Auchenorrhyncha of a new polder were sampled by different kinds of traps in four stations in new habitats, one station on the salt marsh and one just outside the polder. In the latter stations 20 species of Auchenorrhyncha were caught in numbers that declined during the four year study. The proportion of macropters of the common Streptanus sordidus was highest in the largest catches. In the polder proper 392 specimens of 13 species were caught, all had moved at least 1500 m, but some could be traced to origins at greater distances. The number of immigrants also declined during the study. The distribution of the leafhoppers over the trapping stations suggests that leafhoppers, although sometimes transported over great distances, as a rule quickly fall out once they are airborne. Probably only one species had founded a resident population in the new polder at the end of the study.  相似文献

HLA antigens have been purified to homogeneity after detergent solubilization from RPMI 4265, a human lymphoblastoid line. The inhibition of cytotoxicity assay for HLA antigen was modified, using preincubation with bovine serum albumin of antigen samples containing detergent to prevent lysis of target cells by detergent. Solubilization was tested with many types of detergents. A polyethyleneglycol oleyl ether nonionic detergent mixture, Brij 99:Brij 97 (2:1) was selected for solubilization, since it selectively solubilized HLA antigens, had a low absorbance at 280 nm and was uncharded. HLA antigens were then purified by Lens culinaris lectin affinity chromatography and Bio-Gel A-5m filtration. The antigen specifity HLA-A2 was separated from specificities HLA-B7,12 by isoelectric focusing. Purified HLA antigens contained a subunit of Mr=44,000 with NH2-terminal glycine, and a subunit of Mr=12,000, beta2-microglobulin, with NH2-terminal isoleucine.  相似文献

A rapid method is described for the identification of lectin binding membrane glycocomponents in polyacrylamide gels. The method requires only small quantities of membrane material and is applicable to a wide variety of lectins. Solubilized membrane components are electrophoretically separated according to molecular weight in a SDS-polyacrylamide gel. The gel is then incubated in a solution containing the 125I-lectin. After elution of unbound 125I-lectin, the gel is dried down and those bands which have bound the 125I-lectin are identified by radioautography. The amount of bound 125I-lectin can be quantified by either densitometric scanning of the radioautogram or by liquid scintillation counting of the dried gel.  相似文献

A simplified mathematical model of the origin of the left-handed asymmetry of proteins in living matter is presented. The model is based on the hypothesis of Vester and Ulbricht that the chirality of (lefthanded) electrons from naturally beta-active elements, e.g.,14C,40K, etc., was the specific source of the asymmetry; it requires for its application data on the interaction of electrons having non-zero chirality with racemic mixtures of amino acids. This interaction is here treated theoretically in an order-of-magnitude calculation. Our analysis yields a very approximate value of the induced steady-state asymmetry in the amino acids at the beginning of protein synthesis and indicates that this asymmetry, though small, may have been suffcient to account for the dominant left-handedness of proteins now observed.  相似文献

