The four sperm-specific CatSper ion channel proteins are required for hyperactivated motility and male fertility, and for Ca2+ entry evoked by alkaline depolarization. In the absence of external Ca2+, Na+ carries current through CatSper channels in voltage-clamped sperm. Here we show that CatSper channel activity can be monitored optically with the [Na+]i-reporting probe SBFI in populations of intact sperm. Removal of external Ca2+ increases SBFI signals in wild-type but not CatSper2-null sperm. The rate of the indicated rise of [Na+]i is greater for sperm alkalinized with NH4Cl than for sperm acidified with propionic acid, reflecting the alkaline-promoted signature property of CatSper currents. In contrast, the [Na+]i rise is slowed by candidate CatSper blocker HC-056456 (IC50 ∼3 µM). HC-056456 similarly slows the rise of [Ca2+]i that is evoked by alkaline depolarization and reported by fura-2. HC-056456 also selectively and reversibly decreased CatSper currents recorded from patch-clamped sperm. HC-056456 does not prevent activation of motility by HCO3− but does prevent the development of hyperactivated motility by capacitating incubations, thus producing a phenocopy of the CatSper-null sperm. When applied to hyperactivated sperm, HC-056456 causes a rapid, reversible loss of flagellar waveform asymmetry, similar to the loss that occurs when Ca2+ entry through the CatSper channel is terminated by removal of external Ca2+. Thus, open CatSper channels and entry of external Ca2+ through them sustains hyperactivated motility. These results indicate that pharmacological targeting of the CatSper channel may impose a selective late-stage block to fertility, and that high-throughput screening with an optical reporter of CatSper channel activity may identify additional selective blockers with potential for male-directed contraception. 相似文献

