Alkaloid extracts from 12 plant species of the families Amaryllidaceae, Fumariacae and Papaveraceae were studied with respect to their acetylcholinesterase inhibitory activity and alkaloid patterns. Fifty-three alkaloids were identified by GC-MS, including known acetylcholinesterase (AChE) inhibitors such as galanthamine, epigalanthamine, sanguinine and epinorgalanthamine in extracts of Amaryllidaceae plants and protopine in extracts of Fumariaceae and Papaveraceae plants. The galanthamine-containing extracts of the amaryllidaceous plants were found to be the most active while the extract of Corydalis bulbosa was the most active among the extracts of the tested plants from the Fumariaceae and Papaveraceae plants. TLC bioautographic assay, preparative TLC and GC-MS analysis were combined to identify the active compounds in the studied extracts. Galanthamine was isolated from the known AChE inhibitors in the extracts of Amaryllidaceae plants. Corydaline, bulbocapnine and stylopine were found to be active in the extracts of plant species of the families Fumariaceae and Papaveraceae. Available standards of deshydrocorydaline--a precursor of corydaline, corydaline and stylopine--were tested for AChE inhibitory activity. The taphonomy is a powerful and requisite tool for environmental reconstructions of ancient plant communities. Necrobiotic processes, which lead to the production of plant fragments, inform us on fossil plant physiology. Among the processes that can be drawn from necrobiotic studies is the retention of leaf organs on plants, the relative quantity of pollen grains produced by different fossil species or the significance of wildfire dynamics in ancient plant communities. Biostratinomy examination is a fundamental tool for elucidating fossil plant habitats. Numerous experimental data allow paleobotanists for evaluating the role of transport in the origin of fossil assemblages. Autochthonous plant assemblages, which are characterised by the preservation of fossil rooting structures, are relatively rare in the nature. In consequence, the search for palaeoecological information from parautochthonous to allochthonous assemblages has been a priority in taphonomy. As a result, taphonomic models have been elaborated in well-known sedimentological contexts, such as small lacustrine deltas, which allow for the distinction between riparian or perideltaic plant remains. Lithospheric processes modify plant debris after burial. The differences in the degrees of transformations (or alterations) during the diagenesis provide for information about the original morphology and biochemical composition of the plant tissues, which are also paleoecologically useful. Thus, amber diagenesis modifies resin biochemistry into new molecules that are still informative from the chemotaxonomical point of view. 相似文献

The morphology (mainly prosthecae length), ultrastructure, and antenna pigment composition of the green sulfur bacterium Prosthecochloris aestuarii changed when grown under different light intensities. At light intensities of 0.5 and 5 micromol quanta m(-2) s(-1), the cells had a star-like morphology. Prosthecae, the characteristic appendages of the genus Prosthecochloris, were 232 nm and 194 nm long, respectively. In contrast, when grown at 100 micromol quanta m(-2) s(-1), these appendages were shorter (98 nm) and the cells appeared more rod-shaped. Transmission electron microscopy revealed a significant decrease in the cell perimeter to area ratio and in the number of chlorosomes per linear microm of membrane as light intensity increased. In addition to these morphological and ultrastructural responses, Prosthecochloris aestuarii exhibited changes in its pigment composition as a function of light regime. Lower specific pigment content and synthesis rates were found in cultures grown at light intensities above 5 micromol quanta m(-2) s(-1). A blue shift in the bacteriochlorophyll (BChl) c Q(y) absorption maximum of up to 17.5 nm was observed under saturating light conditions (100 micromol quanta m(-2) s(-1)). This displacement was accompanied by changes in the composition of BChl c homologs and by a very low carotenoid content. The morphological, ultrastructural and functional changes exhibited by Prosthecochloris aestuarii revealed the strong light-response capacity of this bacterium to both high and low photon-flux densities. 相似文献

