A total of 41 faeces and 5 vomits of the Weddell seal Leptonychotes weddelli was collected at Harmony Point, Nelson Island, South Shetland Islands, from 14 January to 1 February 1996. The diet indicated by the remains in the samples was diverse and comprised both pelagic and benthic-demersal species. Fish were the most frequent (95.7%) and numerous prey (46.2%), but molluscs were the most important by mass (65.8%). Octopods, mainly Pareledone charcoti, constituted the bulk of the diet (63.1% by mass), but the importance of the remaining molluscs was negligible. Otoliths represented 510 fish of which 491 were identified as belonging to 5 species: Gymnoscopelus nicholsi, Electrona antarctica, Lepidonotothen nudifrons, Gobionotothen gibberifrons and Nototheniops nybelini. The myctophid Gymnoscopelus nicholsi was the most important fish prey, and the contribution of benthic-demersal species was low. However, the importance of that fish was over-estimated since 96% of the specimens were obtained from the five vomits analysed. The biases associated with the faecal analysis technique are discussed. Site-directed mutations in the Escherichia coli ssb gene were tested for the ability to complement a chromosomal ssb deletion for viability, and only the ssb W54→G mutation failed to do so at the pSC101 copy level. Non-aromatic amino acid substitutions for SSB Trp-54 ( ssb W54→L and ssb W54→S) produced the greatest effects on in vivo protein function including altered marker linkage subsequent to generalized transduction, extreme UV sensitivity, and a lack of ability to support SOS induction. Additionally, the ssb-113 ( ssb P176→S) mutation demonstrated the existence of both uvrA -dependent and uvrA -independent components of SOS induction. Although nucleotide excision repair appeared unaffected by alterations in the SSB protein, the mutational analysis suggests a direct role for SSB in recombinational repair.  相似文献

Serum free fermentation procedures of cell cultures have got a wide application in production of biochemicals. But, cells cultured in serum free media in general are more sensitive to changes in culture condition, especially to nutrient limitation. There are no substances from serum which can support the cells when conditions are changing. In this study special attention is directed to amino acid utilization of mouse hybridoma in batch, chemostat and perfusion fermentations. Detailed data are presented which show the considerable difference of amino acid consumption rates in different fermentation modes. Already, in batch mode there are differences of the two investigated mouse hybridoma cell lines, although they are derived from the same myeloma line. In chemostat running at a dilution rate representing maximal growth rate most of the consumption rates are significant higher than in batch. On the other hand, in perfusion mode the rates are lower than in batch. This indicates clearly the different conditions of the fermentation modes. Therefore, it is necessary to develop serum free processes under the desired production conditions. An accurate analysis of the process is strongly recommended.  相似文献

Berkowski, B & Klug, C. 2011: Lucky rugose corals on crinoid stems: unusual examples of subepidermal epizoans from the Devonian of Morocco. Lethaia, Vol. 45, pp. 24–33. In the fossil record, evidence for true epizoans, i.e. living animals inhabiting other living host‐animals, is rather rare. A host reaction is usually needed to proof the syn vivo‐settling of the epizoan. Herein, we provide a first report of such an epizoan biocoenosis from various strata of the Early Devonian of Hamar Laghdad, the world‐renowned Moroccan mud‐mound locality. In this case, solitary rugose corals settled as larvae on crinoid stems, perhaps at a spot where the epidermis was missing for some reason (injury, disease). Both the crinoid and the coral began to grow around each other. By doing so, the affected crinoid columnals formed a swelling, where ultimately only an opening slightly larger than the coral orifice remained. We discuss both macroecological and small‐scale synecological aspects of this biocoenosis. The coral profited from its elevated home because it reached into more rapid currents providing the polyp with more food than at the densely populated seafloor, which was probably covered by a coral‐meadow around the mounds and hydrothermal vents. □Corals, crinoids, Early Devonian, epizoans, Morocco, Rugosa.  相似文献

