We have identified the major antigens or IgE binding components from wheat flour. Thirty-five sera from patients with baker's asthma were used to analyze the reaction with wheat salt-soluble proteins. We found a 15 kDa SDS-PAGE band which reacted with all sera tested. Purified members of the alpha-amylase inhibitor family, which are the main components of the 15 kDa band, were recognized by specific IgE when tested with a pool of reactive sera. Immunodetection after two-dimensional electrophoretic fractionation of crude inhibitor preparations from wheat endosperms also detected several inhibitor subunits as major low-molecular-weight allergens.  相似文献

The Shaker complex (ShC) spans over 350 kb in the 16F region of the X chromosome. It can be dissected by means of aneuploids into three main sections: the maternal effect (ME), the viable (V) and the haplolethal (HL) regions. The mutational analysis of ShC shows a high density of antimorphic mutations among 12 lethal complementation groups in addition to 14 viable alleles. The complex is the structural locus of a family of potassium channels as well as a number of functions relevant to the biology of the nervous system. The constituents of ShC seem to be linked by functional relationships in view of the similarity of the phenotypes, antimorphic nature of their mutations and the behavior in transheterozygotes. We discuss the relationship between the genetic organization of ShC and the functional coupling of potassium currents with the other functions encoded in the complex.  相似文献

Summary In developing and normal adult rat liver the expression patterns of the mRNAs for -fetoprotein (AFP) and albumin (ALB) were analysed byin situ hybridization using specific35S-labelled complementary DNA probes. In the developing liver AFP and ALB mRNA are found from embryonic day (ED) 11 and 12, respectively, onward. At ED 20 the first signs of a zonal distribution of these mRNAs across the liver lobule can be observed, AFP mRNA concentration being higher in the pericentral area and ALB mRNA concentration higher in the periportal area. This distribution pattern of reciprocal, overlapping gradients of mRNA can be clearly recognized in the neonatal period. In the adult liver AFP mRNA can no longer be detected and similar to the neonatal situation, ALB mRNA is expressed across the entire porto-central distance decreasing in concentration going from the portal to the central area.Transient extra-hepatic expression of AFP mRNA is found in the embryonic heart and in the epithelial lining of intestine and lung furthermore, AFP and ALB mRNA are found to be transiently expressed in the developing renal tubules. Similar expression patterns have been observed for other liver-characteristic mRNAs (Moormanet al., 1990), suggesting that common regulatory factors are operative during development.  相似文献

Phytoplankton primary production, nutrient concentrations and turbidity were monitored at three stations in the Oosterschelde during 1980–1984 as part of an ecosystem study.From comparisons of dissolved nutrient ratios with the nutrient requirements of phytoplankton, and of ambient nutrient concentrations with half-saturation constants for nutrient uptake by natural phytoplankton populations it was concluded that silicate was a limiting nutrient for diatoms after the spring bloom until the end of the summer. Dissolved inorganic nitrogen and phosphate were not considered to be limiting to phytoplankton growth.In general, the phytoplankton growing season started during the first fortnight of April and ended at the end of September. Column production in the whole Oosterschelde varied between 201 and 540 g C m–2 yr–1 and was, on average, 25% higher in the western part than in the eastern part. Basin production in the Oosterschelde varied between 120 and 466 g C m–2 yr–1 and was, on average, 55% higher in the western part than in the eastern part; this difference could be explained by differences in the ratio of euphotic depth to mean depth of the compartments.Estimated carbon-specific growth rates in the eastern part varied between < 0.1 and 3 d–1 and in the western part between < 0.1 and 1 d–1. This difference could be explained by the great differences in depth of the compartments. Carbon-specific growth rates are discussed in relation to phytoplankton loss rates. In the evaluation of the role of lake restoration programmesin situ measurements of the filtration rate of the freshwater musselDreissena polymorpha have been performed in Lake Wolderwijd, The Netherlands. The filtration rate mainly depends on the suspended matter content of the water, and shows an inverse exponential relationship with this factor. The filtration activity is temperature indifferent between approx. 5 and 20 °C. At low temperatures the filtration rate drops abruptly, at high temperatures gradual inhibition occurs. The filtration rate shows a sigmoidal relation with the length of the mussel. The largestD. polymorpha have a diminished filtration rate compared to animals of smaller size. This might be a degenerative feature of the oldest mussels. In Lake Wolderwijd a population density of 675 per m2 is required to compensate phytoplankton growth by grazing. Manipulation of the size of the population can be executed by adding suitable substrates for the mussel.  相似文献

