In 2005, southwestern Amazonia experienced the effects of an intense drought that affected life and biodiversity. Several major tributaries as well as parts of the main river itself contained only a fraction of their normal volumes of water, and lakes were drying up. The consequences for local people, animals and the forest itself are impossible to estimate now, but they are likely to be serious. The analyses indicate that the drought was manifested as weak peak river season during autumn to winter as a consequence of a weak summertime season in southwestern Amazonia; the winter season was also accompanied by rainfall that sometimes reached 25% of the climatic value, being anomalously warm and dry and helping in the propagation of fires. Analyses of climatic and hydrological records in Amazonia suggest a broad consensus that the 2005 drought was linked not to El Niño as with most previous droughts in the Amazon, but to warming sea surface temperatures in the tropical North Atlantic Ocean.  相似文献

This work presents the binding of AZT and nine novel AZT derivatives to human serum albumin (HSA), both defatted (HSA(D)) and complexed with fatty acids (HSA(FA)). The bound fractions and binding site were determined by applying an ultrafiltration procedure, with an increased affinity for the majority of these derivatives to HSA(D) being found with respect to that of AZT, while only one derivative exhibited an increased affinity for HSA(FA). By means of computational methods, we observed that specific electrostatic interactions are responsible for the increased affinity for HSA(D), while the presence of fatty acids complexed to HSA caused an intense electrostatic repulsion with negatively charged ligands located in Sudlow site I, thus diminishing their bound fractions. A strong relationship between the calculated energetic components and the observed experimental affinity was identified.  相似文献

Angiotensin II (Ang II) AT(2) receptors are abundantly expressed in rat fetal tissues where they probably contribute to development. In the present study we examine the effects of Ang II type 2 receptor stimulation on SHP-1 activation. Ang II (10(-7) M) elicits a rapid and transient tyrosine phosphorylation of SHP-1, maximal at 1 min, in a dose-dependent form, blocked by the AT(2) antagonist, PD123319. SHP-1 phosphorylation is followed in time by tyrosine dephosphorylation of different proteins, suggesting a sequence of events. Ang II induces association of SHP-1 to AT(2) receptors as shown by co-immunoprecipitation, Western blot and binding assays. SHP-1 activity was determined in immunocomplexes obtained with either anti-AT(2) or anti-SHP-1 antibodies, after Ang II stimulation (1 min), in correlation with the maximal level of SHP-1 phosphorylation. Interestingly, following receptor stimulation (1 min) c-Src was associated to AT(2) or SHP-1 immunocomplexes. Preincubation with the c-Src inhibitor PP2 inhibited SHP-1 activation and c-Src association, thus confirming the participation of c-Src in this pathway. We demonstrated here for the first time the involvement of c-Src in SHP-1 activation via AT(2) receptors present in an ex vivo model expressing both receptor subtypes. In this model, AT(2) receptors are not constitutively associated to SHP-1 and SHP-1 is not constitutively activated. Thus, we clearly establish that SHP-1 activation, mediated by the AT(2) subtype, involves c-Src and precedes protein tyrosine dephosphorylation, in rat fetal membranes.  相似文献

Evolutionary studies indicate that a high proportion of alternative splicing (AS) events are species-specific; just 28% of minor-form alternatively spliced exons are conserved between mice and humans. We employed a splicing-sensitive microarray to study the evolution of allele-specific AS in nematodes. We compared splicing levels among five distinct Caenorhabditis elegans lines. Our results indicate that AS is less variable between natural isolates (NIs) from England, Hawaii, and Australia than when compared with mutation accumulation lines (6% vs. 21%, respectively, vary compared with N2). This suggests that strong stabilizing selection shapes the evolution of the ratios of isoforms generated by AS in C. elegans. When we analyzed some of the splicing changes between the NIs, we found examples of changes in both cis and trans that lead to alterations in gene-specific AS. This indicates that both these mechanisms for changing AS are employed along the path toward speciation in nematodes.  相似文献

Do neurons in primary motor cortex encode the generative details of motor behavior, such as individual muscle activities, or do they encode high-level movement attributes? Resolving this question has proven difficult, in large part because of the sizeable uncertainty inherent in estimating or measuring the joint torques and muscle forces that underlie movements made by biological limbs. We circumvented this difficulty by considering single-neuron responses in an isometric task, where joint torques and muscle forces can be straightforwardly computed from limb geometry. The response for each neuron was modeled as a linear function of a "preferred" joint torque vector, and this model was fit to individual neural responses across variations in limb posture. The resulting goodness of fit suggests that neurons in motor cortex do encode the kinetics of motor behavior and that the neural response properties of "preferred direction" and "gain" are dual components of a unitary response vector.  相似文献

We discuss the influence of absorption flattening (AF) on CD spectra, aiming at finding a simple way to compensate the induced distortions. A simple algebraic formalism is proposed based on an unconventional measuring approach, which makes use of a commercial unit. Validation of the proposed compensation method for AF is conducted not only on the already studied CD Vis spectral region of a Co chiral complex with the use of an AF emulator, but also on the far UV CD spectra of poly-L-glutamic acid suspensions with different turbidity levels. Comparison with some correction factors proposed in the literature is presented.  相似文献

A hierarchical animal model was developed for inference on genetic merit of livestock with uncertain paternity. Fully conditional posterior distributions for fixed and genetic effects, variance components, sire assignments and their probabilities are derived to facilitate a Bayesian inference strategy using MCMC methods. We compared this model to a model based on the Henderson average numerator relationship (ANRM) in a simulation study with 10 replicated datasets generated for each of two traits. Trait 1 had a medium heritability (h2) for each of direct and maternal genetic effects whereas Trait 2 had a high h2 attributable only to direct effects. The average posterior probabilities inferred on the true sire were between 1 and 10% larger than the corresponding priors (the inverse of the number of candidate sires in a mating pasture) for Trait 1 and between 4 and 13% larger than the corresponding priors for Trait 2. The predicted additive and maternal genetic effects were very similar using both models; however, model choice criteria (Pseudo Bayes Factor and Deviance Information Criterion) decisively favored the proposed hierarchical model over the ANRM model.  相似文献

The Mondego estuary (Portugal) has suffered severe ecological stress over the last two decades, as manifested in the replacement of seagrasses by opportunistic macroalgae, degradation of water quality and increased turbidity. A restoration plan was implemented in 1998, which aimed to reverse the eutrophication effects, and especially to restore the original natural seagrass (Zostera noltii) community. This article explores the long-term changes in Ampithoe valida and Melita palmata (Amphipoda) populations in response to eutrophication (with consequent seagrass loss and macroalgal proliferation) and to the subsequent restoration plan (with progressive seagrass recovery and macroalgal biomass decline). Until the early 1990s, high densities of A. valida and M. palmata were recorded in the Mondego estuary, especially during the occurrence of the macroalgal bloom and during all the periods in which green macroalgae were available. After the implementation of the restoration plan, species abundance, biomass and production levels decreased considerably due to the progressive decline of green macroalgae. This implied the virtual disappearance of the amphipod population, mainly A. valida. Distinct behaviours displayed by the two species could be related to different food strategies and habitat preferences. Ampithoe valida showed feeding preferences for ephemeral softer, filamentous or bladed algae (e.g. Ulva sp.) due to its high caloric content, using the Z. noltii bed only as a habitat for protection against predators or shelter from wave action. On the other hand, M. palmata did not suffer a strong decline in its population density, biomass and production, which may indicate that this species is probably not a primary consumer of green macroalgae and may readily shift to alternative ecological niches.
