Heparan sulfate 3-O-sulfotransferase transfers sulfate to the 3-OH position of a glucosamine to generate 3-O-sulfated heparan sulfate (HS), which is a rare component in HS from natural sources. We previously reported that 3-O- sulfotransferase isoform 5 (3-OST-5) generates both an antithrombin-binding site to exhibit anticoagulant activity and a binding site for herpes simplex virus 1 glycoprotein D to serve as an entry receptor for herpes simplex virus. In this study, we characterize the substrate specificity of 3-OST-5 using the purified enzyme. The enzyme was expressed in insect cells using the baculovirus expression approach and was purified by using heparin-Sepharose and 3',5'-ADP- agarose chromatographies. As expected, the purified enzyme generates both an antithrombin binding site and a glycoprotein D binding site. We isolated IdoUA-AnMan3S and IdoUA-AnMan3S6S from nitrous acid-degraded 3-OST-5-modified HS (pH 1.5), suggesting that 3-OST-5 enzyme sulfates the glucosamine residue that is linked to an iduronic acid residue at the nonreducing end. We also isolated a disaccharide with a structure of DeltaUA2S-GlcNS3S and a tetrasaccharide with a structure of DeltaUA2S-GlcNS-IdoUA2S-GlcNH23S6S from heparin lyases-digested 3-OST-5-modified HS. Our results suggest that 3-OST-5 enzyme sulfates both N-sulfated glucosamine and N-unsubstituted glucosamine residues. Taken together, the results indicate that 3-OST-5 has broader substrate specificity than those of 3-OST-1 and 3-OST-3. The unique substrate specificity of 3-OST-5 serves as an additional tool to study the mechanism for the biosynthesis of biologically active HS.  相似文献

We describe the first validated scintillation proximity assay (SPA) binding method for quantitation of 3H-labeled d-lysergic acid diethylamide (LSD) binding to recombinant human 5-hydroxytryptamine 6 (5-HT6) receptors expressed in Chinese hamster ovary (CHO)-Dukx and HeLa cells. The assay was developed using intact cells as a receptor source because membrane fractions derived from these cells failed to discern specific binding from a high level of nonspecific binding. The pharmacological binding profile of seven 5-HT6 agonists and antagonists using intact CHO-Dukx/5-HT6 cells in the SPA format was similar to data obtained from a filtration binding assay using HeLa/5-HT6 membranes. Ki values and rank order of potencies obtained in the SPA format were consistent with published filtration data as follows: SB-271046 (Ki = 1.9 nM) > methiothepin (Ki = 6.2 nM) > mianserin (Ki = 74.3 nM) > 5-methoxytryptamine (5-MeOT, Ki = 111 nM) > 5-HT (Ki = 150 nM) > ritanserin (Ki = 207 nM) > 5-carboxamidotryptamine (5-CT, Ki = 704 nM). Additional evaluation with four antipsychotics demonstrated strong agreement with previous literature reports. A high specific binding signal and low assay variability, as determined by Z′ = 0.81 ± 0.017, make the SPA format amenable to automation and higher throughput; hence, this assay can be a viable alternative to the more labor-intensive filtration and centrifugation methods.  相似文献

The magnitude by which plasma cortisol levels increase following exposure to a stressor is a heritable trait in rainbow trout. The relative growth in coculture of F1 lines selected for high responsiveness (HR) and low responsiveness (LR) to a confinement stressor suggested that behavioral characteristics related to food acquisition, aggression, or competitive ability might differ between the two lines. This hypothesis was tested using the F2 generation of the selected lines. The F2 lines clearly exhibited the characteristics of the F1 parents, displaying significantly divergent plasma cortisol responses to a 1-h confinement stressor and a high heritability for the trait. Behavioral differences between the lines were assessed by observing the outcome of staged fights for dominance in size-matched pairs of HR and LR fish. The identification of dominant and subordinate fish within each pair on the basis of their behavior was supported by the levels of blood cortisol in the fish attributed to each group (dominant < subordinate). Fish from the LR line were identified as dominant in significantly more trials than were HR individuals. The results suggest that behavioral attributes that affect the outcome of rank-order fights are closely linked to the magnitude of the plasma cortisol response to stress in rainbow trout. Whether the link is causal or circumstantial is not yet evident.  相似文献

A proteinase from Pseudomonas aeruginosa exhibiting collagenolytic activity was purified 1575-fold with a recovery of 24% by use of chemical and chromatographic technics. The enzyme preparation appeared to be homogeneous when subjected to chromatographic, electrophoretic and ultracentrifugational analyses. A standard state sedimentation coefficient of 2.10 S was calculated and further analyses indicated that the enzyme had a molecular weight of 17 500 and dimerizes under certain conditions to yield an apparent molecular weight of 34 000. In addition to insoluble collagen, the enzyme catalyzed the hydrolysis of congocoll, azocoll, soluble collagen and casein, but did not attack orcein-elastin, azoalbumin, p-toluene eulfonyl-L-arginine methyl ester, benzoyl-L-tyrosine ethyl ester, and the hexapeptide N-benzyloxycarbonyl-glycyl-L-prolyglycylglycyl-L-prolyl-L-alanine. Enzymatic activity against congocoll was 6-fold greater at pH 7.5 in Tris with HCl than in phosphate buffer at the same ionic strength. Cobalt, and to a lesser extent, Zn2+ appeared to activate the enzyme, especially in phosphate buffer. NcCN and p-chloromercuribenzoate did not appreciably inhibit enzyme activity, while (NH4)2 SO4, EDTA and cysteine displayed a significant inhibitory effect under certain conditions.  相似文献

