The basic properties of bacteriophage N4 development have been investigated in Escherichia coli Hfr 3300 under one-step growth and high cell density conditions. N4r(+) -infected bacteria are lysis inhibited in mass culture, burst asynchronously starting 180 min postinfection, and release over 3,000 phage per cell. During lysis inhibition the bacteria continuously elongate, increase in girth, and undergo characteristic morphological changes represented by the appearance of dark spots located at the cell poles. In thin sections, during the late stages of replication and assembly, the phage particles are localized exclusively in restricted areas of the cytoplasm near the polar regions. Large paracrystalline arrays of virions are found in over 7% of the cells before lysis. The most common mechanism of lysis consists in the formation of bulges located at random in the cell circumference; these burst and, without extensive disruption of the cell wall, the phage progeny escapes into the medium.  相似文献

The predictability of temporal changes was analysed in the population density of three meiobenthic copepod species, and for three parameters characterizing this community. For Paronychocamptus nanus and Tachidius discipes a large part of the variation in time scales longer than 1 year is explainable as cyclic factors. In P. nanus clear cyles with periods of 2 years and 1.5 years were found. These cyclic factors can be extrapolated for prediction. In Canuella perplexa almost all the variation was due to unpredictable long-term drift. Community parameters are generally better predicted than population densities. In our data set total density of the copepods was the best parameter to monitor. Monitoring does not necessitate a strict sampling scheme; irregular quarterly sampling from the P. nanus series showed that the essential features of the series were still discernible.  相似文献

We have carried out a systematic analysis of the solid-state conformational preferences of a number of linear homo-oligoprolines (to the tetramer) by ir absorption and x-ray diffraction. The peptides present different chiral sequences (tacticities), various types (urethane and amide) of N-protecting groups, and free and blocked C-termini (which imply different capabilities of forming H-bonds). The following conclusions can be drawn: (i) values for the geometry of the prolyl residue and the peptide bond in the cis and in the trans conformations are proposed; (ii) in general the conformational angles φ and ψ in the linear homo-oligoprolines have values appropriate for the polyproline II structure (conformation F); (iii) the pyrrolidine ring shows various types of puckering with no apparent relation to the backbone conformation; (iv) Pro-Pro peptide bonds generally take the trans conformation, the few cases of cis conformation being formed by Pro residues of different chirality; (v) the single H-bond donor — OH, when present, is always bonded to H-acceptors, which can be either the urethane or the amide or the peptide carbonyl but never the carbonyl group of the — COOH moiety.  相似文献

A coordinated activation of upper airway and chest wall muscles may be crucial in maintaining airway patency and ventilation. The alae nasi (AN) and diaphragm (DIA) electromyograms (EMG) were recorded with surface electrodes in 17 unsedated healthy preterm infants during both active (AS) and quiet sleep (QS). Airflow was measured via a nasal mask pneumotachograph and integrated to obtain tidal volume. Studies were performed during inhalation of room air and mixtures of 2 and 4% CO2 in air. In room air, phasic AN EMG accompanied 45 +/- 7% of breaths during AS compared with 14 +/- 5% of breaths during QS (P less than 0.001); however, with inhalation of 4% CO2 the incidence of AN EMG increased to comparable levels in both sleep states. During room air breathing onset of AN EMG preceded that of the DIA EMG and inspiratory airflow by 41 +/- 8 ms (P less than 0.01) and 114 +/- 29 ms (P less than 0.05), respectively. Peak AN activity preceded peak DIA activity by 191 +/- 36 ms (P less than 0.01). Alteration in sleep state or increasing chemical drive did not significantly alter these temporal relationships. Nevertheless, with each increase in end-tidal CO2, peak DIA EMG and tidal volume increased while peak AN EMG only showed a consistent increase during 4% CO2 inhalation. We conclude that although there exists a mechanism that temporally coordinates AN and DIA activation, the amount of AN EMG activity with each breath is not clearly correlated with DIA activation, which may contribute to the high incidence of respiratory dysrhythmias in preterm neonates.  相似文献

Summary The cholinergic innervation of the human liver was studied. Slices (150–200m thick) of human liver and of the greater hepatic blood vessels (hepatic artery and vein, portal vein) were incubated in a solution of 6-hydroxydopamine (6-HDA) in order to obtain a selective degeneration of adrenergic nerves. Controls were prepared from samples incubated with buffer alone. The slices were cut on a cryostat into 15–20m thick sections and processed for the histochemical detection of cholinesterases.Cholinergic nerve fibres innervate the extra hepatic and the intrahepatic branches of the hepatic artery, the portal vein as well as the hepatic vein. Fewer cholinergic fibres innervate the hepatocytes and the hepatic sinusoids. The 6-HDA treatment does not seem to alter the pattern of the cholinergic innervation of the liver. The findings indicate the presence of a cholinergic parasympathetic innervation in the human liver.  相似文献

Abstract: The uptake of amino acids by microvessels isolated from brains of rats was studied. Previous studies have demonstrated alterations in blood-brain amino acid transport after portacaval shunt in rats. In order to elucidate whether such changes in the blood-brain barrier were located in the microvessels, brain microvessels were isolated from both rats with portacaval shunt and controls. Brain microvessels from rats 2 weeks after shunt operations took up significantly greater amounts of 14C-labeled neutral amino acids, but not of glutamic acid. lysine, or α-methylaminoisobutyric acid than microvessels from sham-operated controls. Measurement of uptake kinetics showed a higher V max for phenylalanine and leucine uptake and a lower V max for lysine uptake in microvessels from shunted rats compared with control, whereas the respective K m's of uptake were similar in both preparations. The results suggest that changes in brain microvessel transport activity account for altered brain neutral amino acid concentrations after portacaval shunt and that such changes can be studied in vitro in isolated microvessels.  相似文献

From Verbascum sinuatum, besides aucubin, harpagide, 6-O-β-d-xylopyranosylaucubin and sinuatol (6-O- α-l-rhamnopyranosylaucubin), a new iridoid glycoside, sinuatoside, has been isolated and its structure elucidated as 6-O-(3-O-β-d-xylopyranosyl)α-d-galactopyranosyl aucubin on the basis of spectral data and chemical modifications.
