The role of Grb7 adapters, Grb7, Grb10, and Grb14, was investigated in Xenopus oocytes expressing fibroblast growth factor receptors (FGFR). FGF-induced maturation of FGFR-expressing oocytes was blocked by previous injection of Grb7 or Grb14, but not Grb10. This effect correlated with Grb7/14 binding to the receptor, and inhibition of the Ras-dependent pathway. Interestingly, the phosphorylated insulin receptor interacting region (PIR) and Src 2 homology domains (SH2) of Grb7 and Grb14 were differently implicated in the inhibition of FGFR signalling. This study provided further evidence for specificity of the biological action of the Grb7 adapters on receptor tyrosine kinase signalling.  相似文献

We show that p27 localization is cell cycle regulated and we suggest that active CRM1/RanGTP-mediated nuclear export of p27 may be linked to cytoplasmic p27 proteolysis in early G1. p27 is nuclear in G0 and early G1 and appears transiently in the cytoplasm at the G1/S transition. Association of p27 with the exportin CRM1 was minimal in G0 and increased markedly during G1-to-S phase progression. Proteasome inhibition in mid-G1 did not impair nuclear import of p27, but led to accumulation of p27 in the cytoplasm, suggesting that export precedes degradation for at least part of the cellular p27 pool. p27-CRM1 binding and nuclear export were inhibited by S10A mutation but not by T187A mutation. A putative nuclear export sequence in p27 is identified whose mutation reduced p27-CRM1 interaction, nuclear export, and p27 degradation. Leptomycin B (LMB) did not inhibit p27-CRM1 binding, nor did it prevent p27 export in vitro or in heterokaryon assays. Prebinding of CRM1 to the HIV-1 Rev nuclear export sequence did not inhibit p27-CRM1 interaction, suggesting that p27 binds CRM1 at a non-LMB-sensitive motif. LMB increased total cellular p27 and may do so indirectly, through effects on other p27 regulatory proteins. These data suggest a model in which p27 undergoes active, CRM1-dependent nuclear export and cytoplasmic degradation in early G1. This would permit the incremental activation of cyclin E-Cdk2 leading to cyclin E-Cdk2-mediated T187 phosphorylation and p27 proteolysis in late G1 and S phase.  相似文献

In this study, the phylogenetic relationships of 164 species of the family Drosophilidae are discussed, using the Amyrel gene, a member of the α -amylase multigene family. This study focuses on numerous species groups in the subgenera Sophophora and Drosophila of the genus Drosophila but also includes other closely related genera. Nucleotide data were analysed by several methods: maximum parsimony, neighbour joining, maximum likelihood and Bayesian inference. Heterogeneity of base composition (mainly low GC contents in the species groups willistoni and saltans ) has been addressed. In all analyses, the genus Drosophila appeared paraphyletic. The subgenus Sophophora clearly appeared to be a monophyletic group, showing well-resolved clades, with the Neotropical groups arising in a basal position. Here, it is proposed to raise the species subgroups ananassae and montium to the rank of species group, and to restrict the melanogaster species group to the melanogaster subgroup plus the 'Oriental' subgroups, among which the suzukii subgroup is polyphyletic. Some related genera such as Zaprionus , Liodrosophila , Scaptomyza and Hirtodrosophila are clustered with, or inside the subgenus Drosophila , which is therefore paraphyletic and should be reviewed.  相似文献

Using isoelectric focusing, the zymograms of 23 pathogenic and nonpathogenic Naegleria strains were studied for the activity of 16 enzymes. Certain enzymes (lactate dehydrogenase, L-threonine dehydrogenase, superoxide dismutase, acid phosphatase, malic enzyme, and leucine aminopeptidase) proved particularly useful from a practical point of view as they allow easy and reliable identification of pathogenic N. fowleri and N. australiensis as well as nonpathogenic N. lovaniensis strains. Genetic interpretation of these zymograms gave estimates of genetic distances that largely confirmed the taxonomic position of the Naegleria species. In addition, the genetic data suggest that there are two main phylogenetic groups in the genus Naegleria.  相似文献

Made up of calcareous coralline algae, maerl beds play a major role as ecosystem engineers in coastal areas throughout the world. They undergo strong anthropogenic pressures, which may threaten their survival. The aim of this study was to gain insight into the future of maerl beds in the context of global and local changes. We examined the effects of rising temperatures (+3°C) and ocean acidification (?0.3 pH units) according to temperature and pH projections (i.e., the RCP 8.5 scenario), and nutrient (N and P) availability on three temperate maerl species (Lithothamnion corallioides, Phymatolithon calcareum, and Lithophyllum incrustans) in the laboratory in winter and summer conditions. Physiological rates of primary production, respiration, and calcification were measured on all three species in each treatment and season. The physiological response of maerl to global climate change was species‐specific and influenced by seawater nutrient concentrations. Future temperature–pH scenario enhanced maximal gross primary production rates in P. calcareum in winter and in L. corallioides in both seasons. Nevertheless, both species suffered an impairment of light harvesting and photoprotective mechanisms in winter. Calcification rates at ambient light intensity were negatively affected by the future temperature–pH scenario in winter, with net dissolution observed in the dark in L. corallioides and P. calcareum under low nutrient concentrations. Nutrient enrichment avoided dissolution under future scenarios in winter and had a positive effect on L. incrustans calcification rate in the dark in summer. In winter conditions, maximal calcification rates were enhanced by the future temperature–pH scenario on the three species, but P. calcareum suffered inhibition at high irradiances. In summer conditions, the maximal calcification rate dropped in L. corallioides under the future global climate change scenario, with a potential negative impact on CaCO3 budget for maerl beds in the Bay of Brest where this species is dominant. Our results highlight how local changes in nutrient availability or irradiance levels impact the response of maerl species to global climate change and thus point out how it is important to consider other abiotic parameters in order to develop management policies capable to increase the resilience of maerl beds under the future global climate change scenario.  相似文献