Summary Myosin was isolated in high purity from the bovine adrenal medulla by gel filtration and ion exchange chromatography. The purified myosin was analyzed by electrophoresis in gels containing SDS and found to contain a 200,000 molecular weight heavy chain and major light chains of molecular weights 20,000 and 17,000 in a 111 molar ratio. At high ionic strength the myosin had high Ca-ATPase and K-EDTA-ATPase activities and low Mg-ATPase activity. At low ionic strength, the Mg-ATPase was activated to a low level by rabbit muscle actin. The myosin was found to decorate F-actin in the absence, but not the presence of ATP. In low ionic strength solutions, the myosin assembled into characteristic bipolar filaments.The distribution of this myosin in the adrenal medulla and of cross-reacting myosin in several other bovine tissues was determined with the use of antimedullary myosin immunoglobulin G as a specific stain that was detected by direct and indirect immunofluorescence. In the medulla strong staining was seen between the chords of chromaffin cells indicating the presence of a highly muscular vasculature that may perform functions analogous to those of the myoepithelium of exocrine glands. The chromaffin cells showed weak positive staining around the nuclei and in a pattern radiating toward adjacent blood vessels. Cells of the inner zone of the adrenal cortex showed strong staining in the peripheral cytoplasm while cells in the intermediate and outer zones did not stain. In a blood smear, platelets and the cytoplasm of leukocytes stained strongly while erythrocytes did not stain. In striated muscle and the gray and white matter of the cerebrum only the capillaries and larger vessels stained. In the liver the phagocytic cells bordering vascular sinuses stained strongly while the hepatocytes were separated from one another by a 2 micron trilaminar band possibly representing the microfilament web surrounding the bile canaliculi and associated with junctional complexes.The results suggest that myosin is present in several highly differentiated, non-motile tissue cells where it may play a role in secretion or other specialized functions.The author gratefully acknowledges the support and encouragement received from Francis D. A stable kelp bed ecosystem in St. Margaret's Bay, Nova Scotia (Canada), had as its main producersLaminaria longicruris andL. digita. Most algal production was exported as detritus, but there was a moderate population of herbivores, mainly the sea urchinsStrongylocentrotus droebachiensis. These were eaten by crabs,Cancer irroratus and by lobsters,Homarus americanus. Lobsters also preyed on crabs. Beginning in 1968, sea urchins became locally abundant and overgrazed the kelp beds, converting large areas to urchindominated barren grounds. Almost all kelp beds in St. Margaret's Bay (140 km2) have now been destroyed. During the same period, lobster biomass decreased, and the hypothesis was put forward that reduction in lobster predation led to increased urchin abundance and kelp bed destruction. Evidence is presented for the hypothesis that urchin-dominated barren grounds are a new, stable configuration of the ecosystem, and that a long-term decrease in primary and secondary productivity of these coastal waters can be expected.  相似文献

Microbial transformations of 14C-labeled substrates (sodium glutamate, Casamino Acids, glucose, and sodium acetate) were measured in undecompressed seawater samples collected from depths of 1,800 to 6,000 m, during 14- to 21-day incubation periods at in situ temperature (3°C). Each substrate was tested at two concentrations (ca. 0.5 and 5.0 μg/ml) and two in situ pressures. The data were compared to 1-atmosphere (ca. 1.013 × 102 kPa) controls. The rates of 14C incorporation and 14CO2 production as well as the amounts of total substrate utilization were generally lower at pressure than in the decompressed controls but were significantly different for each of the four substrates used. The utilization of acetate was the least affected by pressure; rates were similar to those measured at 1 atmosphere in two out of four experiments. In contrast, transformation rates of the amino acids at pressure averaged to only 38% of those in the controls. A single but reproducible “barophilic” response was observed with glucose as a substrate in samples collected from a depth of 4,500 m at a specific area in the northwestern Atlantic Ocean. Except for this latter set of experiments, the transformation of all substrates showed an increased lag period at pressure as compared to the 1-atmosphere controls.  相似文献

A cytosolic glutathione S-transferase from pig lung was purified 210-fold to apparent homogeneity. The enzyme was classified as a class pi isoenzyme on the basis of its physical and chemical properties. It is homodimeric with a subunit Mr of 23,500, has a pI of 7.2, and shows a high specific activity towards ethacrynic acid. The glutathione analogues, S-hexylglutathione and glutathione sulfonate, were strong reversible inhibitors. The enzyme's primary structure, established entirely by protein chemical methods, consists of 203 amino acids and is highly similar (82-84% residue identity) to the rat and human class pi isoenzymes. Furthermore, there was no evidence of microheterogeneity or post-translational modifications. Each subunit contains a highly reactive cysteine residue, the modification of which leads to enzyme inactivation. None of the cysteine residues in the pig enzyme appear to form intramolecular disulfide bonds. Singel crystals of the glutathione-S-transferase-glutathione-sulfonate complex were obtained by the hanging-drop method of vapour diffusion from poly(ethylene glycol) 4000 solutions. The crystals belong to the orthorhombic space group P212121 with unit cell dimensions of a = 10.125 nm, b = 8.253 nm and c = 5.428 nm and diffract to better than 0.22 nm.  相似文献

A 5 kilobase deletion in mitochondrial DNA (mtDNA) has been reported to be responsible for the specific complex I deficiency in the substantia nigra (SN) of the Parkinson's disease (PD) brain. We have studied mitochondrial respiratory chain function in the SN from control and PD subjects, and analysed mtDNA, extracted from the same tissues, by Southern blot and the polymerase chain reaction (PCR). Quantitation of the levels of the deletion indicate that it does not contribute to the pathogenesis of PD nor to a complex I deficiency but seems likely to be an age-related observation.  相似文献
