Members of the marine bacterial genus Pseudoalteromonas have been found in association with living surfaces and are suggested to produce bioactive compounds against settlement of algal spores, invertebrate larvae, bacteria and fungi. To determine the extent by which these antifouling activities and the production of bioactive compounds are distributed amongst the members of the genus Pseudoalteromonas, 10 different Pseudoalteromonas species mostly derived from different host organisms were tested in a broad range of biofouling bioassays. These assays included the settlement of larvae of two ubiquitous invertebrates Hydroides elegans and Balanus amphitrite as well as the settlement of spores of the common fouling algae Ulva lactuca and Polysiphonia sp. The growth of bacteria and fungi, which are the initial fouling organisms on marine surfaces, was also assayed in the presence of each of the 10 Pseudoalteromonas species. It was found that most members of this genus produced a variety of bioactive compounds. The broadest range of inhibitory activities was expressed by Pseudoalteromonas tunicata which inhibited all target fouling organisms. Only two species, Pseudoalteromonas haloplanktis and Pseudoalteromonas nigrifaciens, displayed negligible activity in the bioassays. These were also the only two non-pigmented species tested in this study which indicates a correlation between production of bioactive compounds and expression of pigment. Three members, P. tunicata, Pseudoalteromonas citrea and Pseudoalteromonas rubra, were demonstrated to express autoinhibitory activity. It is suggested that most Pseudoalteromonas species are efficient producers of antifouling agents and that the production of inhibitory compounds by surface associated Pseudoalteromonas species may aid the host against colonisation of its surface.  相似文献

The binding of a ligand molecule to a protein is often accompanied by conformational changes of the protein. A central question is whether the ligand induces the conformational change (induced-fit), or rather selects and stabilizes a complementary conformation from a pre-existing equilibrium of ground and excited states of the protein (selected-fit). We consider here the binding kinetics in a simple four-state model of ligand-protein binding. In this model, the protein has two conformations, which can both bind the ligand. The first conformation is the ground state of the protein when the ligand is off, and the second conformation is the ground state when the ligand is bound. The induced-fit mechanism corresponds to ligand binding in the unbound ground state, and the selected-fit mechanism to ligand binding in the excited state. We find a simple, characteristic difference between the on- and off-rates in the two mechanisms if the conformational relaxation into the ground states is fast. In the case of selected-fit binding, the on-rate depends on the conformational equilibrium constant, whereas the off-rate is independent. In the case of induced-fit binding, in contrast, the off-rate depends on the conformational equilibrium, while the on-rate is independent. Whether a protein binds a ligand via selected-fit or induced-fit thus may be revealed by mutations far from the protein's binding pocket, or other "perturbations" that only affect the conformational equilibrium. In the case of selected-fit, such mutations will only change the on-rate, and in the case of induced-fit, only the off-rate.  相似文献

Bacterial virulence and antibiotic resistance have a significant influence on disease severity and treatment options during bacterial infections. Frequently, the underlying genetic determinants are encoded on mobile genetic elements (MGEs). In the leading human pathogen Staphylococcus aureus, MGEs that contain antibiotic resistance genes commonly do not contain genes for virulence determinants. The phenol-soluble modulins (PSMs) are staphylococcal cytolytic toxins with a crucial role in immune evasion. While all known PSMs are core genome-encoded, we here describe a previously unidentified psm gene, psm-mec, within the staphylococcal methicillin resistance-encoding MGE SCCmec. PSM-mec was strongly expressed in many strains and showed the physico-chemical, pro-inflammatory, and cytolytic characteristics typical of PSMs. Notably, in an S. aureus strain with low production of core genome-encoded PSMs, expression of PSM-mec had a significant impact on immune evasion and disease. In addition to providing high-level resistance to methicillin, acquisition of SCCmec elements encoding PSM-mec by horizontal gene transfer may therefore contribute to staphylococcal virulence by substituting for the lack of expression of core genome-encoded PSMs. Thus, our study reveals a previously unknown role of methicillin resistance clusters in staphylococcal pathogenesis and shows that important virulence and antibiotic resistance determinants may be combined in staphylococcal MGEs.  相似文献

Two methods were used to separate free-ranging mink Mustela vison into wild mink and escaped farm mink. Analysis of stable carbon isotopes was performed on teeth and claws of 226 free-ranging mink from two areas in Denmark. A classification based on empirical data resulted in three groups (n=213); 47% were newly escaped farm mink and another 31% had been born in farms and lived in nature for more than ca. 2 months. The remaining 21% may or may not have been born in nature, but they had been free ranging for more than a year and were thus considered wild. A genetic analysis by means of microsatellites was performed on a subsample of the trapped mink (86 individuals) and on 70 farm mink, in order to assess the proportion of escaped farm mink in the free-ranging population. Strong genetic evidence for a high percentage of escaped farm mink in the free-ranging population (86%) was found, in agreement with the carbon isotope results. Both methods can be used to distinguish between farm and wild mink, but whereas microsatellites can only say whether a given mink originated from a farm or not, carbon isotopes can give some more detail on the period of time that a farm mink has been living in natural habitats. The high proportion of escaped farm mink in the Danish nature could have serious implications for the preservation of other vulnerable species and should be carefully considered when designing conservation strategies.  相似文献

