Forage nitrogen concentrations, nitrogen yields, and in vitro digestibilities were assessed in shortgrass steppe that had been ungrazed, lightly, or heavily grazed for 50 years. Caged plots were defoliated in amounts based upon removals observed in naturallygrazed reference plots or not defoliated. This was done in a year of average precipitation and with a supplemental water treatment to simulate a wet year. In general, current-year defoliation had positive effects, and longterm grazing and supplemental water had negative effects, on forage nitrogen concentrations and digestibilities. However, defoliation interacted with long-term grazing in determning forage nitrogen concentrations, and with grazing and with watering in determining digestibilities. Nitrogen concentration and digestibility increased with defoliation in lightly, but not in heavily, grazed treatments. The dilution effect of supplemental water an digestibilities through increased plant growth was offset by defoliation. The negative effects of long-term grazing on forage quality were small, equally or more than compensated for by defoliation in a year of average precipitation, but more pronounced in the simulated wet year. Nitrogen yields and digestible forage production were usually increased by defoliation, but this depended upon grazing and watering treatments. Increased nitrogen and digestible forage yields and concentrations in response to defoliation were greater than the biomass response in lightly grazed grassland. For both nitrogen and digestibility, yields were greater in grazed than ungrazed treatments in the year of average precipitation, but less in the simulated wet year. Optimizing quantity and year-to-year stability of nitrogen and digestible forage yield may best be achieved with light grazing rather than no or heavy grazing. Clipping was conducted in a manner closely resembling the natural pattern and intensity of defoliation by the cattle, and confirms the potential for a positive feedback of increased forage quality with defoliation observed in pot experiments. Long-term heavy grazing can diminish this response. Quantily (aboveground primary production, ANPP), quantity of quality (digestible and N yields), and quality (concentrations) do not necessarily respond similarly in interactions between current-year defoliation, long-term grazing history, and level of water resource.  相似文献

Lysergyl peptide synthetase 1 catalyzes the assembly of toxic ergopeptines from activated D-lysergic acid and three amino acids. The gene encoding this enzyme in the endophytic fungus Neotyphodium lolii was analyzed and compared to a homologous gene from the ergot fungus Claviceps purpurea. Each gene contained two introns, which were found in the same relative position within two modules of the gene. The 5' ends of the two genes were unusually divergent. Signature sequences determining substrate specificity were similar in adenylation domains that recognized identical amino acids but differed within the adenylation domain for the amino acid that varies between the major ergopeptines of the two fungi. Homologues were detected in several related endophytic fungi; the tall fescue endophyte Neotyphodium coenophialum contained a divergent, second copy of the gene. Our results provide new information on the structure and distribution of this important peptide synthetase involved in ergot alkaloid biosynthesis.  相似文献

Decision making in risky situations is frequently required in our everyday lives and has been shown to be influenced by various factors, some of which are independent of the risk context. Based on previous findings and theories about the central role of perceptions of control and their impact on subsequent settings, spillover effects of subjective loss of control on risky decision-making are assumed. After developing an innovative experimental paradigm for inducing loss of control, its hypothesized effects on risky decision-making are investigated. Partially supporting the hypotheses, results demonstrated no increased levels of risk perceptions but decreased risk-taking behavior following experiences of loss of control. Thus, this study makes a methodological contribution by proposing a newly developed experimental paradigm facilitating further research on the effects of subjective loss of control, and additionally provides partial evidence for the spillover effects of loss of control experiences on risky decision-making.  相似文献

Masking of circadian activity rhythms in canaries by light and dark   总被引:1,自引:0,他引:1  
Canaries (Serinus canaria) were kept singly in cages placed in an artificially illuminated, soundproof cabinet. Perch-hopping activity was recorded by means of a computer system. In three series of experiments, the activity rhythms of the birds were entrained to 24 hr by light-dark (LD) cycles with 4, 12, or 20 hr of light (L), respectively. The intensity of illumination was 10 lux in L and 0.25 lux in darkness (D). Under LD 4:20 and 12:12, the intensity of D was increased daily at the same zeitgeber time to 1 lux for 1 hr (L pulse) during about 8 consecutive days. This sequence was followed by 8 days without L pulses before giving another series of L pulses at a different zeitgeber time. Under LD 20:4, the intensity of L was decreased to 1 lux for 1 hr (D pulse). The activity of all birds was more or less increased by the L pulses (positive masking) and decreased by the D pulses (negative masking). The level of masking activity during the L and D pulses depended on the circadian phase at which the pulses were administered. Positive masking by L pulses was minimal about 5 hr after the beginning of D, and increased steadily thereafter. Negative masking by D pulses was maximal at the beginning and the end of L, and minimal during the middle.  相似文献