Background  

Parkinson's disease (PD) is the second most common neurodegenerative disorder. One of the most widely used techniques to diagnose PD is a Single Photon Emission Computer Tomography (SPECT) scan to visualise the integrity of the dopaminergic pathways in the brain. Despite this there remains some discussion on the value of SPECT in the differential diagnosis of PD. We did a meta-analysis of all the existing literature on the diagnostic accuracy of both pre- and post-synaptic SPECT imaging in the differential diagnosis of PD.  相似文献

The soybean ubiquitous urease (encoded by GmEu4) is responsible for recycling metabolically derived urea. Additional biological roles have been demonstrated for plant ureases, notably in toxicity to other organisms. However, urease enzymatic activity is not related to its toxicity. The role of GmEu4 in soybean susceptibility to fungi was investigated in this study. A differential expression pattern of GmEu4 was observed in susceptible and resistant genotypes of soybeans over the course of a Phakopsora pachyrhizi infection, especially 24 h after infection. Twenty-nine adult, transgenic soybean plants, representing six independently transformed lines, were obtained. Although the initial aim of this study was to overexpress GmEu4, the transgenic plants exhibited GmEu4 co-suppression and decreased ureolytic activity. The growth of Rhizoctonia solani, Phomopsis sp., and Penicillium herguei in media containing a crude protein extract from either transgenic or non-transgenic leaves was evaluated. The fungal growth was higher in the protein extracts from transgenic urease-deprived plants than in extracts from non-transgenic controls. When infected by P. pachyrhizi uredospores, detached leaves of urease-deprived plants developed a significantly higher number of lesions, pustules and erupted pustules than leaves of non-transgenic plants containing normal levels of the enzyme. The results of the present work show that the soybean plants were more susceptible to fungi in the absence of urease. It was not possible to overexpress active GmEu4. For future work, overexpression of urease fungitoxic peptides could be attempted as an alternative approach.  相似文献

Cephalopods play a key role in marine environments as food resources for top predators such as marine mammals and seabirds. However, detailed information on their trophic relationships with Antarctic seals is scarce. The aim of the present study was to examine the cephalopod portion of the diet of adult and subadult Weddell seals, Leptonychotes weddellii, at Hope Bay, Antarctic Peninsula, through the analysis of scats collected during three consecutive summers (2003, 2004 and 2005). Cephalopods occurred in almost 45% of the 217 samples collected during the whole period of study. A total of 662 beaks (358 upper and 304 lower) were removed from scats containing cephalopod remains (n = 93). Octopods were largely dominant in comparison with teuthoids constituting in numerical abundance over 95% of the cephalopod prey. The octopod Pareledone turqueti was the most frequent and dominant prey species representing, respectively, 57.9 and 71.1% in numbers and biomass of cephalopods consumed. Species belonging to the group of papillated Pareledone were second in importance. The results were compared with information from previous dietary studies of L. weddellii at other localities of Antarctica. Based on the examination of the cephalopod prey taxa identified in this study, it is suggested that during the study period Weddell seal individuals foraged mainly on benthic prey resources close to the coast, in inshore waters where octopods were dominant in comparison with pelagic squid.  相似文献

Caves provide excellent settings to examine evolutionary questions. Subterranean environments are characterized by similar and consistent conditions. Cave-adapted species often share characteristics such as diminished pigmentation, elongated limbs and reduced or absent eyes. Relatively little is known about the evolution and development of troglomorphic traits in invertebrates. In this study, we compare expression of the eye development genes hedgehog, pax6, sine oculis and dachshund in individuals from multiple independently derived cave populations of the amphipod Gammarus minus. hedgehog expression was significantly reduced in cave populations, compared to genetically related surface populations. Interestingly, no differences were found in pax6, sine oculis or dachshund expression. Because hedgehog-related genes are also involved in eye reduced in Astyanax mexicanus, these genes may be consistent targets of evolution during cave adaptation. These results provide support for the hypothesis of genomic 'hotspots' of evolution and allow comparison of adaptive mechanisms among diverse animals in subterranean environments.  相似文献