An unusual compound, cyclic-bis(3'----5') diguanylic acid (c-di-GMP or cGpGp), is involved in the regulation of cellulose synthesis in the bacterium Acetobacter xylinum. This cyclic dinucleotide acts as an allosteric, positive effector of cellulose synthase activity in vitro (Ka = 0.31 microM) and is inactivated via degradation by a Ca2(+)-sensitive phosphodiesterase, PDE-A (Km = 0.25 microM). A series of 13 analogs cyclic dimer and trimer nucleotides were synthesized, employing a phosphotriester approach, and tested for the ability to mimick c-di-GMP as activators of cellulose synthase and as substrates for PDE-A. Seven of the synthetic compounds stimulate cellulose synthase activity and all of these activators undergo the Ca2(+)-inhibited degradation reaction. The order of affinities for synthase activators is cGpGp approximately cdGpGp approximately cGp(S)Gp (S-diastereomer) greater than cIpGp greater than cdGpdGp greater than cXpGp greater than cIpIp greater than cGp(S)Gp (R-diastereomer). Three cyclic dinucleotides of negligible affinity for either enzyme are cApAp, cUpUp, and cCpCp. This same order of affinities essentially pertains to the analogs as inhibitors of PDE-A activity, but at least one cyclic dinucleotide, cXpXp, which does not bind to cellulose synthase, is also a substrate for the degradation reaction, demonstrating that although the two enzymes share a similar, high degree of specificity for c-di-GMP, their cyclic dinucleotide binding sites are not identical. Phosphodiester bonds of activators in which an exocyclic oxygen is replaced with an atom of sulfur (cGp(S)Gp isomers) resist the action of PDE-A, and such derivatives may be prototypes for synthetic non-hydrolyzable c-di-GMP analogs.  相似文献

The reduction of a series of 2,5-bis(1-aziridinyl)-1,4-benzoquinone (BABQ) derivatives with various 3,6 substituents by the enzyme xanthine oxidase has been studied. The reduction rate has been assayed by measuring the rate of reduction of cytochrome c, which is very efficiently reduced by reduced BABQ species. Under nitrogen, the reduction rate correlated with the quinone reduction potential and steric parameters. Comparing reduction rates under nitrogen and air demonstrates that at BABQ concentrations greater than 25 microM the competition for electrons from xanthine oxidase between oxygen and the BABQ derivative is dominated by the latter. This is also confirmed by the effect of superoxide dismutase (SOD): in the presence of a BABQ derivative, cytochrome c reduction can be totally inhibited by SOD, although the required amount of SOD depends on the redox potential of the quinones. This indicates that SOD causes the equilibrium between semiquinone and superoxide to shift, resulting in a decrease of the semiquinone concentration. It is concluded that reduction by xanthine oxidase is a simple and effective method for reducing aziridinylbenzoquinones.  相似文献

The effect of guanidine chloride on the activity of bovine heart lactate dehydrogenase transferred to a system that was made with toluene, phospholipids, Triton X-100 and 3.8% water (v/v) was studied. The activity of the enzyme in the latter system was about 30 times lower than in standard water mixtures. In the low water system, 1.5 and 2.0 M guanidine chloride increased the activity by approximately 20 times. These concentrations of guanidine chloride caused complete inactivation of the enzyme in conventional water systems. The activating effect of the denaturant was independent of enzyme concentration. It is suggested that the increase in activity produced by guanidine chloride was due to a facilitation of the protein-solvent interactions that operate in a catalytic cycle.  相似文献