Effects of feeding cycles on circadian rhythms in squirrel monkeys   总被引:1,自引:0,他引:1  
Squirrel monkeys (Saimiri sciureus) were housed singly in cages equipped with a tree for climbing to measure locomotor activity, and with a movable food cup that could be arrested automatically. The animals were kept in continuous dim illumination (LL), twice interrupted by several weeks of entrainment by a light-dark (LD) 12:12 cycle. Apart from three control sections in which the food cups were unlocked continuously (ad libitum feeding), food was accessible for 3 hr per day only, with interfeeding intervals varying from 23 to 26 hr (periodic restricted feeding, or RF). During LD entrainment, the imposition of an RF schedule resulted in anticipatory behaviors, represented by increased tugs at the food cup and a pause in locomotor activity preceding the feeding time. In LL, the animals showed free-running circadian rhythms of locomotor and "feeding" activity that nearly always persisted when ad libitum feeding was replaced by RF. The period (tau) of the free-running rhythm was slightly modulated in relation to the varying interfeeding intervals (T), but entrainment was never achieved except in one test with an animal whose tau was very close to T. It is concluded that periodic availability of food represents an extremely weak zeitgeber, if any, for the circadian pacemaker of squirrel monkeys.  相似文献

In an underground isolation unit, 46 subjects lived singly without time cues under conditions of self-selected light-dark (LDs) cycles or constant illumination (LL). They all developed free-running circadian rhythms that either remained internally synchronized (i.e., with equal periods in all functions; n = 24) or became desynchronized by a sudden lengthening (n = 14) or shortening (n = 8) of the sleep-wake cycle. Six further subjects were synchronized to 24 hr by an externally controlled LD cycle. All subjects had to prepare their own meals. Signals were given by the subjects when they woke up, had a meal, and retired. Of the 52 subjects, 43 consistently had three meals per "day," and 9 had two meals, irrespective of the length of the circadian cycle and despite the fact that, due to desynchronization, wakefulness varied from over 30 hr to less than 12 hr. The intervals between meals, between wake-up time and breakfast, and between last meal and bedtime were "stretched" or "compressed" in strong proportionality to the duration of wakefulness.  相似文献

Several months following parenteral injections of saccharated iron oxide into DBA/2J mice, granules rich in iron were found in nuclei of scattered parenchymal liver cells as well as in the cytoplasm. As seen in the light microscope, the intranuclear granules were brown; most of them measured between 0.5 µ and 1 µ in cross-section. They gave positive Prussian blue tests, and were not selectively stainable with pyronine. Electron micrographs of the granules showed closely packed aggregates of ferritin molecules, occasionally in paracrystalline order. The intranuclear collections were often surrounded by bands of material of moderate opacity. Scattered ferritin molecules and collections of such molecules were also present in the cytoplasm of many liver cells, but there seemed to be no quantitative relationship between intranuclear and cytoplasmic ferritin. Liver cells from untreated control mice failed to reveal intranuclear deposits of ferritin. Although the site of origin of the intranuclear aggregates of ferritin is unknown, the findings suggest the possibility that under suitable circumstances ferritin synthesis may take place within nuclei of liver cells—perhaps induced by the presence of colloidal iron.  相似文献

Increases in plasma vasopressin and renin activity that occur in response to haemorrhage have been attributed in part to reflex effects from cardiac receptors and sinoaortic baroreceptors, but the relative importance of these different receptors in causing humoral changes during haemorrhage in conscious dogs has not been reported. We investigated this question by hemorrhaging 6 sham-operated (SO), 6 cardiac-denervated (CD), 4 sinoaortic-denervated (SAD), and 4 combined sinoaortic and cardiac-denervated (SACD), conscious dogs. Blood was removed at a rate of 0.9 ml/kg X min. Plasma vasopressin and renin samples were taken during a control period and after 10, 20, and 30 ml/kg of blood had been removed. Results (mean +/- SE) are shown in the tables below. (table; see text) These experiments illustrate that: resting plasma levels of vasopressin and renin in conscious dogs are unaffected by the denervation procedures used in these experiments, the increase in plasma vasopressin that occurs during haemorrhage is mediated largely via cardiac receptors, with a considerably smaller contribution from the sinoaortic baroreceptors, during moderately severe haemorrhage (30 ml/kg) vasopressin secretion can be increased by a mechanism independent of sinoaortic and cardiac reflexes, the increase in plasma renin activity that occurs during haemorrhage is not dependent upon either cardiac or sinoaortic reflexes.  相似文献