A longitudinal study was conducted to investigate the nature of Escherichia coli O157:H7 colonization of feedlot cattle over the final 100 to 110 days of finishing. Rectal fecal grab samples were collected from an initial sample population of 788 steers every 20 to 22 days and microbiologically analyzed to detect E. coli O157:H7. The identities of presumptive colonies were confirmed using a multiplex PCR assay that screened for gene fragments unique to E. coli O157:H7 (rfbE and fliCh7) and other key virulence genes (eae, stx1, and stx2). Animals were classified as having persistent shedding (PS), transient shedding (TS), or nonshedding (NS) status if they consecutively shed the same E. coli O157:H7 genotype (based on the multiplex PCR profile), exhibited variable E. coli O157 shedding, or never shed morphologically typical E. coli O157, respectively. Overall, 1.0% and 1.4% of steers were classified as PS and NS animals, respectively. Characterization of 132 E. coli O157:H7 isolates from PS and TS animals by pulsed-field gel electrophoresis (PFGE) typing yielded 32 unique PFGE types. One predominant PFGE type accounted for 53% of all isolates characterized and persisted in cattle throughout the study. Isolates belonging to this predominant and persistent PFGE type demonstrated an enhanced (P < 0.0001) ability to adhere to Caco-2 human intestinal epithelial cells compared to isolates belonging to less common PFGE types but exhibited equal virulence expression. Interestingly, the attachment efficacy decreased as the genetic divergence from the predominant and persistent subtype increased. Our data support the hypothesis that certain E. coli O157:H7 strains persist in feedlot cattle, which may be partially explained by an enhanced ability to colonize the intestinal epithelium.Escherichia coli serotype O157:H7 was first linked to human illness in the early 1980s, when it was determined to cause severe abdominal pain with initially watery diarrhea that progressed to grossly bloody diarrhea accompanied by little or no fever (42). Initially, E. coli O157:H7 can cause nonbloody diarrhea through attachment to, and subsequent destruction of, intestinal microvilli (24). In addition to microvillus damage, serious health complications can arise due to the ability of E. coli O157:H7 to produce Shiga toxins (Stx1 and Stx2). Shiga toxins are very potent cytotoxins that are absorbed into the intestinal microvasculature and initiate apoptosis of vascular epithelium, resulting in hemorrhagic colitis (41). Persistent uptake of these toxins may lead to more severe manifestations of disease, such as hemolytic-uremic syndrome, which may ultimately result in kidney failure (24). Most recent estimates have identified E. coli O157:H7 as the cause of at least 70,000 cases of food-borne illness annually in the United States, and in 4% of cases life-threatening hemolytic-uremic syndrome develops (37). Epidemiological studies have implicated the consumption of meat, dairy products, produce, and water contaminated by animal feces, as well as person-to-person contact and direct contact with farm animals or their environment, as routes of E. coli O157:H7 transmission leading to human illness (36).It is generally accepted that cattle and other animals are the major reservoir of E. coli O157:H7, but it is still not clear if animals are colonized for prolonged periods with E. coli O157:H7 or if they transiently shed this organism following repeated exposure to it through ingestion of contaminated feedstuffs or water or through exposure to other contaminated environmental sources. Based on results of numerous epidemiological studies (4, 6, 21, 30, 32), the prevalence of E. coli O157:H7 in feedlot cattle is highly variable and can range from less than 1% to 80%. Several other studies (7, 8, 23) have found evidence of persistent E. coli O157:H7 colonization in individual cattle, supporting the hypothesis that at least some animals are susceptible to persistent E. coli O157:H7 colonization. Multiple experimental inoculation studies (15, 23, 39, 46) showed that E. coli O157:H7 persists in the bovine gastrointestinal (GI) tract for at least 14 days up to 140 days postinfection. Studies have implicated the lower GI tract and specifically the recto-anal junction (RAJ) as the major location of E. coli O157:H7 colonization and proliferation (9, 12, 23, 39); however, this organism also can be found throughout the bovine GI tract (7, 8, 31, 40, 54).It stands to reason that if the E. coli O157:H7 prevalence in cattle presented for harvest were reduced, there would be a decrease in the probability of beef product contamination, if good manufacturing procedures were used. Although there is consensus concerning the importance of preharvest pathogen mitigation and its role in minimizing entry of E. coli O157:H7 into harvest facilities, there is disagreement about the significance of “supershedders” (animals that excrete large quantities of a pathogen for various amounts of time) for E. coli O157:H7 transmission dynamics at the preharvest level (12, 34, 35, 39). Utilizing statistical modeling, researchers have estimated that, on average, the prevalence of “supershedders” in a population is 4% and that these animals excrete 50 times more E. coli O157:H7 than other animals colonized by this organism (34). Additionally, the same researchers suggested that approximately 80% of E. coli O157:H7 transmission is generated by a few “supershedders” (35).Research by our group discovered a unique association between E. coli O157:H7 prevalence in pen floor fecal pats and carcass contamination by this pathogen (57). When the prevalence in fecal pats from a pen floor exceeded 20%, carcasses of animals from the pen had E. coli O157:H7 prevalence values of 14.3, 2.9, and 0.7% before evisceration, after evisceration, and after final intervention, respectively. However, when the prevalence in pen floor fecal pats was less than 20%, the preeviscerated carcass prevalence value was 6.3%, and there was no detectable E. coli O157:H7 contamination of carcass samples after evisceration and after final intervention (57). Thus, we hypothesize that animals which persistently excrete normal levels of E. coli O157:H7 over prolonged periods (persistent shedders [PS]) rather than animals that periodically shed abnormally high levels (supershedders) are the most significant source of E. coli O157:H7 contamination in the food continuum. Although previous studies suggested that cattle may be persistently colonized by E. coli O157:H7 and shed this organism in their feces for prolonged periods, molecular subtyping data are required to further investigate whether cattle are persistently colonized by the same strain (i.e., molecular subtype) or if they are repeatedly exposed to different strains through contaminated feedstuffs, water, or other environmental sources. Thus, the objectives of this study were to determine if naturally colonized feedlot cattle persistently shed E. coli O157:H7, using combined cultural microbiological analyses, molecular subtyping approaches, and in vitro virulence phenotype assays to probe the factors (agent, host, environment, or a combination of these factors) that contribute to the complex ecology of E. coli O157:H7 persistence at the preharvest level. 相似文献

Many insects harbor specific bacteria in their digestive tract, and these gut microbiota often play important roles in digestion and nutrient provisioning. While it is common for a given insect species to harbor a representative gut microbial community as a population, how this community is acquired and maintained from generation to generation is not known for most xylophagous insects, except termites. In this study, we examined acquisition of gut microbiota by the wood-feeding beetle, Anoplophora glabripennis, by identifying and comparing microbial community members among different life stages of the insect and with microbes it encounters in the environment. Automated ribosomal intergenic spacer analysis was employed to compare bacterial communities present in the egg and larval stages of A. glabripennis as well as with microbes found in the oviposition site and the surrounding woody tissue. Multivariate analyses were used to identify relationships between sample type and specific bacterial types (operational taxonomic units). From this analysis, bacteria that were derived from the environment, the oviposition site, and/or the egg were identified and compared with taxa found in larvae. Results showed that while some larval microbes were derived from environmental sources, other members of the larval microbial community appear to be vertically transmitted. These findings could lead to a better understanding of which microbial species are critical for the survival of this insect and to development of techniques that could be used to alter this community to disrupt the digestive physiology of the host insect as a biological control measure. 相似文献

