Gas Chromatographic-Mass Spectrometric Determination of myo-Inositol in Human Cerebrospinal Fluid

The concentration of free myo-inositol in CSF was determined with a gas chromatographic-mass spectrometric method using deuterated myo-inositol as an internal standard after conversion to the hexa-O-acetyl derivative with acetic anhydride and pyridine. Twenty microliters of CSF is sufficient for the analysis which has a coefficient of variation of 9%. Identical analytical results were obtained on two different mass numbers. Schizophrenic patients were compared with healthy control persons. In addition, patients with rheumatoid arthritis or with neurological illnesses were studied. No consistent differences related to the illness could be found. The mean concentration of myo-inositol was about 25 micrograms/ml. Treatment of schizophrenic patients with chlorpromazine or sulpiride had no significant effect on the concentration of myo-inositol in CSF.     

Overexpression of the endogenous peroxidase-like gene spi 2 in transgenic Norway spruce plants results in increased total peroxidase activity and reduced growth

Peroxidases constitute a large family of proteins found in all higher plants. Owing to the complexity of the peroxidase isoenzyme family it has been difficult to assess the precise function of individual peroxidase enzymes. In this work we have studied the effects of an endogenous peroxidase-like gene from Norway spruce [Picea abies (L.) Karst], spi 2, on the development and growth of Norway spruce somatic embryo plants. Embryogenic cells of Norway spruce transformed with spi 2 under control of the maize ubi-1 promoter showed up to 40 times higher total peroxidase activity than the control cells; regenerated plants overexpressing spi 2 showed an increased total peroxidase activity. Based on these results and the overall sequence similarity with cationic peroxidases we conclude that spi 2 encodes a peroxidase. Overexpression of spi 2 resulted in increased sensitivity to stress, leading to a reduction in epicotyl formation and in height growth compared with control plants. The plants overexpressing spi 2 also showed a deeper phloroglucinol staining but similar levels of Klason lignin.     

Identification and Determination of Tele-Methylhistamine in Cerebrospinal Fluid by Gas Chromatography-Mass Spectrometry

Abstract: The presence of tele-methylhistamine in human cerebrospinal fluid has been established. The concentration was determined with the use of deuterated tele-methylhistamine. The preparation of the deuterated standard is described. The concentration range in samples from neuropsychiatric patients was 0.1-2.5 ng/ml. The structure of the pentafluoropropionyl derivative used for gas chromatography was studied with the aid of proton nuclear magnetic resonance spectroscopy.