The dominant Arctic Ocean and North Atlantic copepods Calanus hyperboreus, Calanus glacialis, and Calanus finmarchicus were collected in the Greenland Sea and fed 13C labelled diatom Thalassiosira weissflogii to follow the transfer and assimilation of carbon, lipid, and individual fatty acids and alcohols. The diatom was grown with 13C for 3 to 5 days and fed then to the copepods. During the feeding period of 14 days, total carbon increased in the copepodite stages V of C. hyperboreus and C. finmarchicus, whereas carbon remained almost constant in C. glacialis females. However, total lipid increased in all species and stages. Highest lipid accumulation occurred in C. hyperboreus in which nearly all lipids were exchanged already after 11 days of feeding. In the other species lipid accumulation made up between 22% (C. finmarchicus) and 45% of total lipid (C. glacialis). The proportion of wax esters was high ranging from 76% of total lipid in C. glacialis to 92% in C. finmarchicus. The fatty acid composition of the alga was dominated by 16:1(n-7), 16:0, 20:5(n-3), and 22:6(n-3). The composition of the copepods was similar because of feeding already on diatoms in the field. In addition, the monounsaturated fatty acids and alcohols, 20:1(n-9) and 22:1(n-11), were major components of the copepod lipids. During the feeding period the highest 13C labelling was always found in the C16 polyunsaturated fatty acids and in the 16:1(n-7) alcohol. Because these components occurred only in trace amounts in the copepods they totally originated from the diet explaining the high labelling. It is noteworthy that the 16:1(n-7) alcohol originated only from the corresponding dietary and not from the abundant internal fatty acid. The long-chain monounsaturated fatty acids and alcohols, 20:1(n-9) and 22:1(n-11), are not existent in phytoplankton and have to be produced de novo. They were less labelled in the smaller species but highly 13C enriched in C. hyperboreus. Although dietary fatty acids were generally retained by the copepods it seems that fatty acids or even lipids were selectively accumulated and turned over due to bodily requirements, and thus, essential polyunsaturated fatty acids were preferentially retained. During feeding mixing, accumulation, and exchange of internal and dietary fatty acids and alcohols occurred as well as utilisation of lipids from both sources for metabolic requirements. The differences in lipid assimilation fit to the different life strategies of the copepods.  相似文献

Capparis dongvanensis (Capparaceae) is described as a new species and illustrated from Ha Giang Province of Vietnam. It is morphologically similar to C. lanceolaris and C. fengii, but differs in the number of flowers on peduncles, length of spine and petiole, apex of the leaf, type of inflorescence, petal color, cylindrical ovary without any beak, fruit shape and number of seeds per fruit. Diagnostic morphological characters along with pollen and seed SEM images of C. dongvanensis and allied species are provided. Its ecology, habitat and conservation status are also described.  相似文献

European agricultural policy increasingly focuses on environmental friendly cropping systems. Intercropping of maize (Zea mays L.) and common beans (Phaseolus vulgaris L.) has been suggested as an alternative cropping system with environmental benefits. The aim of this study was to assess methane yield potential of mixed silages. Based on material from two field experiments at three sites in Germany, mixed silages were produced with proportions of individual components varying from 0 to 100 % of fresh matter in increments of 12.5 %. Chemical parameters (neutral detergent fiber (NDF), acid detergent fiber (ADF), crude protein (XP), starch, sugar, and crude fat) were determined, and batch tests were performed to measure methane yield potential from silages. With increasing bean proportion, concentrations of XP increased while NDF, methane yield, and methane content decreased. While methane yield showed a negative relationship with XP content (R 2 = 0.56***), a positive relation was found with NDF (R 2 = 0.55***). The reduction of methane yield of circa 1 L of methane per each additional bean percentage in the silages could not be explained by the chemical parameters of the silages. It is hypothesized that other chemical compounds, such as lectins, which were not determined in the present study, may have influenced methane production.  相似文献

Cyst and root‐knot nematodes are obligate parasites of economic importance with a remarkable ability to reprogram root cells into unique metabolically active feeding sites. Previous studies have suggested a role for cytokinin in feeding site formation induced by these two types of nematodes, but the mechanistic details have not yet been described. Using Arabidopsis as a host plant species, we conducted a comparative analysis of cytokinin genes in response to the beet cyst nematode (BCN), Heterodera schachtii, and the root‐knot nematode (RKN), Meloidogyne incognita. We identified distinct differences in the expression of cytokinin biosynthesis, catabolism and signaling genes in response to infection by BCN and RKN, suggesting differential manipulation of the cytokinin pathway by these two nematode species. Furthermore, we evaluated Arabidopsis histidine kinase receptor mutant lines ahk2/3, ahk2/4 and ahk3/4 in response to RKN infection. Similar to our previous studies with BCN, these lines were significantly less susceptible to RKN without compromising nematode penetration, suggesting a requirement of cytokinin signaling in RKN feeding site formation. Moreover, an analysis of ahk double mutants using CycB1;1:GUS/ahk introgressed lines revealed contrasting differences in the cytokinin receptors mediating cell cycle activation in feeding sites induced by BCN and RKN.  相似文献