The basic, still unanswered question about visual object representation is this: what specific information is encoded by neural signals? Theorists have long predicted that neurons would encode medial axis or skeletal object shape, yet recent studies reveal instead neural coding of boundary or surface shape. Here, we addressed this theoretical/experimental disconnect, using adaptive shape sampling to demonstrate explicit coding of medial axis shape in high-level object cortex (macaque monkey inferotemporal cortex or IT). Our metric shape analyses revealed a coding continuum, along which most neurons represent a configuration of both medial axis and surface components. Thus, IT response functions embody a rich basis set for simultaneously representing skeletal and external shape of complex objects. This would be especially useful for representing biological shapes, which are often characterized by both complex, articulated skeletal structure and specific surface features. 相似文献

germline polymorphisms in the variable (V) gene segments of the human TCRA/D locus. Thirty different V gene segments were amplified from six to eight unrelated individuals and sequenced from low melting point agarose. Twenty-seven
polymorphisms were identified in 15 V gene segments. These polymorphisms are mainly single nucleotide substitutions, but an insertion/deletion polymorphism and
a single dinucleotide repeat with variable length were also seen. Of the 15 sequence variations found in the coding regions,
six are silent and nine encode amino acid changes. All of the amino acid changes are found at non-conserved residues, frequently
in the hypervariable regions, where they may influence MHC and/or peptide recognition. Therefore, it is possible that germline
variations in TCR genes could influence an individual’s immune response, and may also contribute to susceptibility to diseases such as autoimmunity.

Species are the most commonly recognised unit for conservation management, yet significant variation can exist below the level of taxonomic recognition and there is a lack of consensus around how a species might be defined. This definition has particular relevance when species designations are used to apportion conservation effort and when definitions might be made through legislation. Here, we use microsatellite DNA analyses to test the proposition that the last remaining populations of the endangered grassland earless dragon (Tympanocryptis pinguicolla) harbour substantial cryptic genetic variation. Our study provides strong evidence that long historical isolation and the recent impacts of urbanization, have led to genetic differentiation in microsatellite DNA allele frequencies and high numbers of private alleles among three genetic clusters. This differentiation is partially concordant with previous mitochondrial DNA analyses, which show the two regions (Canberra and Monaro) where this species exists, to be reciprocally monophyletic, but differs through the identification of a third genetic cluster that splits a northern Canberra cluster from that of southern Canberra. Our data also identify a stark contrast in population genetic structure between clusters such that high levels of genetic structure are evident in the highly urbanised Canberra region but not in the largely rural Monaro region. We conclude that this species, like many reptiles, harbours considerable cryptic variation and currently comprises three distinct and discrete units. These units could be classified as separate species for the purpose of conservation under the relevant Australian and international Acts drawing management appropriate to that status. 相似文献

Phosphorylase kinase (PhK) is a hexadecameric (αβγδ)4 enzyme complex that upon activation by phosphorylation stimulates glycogenolysis. Due to its large size (1.3 MDa), elucidating the structural changes associated with the activation of PhK has been challenging, although phosphoactivation has been linked with an increased tendency of the enzyme's regulatory β‐subunits to self‐associate. Here we report the effect of a peptide mimetic of the phosphoryltable N‐termini of β on the selective, zero‐length, oxidative crosslinking of these regulatory subunits to form β–β dimers in the nonactivated PhK complex. This peptide stimulated β–β dimer formation when not phosphorylated, but was considerably less effective in its phosphorylated form. Because this peptide mimetic of β competes with its counterpart region in the nonactivated enzyme complex in binding to the catalytic γ‐subunit, we were able to formulate a structural model for the phosphoactivation of PhK. In this model, the nonactivated state of PhK is maintained by the interaction between the nonphosphorylated N‐termini of β and the regulatory C‐terminal domains of the γ‐subunits; phosphorylation of β weakens this interaction, leading to activation of the γ‐subunits. 相似文献
