A stable deletion derivative pNH602 was obtained when the recently described higher-copy-number point mutant pNH601 of plasmid R6K was introduced to a minicells-producing strain of Escherichia coli. The size of plasmid pNH602 is 18.8 Mg/mol as determined by electron microscopy. The 7.2 Mg/mol fragment of R6K genome missing in pNH602 carries the Smr-determinant and the region finO and, according to the results of restriction analysis, it includes one EcoRI site. With its radioisotopically determined 33 copies of pNH602 per E. coli K-12 chromosome (npc), representing a 23% increase of the point mutant pNH601 and 150% enhancement of R6K npc, plasmid pNH602 differs from another closely related R6K deletion derivative pAS3 of the same size which exhibits only 20 npc. Both pNH602 and pAS3 plasmids are conjugative. 相似文献

The impact of elevated [CO2] on seed germination was studied in different genotypes of Arabidopsis thaliana from natural populations. Two generations of seeds were studied: the maternal generation was produced in the greenhouse (present-day conditions), the offspring generation was produced in two chambers where the CO2 concentration was either the present atmospheric concentration (about 350 ppm) or elevated (700 ppm). The seeds were tested for proportion of germinated seeds and mean germination time in both chambers to study the impact of elevated [CO2] during seed production and germination. Elevated [CO2] during maturation of seeds on the mother-plants decreased the proportion of germinated seeds, while elevated [CO2] during germination had no effect on the proportion of germinated seeds. However, when seeds were both produced and germinated under elevated [CO2] (situation expected by the end of next century), germination was slow and low. Moreover, the effect of the [CO2] treatment differs among genotypes of Arabidopsis: there is a strong treatment × genotype interaction. This means that there is ample genetic variance for a selective response modiying the effects of high levels of [CO2] in natural populations of Arabidopsis thaliana. Bacterial ATP binding cassette (ABC) exporters fulfill a wide variety of transmembrane transport roles and are homologous to the human multidrug resistance P-glycoprotein. Recent X-ray structures of the exporters MsbA and Sav1866 have begun to describe the conformational changes that accompany the ABC transport cycle. Here we present cryo-electron microscopy structures of MsbA reconstituted into a lipid bilayer. Using ATPase inhibitors, we captured three nucleotide transition states of the transporter that were subsequently reconstituted into helical arrays. The enzyme–substrate complex (trapped by ADP-aluminum fluoride or AMPPNP) crystallized in a different helical lattice than the enzyme–product complex (trapped by ADP-vanadate). 20 Å resolution maps were calculated for each state and revealed MsbA to be a dimer with a large channel between the membrane spanning domains, similar to the outward facing crystal structures of MsbA and Sav1866. This suggests that while there are likely structural differences between the nucleotide transition states, membrane embedded MsbA remains in an outward facing conformation while nucleotide is bound. 相似文献

Many insect species rely on their sense of audition to find a mate, to localize prey or to escape from a predator. Cicadas are particularly known for their loud call and the conspicuous tympanal hearing system located in their abdomen. The vibration pattern of the tympanal membrane (TM) has been investigated recently revealing mechanical properties specific to species and sex. Although TM size and shape is likely to affect these patterns, the geometry of the cicada ear has never been examined per se. Focusing on three Mediterranean cicada species, namely Cicadatra atra, Cicada orni and Lyristes plebejus, we investigated the structure of TM shape variation at two levels, within and across species. Applying an elliptic Fourier analysis to the outlines of both male and female TMs, we estimated sexual dimorphism and species effects. Cicadatra atra showed a large TM compared with its small size, probably as a result of selective constraints related to the role of the TM in sound production. Sexual dimorphism seemed to be greater than interspecific variation, indicating that constraints operating on sex might be more selective than those acting on species identification. In addition, C. orni appeared to be significantly different from the two other species. This morphological peculiarity could be related to the unique vibrational pattern of its membrane. The effects of aquaculture related stressors on circulating levels of GH, IGF-I and for the first time, IGF-II in Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) were investigated. Specifically, circulating growth factor levels were measured in four different experiments. Two 24 h confinement stressor procedures, (one with Atlantic salmon, the other with rainbow trout); following a hypo-osmotic stressor (freshwater bath) in salt water acclimated, adult, Atlantic salmon; and during a 22 day starvation and re-feeding protocol with juvenile Atlantic salmon. Handling and confinement resulted in significant decreases in circulating levels of all three growth factors in Atlantic salmon, and IGF-I and IGF-II (but not GH) in rainbow trout. A 2-3 h freshwater bath to remove gill parasites on a commercial Atlantic salmon aquaculture operation caused a significant decrease in circulating GH and IGF-I concentrations, but no significant change in IGF-II concentration, 2 days post bathing. Starvation for a period of 15 days in Atlantic salmon resulted in a significant increase in circulating GH levels and a significant decrease in circulating IGF-I and IGF-II. Re-feeding of starved fish for 7 days resulted in a significant decrease in GH to the concentration measured in continually fed fish, however re-feeding did not change plasma levels of IGF-I and IGF-II relative to continually starved fish. The results presented here confirm previously observed handling and confinement stressor induced effects on GH and IGF-I and, for the first time, on IGF-II in salmonids. Furthermore this study confirms the nutritional regulation of GH, IGF-I and IGF-II in juvenile Atlantic salmon. 相似文献