Abstract Reference strains of ' Pseudomonas diazotrophicus ' produce a range of polar lipids atypical of authentic Pseudomonas species. In addition to the phospholipids common in Gram-negative bacteria (phosphatidylglycerol, diphosphatidylglycerol, and phosphatidylethanolamine), N -methylated derivatives of phosphatidylethanolamine (including phosphatidylcholine) and an ornithine amide lipid are also present. The preponderant ester-bound fatty acid (up to 80% of the total) is cis -vaccenic acid ( cis -octadec-11-enoic acid), while 3-hydroxyoctadecanoic acid is the major amide-bound fatty acid in the ornithine lipid. Possible implications of the data for classification of the organism are discussed.  相似文献

Plasma cortisol levels were measured as an indicator of physiological stress in roach subjected to brief handling, or to a 14-day period of confinement, and in undisturbed control fish, during winter (water temperature 5° C) and summer (16° C), at which time plasma 17 β-oestradiol levels were also determined. Cortisol levels in undisturbed roach were low (mean 8·1 ng ml−1 at 5° C; 1·4 ng ml−1 at 16° C) and both handling and handling+confinement elevated blood cortisol levels significantly to 400 and 140 ng ml−1, respectively (at 5° C) and 700 and 600 ng ml−1, respectively (at 16) C). Blood cortisol levels had almost returned to baseline within 4 h following handling alone but in fish subjected to handling and prolonged confinement cortisol levels remained elevated for up to 168 h. Differences in baseline and poststress levels of cortisol, and in the rate of recovery from acute stress, were observed at the two different temperatures and the possible factors underlying these differences are discussed. Circulating levels of 17 β-oestradiol were reduced significantly within 24 h of exposure to either acute handling or chronic confinement indicating that the reproductive endocrine system in roach is sensitive to disruption by stressors.  相似文献

Phytoplankton pigment distributions during the spring isothermal periods of 1998 and 1999 and their association with episodic sediment resuspension were characterized in coastal waters of southern Lake Michigan. Total and phylogenetic group chl a concentrations (derived using chemical taxonomy matrix factorization of diagnostic carotenoids) corresponded with assemblage and group biovolumes estimated from microscopic enumeration (P≤ 0.001). Diatoms and cryptophytes dominated assemblages and together typically comprised greater than 85% of relative chl a. Total chl a concentrations and both fucoxanthin·chl a ? 1 and alloxanthin·chl a ? 1 ratios were similar across depths (P> 0.05), indicating uniform distributions of and photophysiological states for assemblages and diatoms and cryptophytes, respectively, throughout the mixed water column. Total chl a concentrations were not always spatially uniform from near‐shore to offshore waters, with the greatest variability reflecting the influence of tributary inflows upon coastal assemblages. Sediment resuspension strongly influenced water column particle density and light climate; however, total and group chl a concentrations did not correspond with coefficients of Kd and suspended particulate matter concentrations (P> 0.05). The correspondence of both light attenuation and suspended particulate matter concentration with relative diatom chl a (P≤ 0.001) indicated an apparent association between sediment resuspension and diatoms. This, and the negative association (P≤ 0.0001) between relative diatom and cryptophyte chl a, corresponded with the spatial dominance of diatom and cryptophyte chl a in near‐shore and offshore waters, respectively. The presence of viable chl a and fucoxanthin within the surficial sediment layer, established this layer as a potential source of meroplanktonic diatoms for near‐shore assemblages.  相似文献

In the Laurentian Great Lakes, phytoplankton growth and biomass are secondarily limited by silica (Si), as a result of phosphorus (P) enrichment. Even modest levels of P enrichment can induce secondary Silimitation, which, in turn, promotes a shift from the native diatom phytoplankton flora to chlorophyte and cyanobacteria species. However, very little is known about the nutritional status of benthic populations and their response to nutrient enrichment. Two experiments were performed in the littoral zone of Lake Michigan where nutrients were delivered to in situ benthic algal (episammic and epilithic) assemblages using nutrient‐diffusing substrata. In order to test the hypothesis that benthic algae in Lake Michigan are Si limited, a 2 × 3 factorial experiment was used to deliver all combinations of Si, N, and P to resident assemblages growing on artificial substrata composed of natural (Si rich) versus calcium carbonate (Si poor) sand. A second experiment utilized a serial enrichment to evaluate the role of Si in mediating changes in taxonomic composition. These findings indicate that benthic algae in Lake Michigan exhibit signs of secondary Si limitation, and that their response to enrichment is similar to the phytoplankton. Moreover, natural sand substrata may provide a source of Si to resident benthic algae.  相似文献