In most plants, sucrose is the primary product of photosynthesis, the transport form of assimilated carbon, and also one of the main factors determining sweetness in fresh fruits. Traditional methods for sugar quantification (mainly sucrose, glucose and fructose) require obtaining crude plant extracts, which sometimes involve substantial sample manipulation, making the process time-consuming and increasing the risk of sample degradation. Here, we describe and validate a fast method to determine sugar content in intact plant tissue by using high-resolution magic angle spinning nuclear magnetic resonance spectroscopy (HR-MAS NMR). The HR-MAS NMR method was used for quantifying sucrose, glucose and fructose in mesocarp tissues from melon fruits (Cucumis melo var. reticulatus and Cucumis melo var. cantalupensis). The resulting sugar content varied among individual melons, ranging from 1.4 to 7.3 g of sucrose, 0.4–2.5 g of glucose; and 0.73–2.83 g of fructose (values per 100 g fw). These values were in agreement with those described in the literature for melon fruit tissue, and no significant differences were found when comparing them with those obtained using the traditional, enzymatic procedure, on melon tissue extracts. The HR-MAS NMR method offers a fast (usually <30 min) and sensitive method for sugar quantification in intact plant tissues, it requires a small amount of tissue (typically 50 mg fw) and avoids the interferences and risks associated with obtaining plant extracts. Two new photoactive compounds (1 and 2) derived from the 9-amidoacridine chromophore have been synthesized and fully characterized. Their abilities to produce singlet oxygen upon irradiation have been compared. The synthesized compounds show very different self-aggregating properties since only 1 present a strong tendency to aggregate in water. Biological assays were conducted with two cell types: hepatoma cells (Hep3B) and human umbilical vein endothelial cells (HUVEC). Photodynamic therapy (PDT) studies carried out with Hep3B cells showed that non-aggregating compound 2 showed photoxicity, ascribed to the production of singlet oxygen, being aggregating compound 1 photochemically inactive. On the other hand suspensions of 1, characterized as nano-sized aggregates, have notable antiproliferative activity towards this cell line in the dark. 相似文献

Allee effects reduce the viability of small populations in many different ways, which act synergistically to lead populations towards extinction vortexes. The Sierra Morena wolf population, isolated in the south of the Iberian Peninsula and composed of just one or few packs for decades, represents a good example of how diverse threats act additively in very small populations. We sequenced the genome of one of the last wolves identified (and road‐killed) in Sierra Morena and that of another wolf in the Iberian Wolf Captive Breeding Program and compared them with other wolf and dog genomes from around the world (including two previously published genome sequences from northern Iberian wolves). The results showed relatively low overall genetic diversity in Iberian wolves, but diverse population histories including past introgression of dog genes. The Sierra Morena wolf had an extraordinarily high level of inbreeding and long runs of homozygosity, resulting from the long isolation. In addition, about one‐third of the genome was of dog origin. Despite the introgression of dog genes, heterozygosity remained low because of continued inbreeding after several hybridization events. The results thus illustrate the case of a small and isolated wolf population where the low population density may have favoured hybridization and introgression of dog alleles, but continued inbreeding may have resulted in large chromosomal fragments of wolf origin completely disappearing from the population, and being replaced by chromosomal fragments of dog origin. Two charophyte lineages from the Upper Cretaceous–Palaeocene of southern Europe are described here, constituting a useful biostratigraphic tool for correlating non-marine stratigraphic sequences. The Peckichara pectinata lineage, ranging from the middle Campanian to the middle–upper Maastrichtian, consists of four successive gyrogonite morphotypes that displayed progressively more complex gyrogonite ornamentation and increasingly smaller gyrogonites with time. The Peckichara sertulata lineage, ranging from the middle Campanian to the Danian, consists of three morphotypes that followed a more common evolutionary trend, developing progressively larger gyrogonites without any changes in ornamentation. Intermediate forms between the successive morphotypes suggest that they corresponded to anagenetic varieties of the same evolutionary species rather than to separate species. The different trends observed appear to be palaeoenvironmentally controlled. The grades of the P. pectinata lineage first occurred in permanent lakes that later shifted to fluvial influenced floodplain ponds, this shift accounting for the reduction in gyrogonite size. By contrast, the P. sertulata lineage mainly occurred in permanent lakes, where stable palaeoenvironmental conditions enabled a steady rise in gyrogonite size to increase the space available for the zygote. 相似文献

Cystic fibrosis (CF) is thought to be rare among the Arab populations from the Middle East and little data have been reported
so far. We have studied a sample of 20 families living in Lebanon for several generations and who have at least one child
with CF. These families are mainly from the Maronite, Greek Catholic, Greek Orthodox, Shiite or Sunnite groups. We found a
50% rate of consanguineous marriage, independent of the community of origin. The distribution of CF genotypes was determined
through the screening of all exons of the CFTR (cystic fibrosis transmembrane conductance regulator) gene by the technique
of denaturing gradient gel electrophoresis combined with asymmetric amplification DNA sequencing. A total of ten different
mutations accounting for 87.5% of 32 unrelated CF alleles was identified, including two novel putative mutations (E672del
and IVS21-28G→A). Three mutations, ΔF508 (37.5%), W1282X (15.6%), and N1303K (9.4%) accounted for 62.5% of CF alleles. Interestingly,
in the Maronite group, 66.7% of the ΔF508 chromosomes were found to be associated with allele 7 of the IVS8(T)tract, contrasting
with the absolute linkage disequilibrium between European ΔF508 chromosomes and allele 9. During this study, two previously
undescribed polymorphisms (IVS14a + 17del5 and 2691T/C) were also identified.