Malate dehydrogenase has been detected in four species of the super-species Jaera albifrons. Two polymorphic enzymatic systems occur showing a one banded pattern for homozygotes and a three banded pattern for heterozygotes. Two independant autosomal loci control the variation as is shown by intra- and interspecific crosses. In European populations of the three species studied, MDH-1 is polymorphic and MDH-2 is monomorphic. In the American species, J. posthirsuta, the locus MDH-2 is polymorphic and MDH-1 is monomorphic.  相似文献

l-Lactate dehydrogenase occurs in the five species of the super-species Jaera albifrons (Crustacea, Isopoda). Populations which are genetically homogenous show either a fast or a slow single band whereas others which are polymorphic contain a third phenotype with five zones of activity. The enzyme is thus probably tetrameric. The variation is controlled by a single autosomal locus, as is shown by intra and interspecific crosses.  相似文献

In 1995 a strain of Ectocarpus was isolated from Hopkins River Falls, Victoria, Australia, constituting one of few available freshwater or nearly freshwater brown algae, and the only one belonging to the genus Ectocarpus. It has since been used as a model to study acclimation and adaptation to low salinities and the role of its microbiota in these processes. To provide more background information on this model, we assessed if Ectocarpus was still present in the Hopkins river 22 years after the original finding, estimated its present distribution, described its abiotic environment, and determined its in situ microbial composition. We sampled for Ectocarpus at 15 sites along the Hopkins River as well as 10 neighboring sites and found individuals with ITS and cox1 sequences identical to the original isolate at three sites upstream of Hopkins River Falls. The salinity of the water at these sites ranged from 3.1 to 6.9, and it was rich in sulfate (1–5 mM). The diversity of bacteria associated with the algae in situ (1312 operational taxonomic units) was one order of magnitude higher than in previous studies of the original laboratory culture, and 95 alga-associated bacterial strains were isolated from algal filaments on site. In particular, species of Planctomycetes were abundant in situ but rare in laboratory cultures. Our results confirmed that Ectocarpus was still present in the Hopkins River, and the newly isolated algal and bacterial strains offer new possibilities to study the adaptation of Ectocarpus to low salinity and its interactions with its microbiome.  相似文献

Inferring phylogenetic relationships between closely related taxa can be hindered by three factors: (1) the lack of informative molecular variation at short evolutionary timescale; (2) the lack of established markers in poorly studied taxa; and (3) the potential phylogenetic conflicts among different genomic regions due to incomplete lineage sorting or introgression. In this context, Restriction site Associated DNA sequencing (RAD‐seq) seems promising as this technique can generate sequence data from numerous DNA fragments scattered throughout the genome, from a large number of samples, and without preliminary knowledge on the taxa under study. However, divergence beyond the within‐species level will necessarily reduce the number of conserved and non‐duplicated restriction sites, and therefore the number of loci usable for phylogenetic inference. Here, we assess the suitability of RAD‐seq for phylogeny using a simulated experiment on the 12 Drosophila genomes, with divergence times ranging from 5 to 63 million years. These simulations show that RAD‐seq allows the recovery of the known Drosophila phylogeny with strong statistical support, even for relatively ancient nodes. Notably, this conclusion is robust to the potentially confounding effects of sequencing errors, heterozygosity, and low coverage. We further show that clustering RAD‐seq data using the BLASTN and SiLiX programs significantly improves the recovery of orthologous RAD loci compared with previously proposed approaches, especially for distantly related species. This study therefore validates the view that RAD sequencing is a powerful tool for phylogenetic inference.  相似文献

The effects of shift and time‐on‐shift on alertness and perceived tension, as well as related physiological variables, were investigated in satellite controllers working a rapid forward rotating three‐shift system. In controlled laboratory conditions, subjective tension and HR have been reported to display circadian variation and marked sensitivity to external factors. We examined whether circadian variations were masked for these particular variables in real‐job conditions, unlike for alertness and body temperature, which have been repeatedly shown to display circadian variation in these conditions. This hypothesis was tested in a repeated‐measures design by collecting alertness and tension self‐reports and recording operators' sublingual temperature on three occasions on each shift and HR continuously throughout shifts. Alertness and body temperature varied according to a typical diurnal trend; subjective tension was only enhanced on the initial recording of each shift (compared to the remaining ones), while HR displayed an intermediary trend. Intra‐subject correlations revealed a positive relationship between alertness, oral temperature, and HR, while no such relationship was found for subjective tension. These results support the hypothesis of a close dependence of alertness and temperature, and to a lesser extent for HR, on endogenous mechanisms in this job‐situation. In addition, some situation‐specific factors, such as job‐demand, would affect subjective tension and partially mask the circadian variations in HR.  相似文献
