographical distribution within and across bio geographical subregions and furthermore supports the idea that large‐scale species distributions relate to distributions at the local community level. We finally discuss how our biogeographical species roles may correspond to the stages of the taxon cycle and other prominent theories of species assembly.  相似文献

Human lymphocytes were exposed in vitro to therapeutic levels of ultrasound (1 W/cm2, CW, 0.87 MHz, durations of 80 and 160 sec). There were no significant differences in sister-chromatid exchange frequencies between controls and ultrasound-exposed cells. Exposure of lymphocytes to the positive control (mitomycin C) resulted in a significant increase in sister-chromatid exchanges. The data do not verify a report by Stella et al. (Mutation Res., 138 (1984) 75-85) that such exposures result in increased frequencies of SCEs.  相似文献

Coastal waters have strong gradients in dissolved organic matter (DOM) quantity and characteristics, originating from terrestrial inputs and autochthonous production. Enclosed seas with high freshwater input therefore experience high DOM concentrations and gradients from freshwater sources to more saline waters. The brackish Baltic Sea experiences such salinity gradients from east to west and from river mouths to the open sea. Furthermore, the catchment areas of the Baltic Sea are very diverse and vary from sparsely populated northern areas to densely populated southern zones. Coastal systems vary from enclosed or open bays, estuaries, fjords, archipelagos and lagoons where the residence time of DOM at these sites varies and may control the extent to which organic matter is biologically, chemically or physically modified or simply diluted with transport off-shore. Data of DOM with simultaneous measurements of dissolved organic (DO) nitrogen (N), carbon (C) and phosphorus (P) across a range of contrasting coastal systems are scarce. Here we present data from the Roskilde Fjord, Vistula and Öre estuaries and Curonian Lagoon; four coastal systems with large differences in salinity, nutrient concentrations, freshwater inflow and catchment characteristics. The C:N:P ratios of DOM of our data, despite high variability, show site specific significant differences resulting largely from differences residence time. Microbial processes seemed to have minor effects, and only in spring did uptake of DON in the Vistula and Öre estuaries take place and not at the other sites or seasons. Resuspension from sediments impacts bottom waters and the entire shallow water column in the Curonian Lagoon. Finally, our data combined with published data show that land use in the catchments seems to impact the DOC:DON and DOC:DOP ratios of the tributaries most.

Objective

The Inuit population in Greenland has undergone rapid socioeconomic and nutritional changes simultaneously with an increasing prevalence of obesity. Therefore, the objective was to examine fetal programming as part of the aetiology of obesity among Inuit in Greenland by investigating the association between birth weight and measures of body composition and fat distribution in adulthood.

Methods

The study was based on cross-sectional data from a total of 1,473 adults aged 18–61 years in two population-based surveys conducted in Greenland between 1999–2001 and 2005–2010. Information on birth weight was collected from birth records. Adiposity was assessed by anthropometry, fat mass index (FMI), fat-free mass index (FFMI), and visceral (VAT) and subcutaneous adipose tissue (SAT) estimated by ultrasound. The associations to birth weight were analyzed using linear regression models and quadratic splines. Analyses were stratified by sex, and adjusted for age, birthplace, ancestry and family history of obesity.

Results

Spline analyses showed linear relations between birth weight and adult adiposity. In multiple regression analyses, birth weight was positively associated with BMI, waist circumference, FMI, FFMI and SAT with generally weaker associations among women compared to men. Birth weight was only associated with VAT after additional adjustment for waist circumference and appeared to be specific and inverse for men only.

Conclusions

Higher birth weight among Inuit was associated with adiposity in adulthood. More studies are needed to explore a potential inverse association between birth size and VAT.  相似文献

Aim Wallacea, the vast oceanic boundary between the Oriental and Australian regions, contains 122 true nectarivorous bird species. It is the contact zone of the Oriental sunbirds and the Australian honeyeaters, and at least three more true nectarivorous families are resident in the region. An island–bird matrix for Wallacea was tested for the presence of two patterns, nestedness and modularity. If the matrix is modular, it consists of a number of densely linked subgroups or modules of islands and birds, which are weakly interconnected. These modules are used as a new tool in analysing biogeographical boundaries in Wallacea. Location Wallacea, Indonesia. Methods We constructed an island–bird matrix for Wallacea and used two algorithms, aninhado and sa , to test it for nestedness and modularity, respectively. aninhado calculates the matrix temperature and provides a null model, and sa is a module‐detecting program based on simulated annealing. The results of the sa were compared with those from a hierarchical cluster analysis. Results The matrix had a nested pattern, as is commonly the case for island–species matrices. The SA detected four modules in Wallacea, each consisting of a group of islands sharing a group of nectarivorous birds. The sa algorithm produced a more detailed pattern of the area than did the hierarchical cluster analysis. Main conclusions Modularity and nestedness do not preclude each other as biogeographical patterns. The boundaries of the modules detected by sa compared well with major boundaries from the existing literature and showed a clear division of Wallacea into modules of birds and islands closely linked together. Thus modules are biogeographical units of islands sharing a specific nectarivorous fauna. For some research questions, we suggest that modules may be more appropriate biogeographical units than single islands or traditionally perceived archipelagos. The nectarivorous families showed distinctly different distributions, indicating variation in their colonization history and speciation processes. We recommend sa as a tool for detecting fine‐grained biogeographical patterns.  相似文献
