Native, drought-adapted arbuscular mycorrhizal fungi (AMF) often improve host-plant performance to a greater extent than nonnative AMF in dry environments. However, little is known about the physiological basis for this differential plant response. Seedlings of Olea europaea and Rhamnus lycioides were inoculated with either a mixture of eight native Glomus species or with the nonnative Glomus claroideum before field transplanting in a semiarid area. Inoculation with native AMF produced the greatest improvement in nutrient and water status as well as in long-term growth for both Olea and Rhamnus. Foliar delta18O measurements indicated that native AMF enhanced stomatal conductance to a greater extent than nonnative AMF in Olea and Rhamnus.delta13C data showed that intrinsic water-use efficiency in Olea was differentially stimulated by native AMF compared with nonnative AMF. Our results suggest that modulation of leaf gas exchange by native, drought-adapted AMF is critical to the long-term performance of host plants in semiarid environments. delta18O can provide a time-integrated measure of the effect of mycorrhizal infection on host-plant water relations.  相似文献

In a previous work we observed multilayered plate-like structures surrounding partially denatured HeLa chromosomes at metaphase ionic conditions. This unexpected finding has led us to carry out an extensive investigation of these structures. Our results show that plates can also be found in metaphase chromosomes from chicken lymphocytes. We have used atomic force microscopy (AFM) to image and investigate the mechanical properties of plates in aqueous solution. Plates are thin (~6.5 nm each layer) but compact and resistant to penetration by the AFM tip: their Young’s modulus is ~0.2 GPa and the stress required for surface penetration is ~0.03 GPa in the presence of Mg2+ (5–20 mM). Low-ionic strength conditions produce emanation of chromatin fibers from the edges of uncrosslinked plates. These observations and AFM results obtained applying high forces indicate that the chromatin filament is tightly tethered inside the plates. Images of metal-shadowed plates and cryo-electron microscopy images of frozen-hydrated plates suggest that nucleosomes are tilted with respect to the plate surface to allow an interdigitation between the successive layers and a thickness reduction compatible with the observed plate height. The similarities between denatured plates from chicken chromosomes and aggregates of purified chromatin from chicken erythrocytes suggest that chromatin has intrinsic structural properties leading to plate formation. Scanning electron micrographs and images obtained with the 200-kV transmission microscope show that plates are the dominant component of compact chromatids. We propose that metaphase chromosomes are formed by many stacked plates perpendicular to the chromatid axis.  相似文献

We studied the influence of inoculation with a mixture of three exotic arbuscular mycorrhizal (AM) fungi, Glomus intraradices Schenck & Smith, Glomus deserticola Trappe, Bloss. & Menge and Glomus mosseae (Nicol & Gerd.) Gerd. & Trappe, and the addition of composted sewage sludge (SS) on the activities of the antioxidant enzymes superoxide dismutase (SOD, EC 1.15.1.1) and total peroxidase (POX) and of shoot and root nitrate reductase (NR, EC 1.6.6.1) in Juniperus oxycedrus L. seedlings, an evergreen shrub, grown in a non-sterile soil under well-watered and drought-stress conditions. Both the inoculation with exotic AM fungi and the addition of composted SS stimulated significantly growth and the N and P contents in shoot tissues of J. oxycedrus with respect to the plants neither inoculated nor treated with composted SS that were either well-watered or droughted. Under drought-stress conditions, only inoculation with exotic AM fungi increased shoot and root NR activity (about 188% and 38%, respectively, with respect to the plants neither inoculated nor treated with composted SS). Drought increased the POX and SOD activities in both shoots of J. oxycedrus seedlings inoculated with exotic AM fungi and grown with composted SS, but the increase was less than in the plants neither inoculated nor treated with SS. Both the plants inoculated with exotic AM fungi and the plants grown with composted SS developed additional mechanisms to avoid oxidative damage produced under water-shortage conditions.  相似文献

This study investigated the influence of inoculation with a plant growth-promoting rhizobacterium, Pseudomonas mendocina Palleroni, alone or in combination with an arbuscular mycorrhizal (AM) fungus, Glomus intraradices (Schenk & Smith) or Glomus mosseae (Nicol & Gerd.) Gerd. & Trappe, on antioxidant enzyme activities (catalase and total peroxidase), phosphatase activity, solute accumulation, growth and mineral nutrient uptake in leaves of Lactuca sativa L. cv. Tafalla affected by three different levels of salt stress. Salinity decreased lettuce growth, regardless of the biological treatment and of the salt stress level. The plants inoculated with P. mendocina had significantly greater shoot biomass than the control plants at both salinity levels, whereas the mycorrhizal inoculation treatments only were effective in increasing shoot biomass at the medium salinity level. At the highest salinity level, the water content was greater in leaves of plants treated with P. mendocina or G. mosseae. At the medium salinity level, G. intraradices- or G. mosseae-colonised plants showed the highest concentrations of foliar P. The P. mendocina- and G. mosseae-colonised plants presented higher concentrations of foliar K and lower concentrations of foliar Na under high salt conditions. Salt stress decreased sugar accumulation and increased foliar proline concentration, particularly in plants inoculated with the PGPR. Increasing salinity stress raised significantly the antioxidant enzyme activities, including those of total peroxidase and catalase, of lettuce leaves compared to their respective non-stressed controls. The PGPR strain induced a higher increase in these antioxidant enzymes in response to severe salinity. Inoculation with selected PGPR could serve as a useful tool for alleviating salinity stress in salt-sensitive plants.  相似文献

Mycorrhizae may help plants to thrive in Mediterranean semi-arid ecosystems by altering antioxidant enzyme activities. Our objective was to determine the influence of mycorrhizal inoculation with an allochthonous arbuscular mycorrhizal (AM) fungus, Glomus claroideum , Schenck & Smith, or with a mixture of native AM fungi, on the activity of antioxidant enzymes from shoots of Olea europaea L. ssp. sylvestris , Retama sphaerocarpa (L.) Boissier and Rhamnus lycioides L. seedlings afforested in a degraded Mediterranean semi-arid soil. One year after planting, shoot biomass of inoculated O. europaea seedlings was about 630%, of non-inoculated plants. Shoot biomass of G. claroideum -colonized R. sphaerocarpa was greater than that of seedlings inoculated with the mixed native AM fungi after 12 months. Inoculation with a mix of native AM fungi was the most effective treatment for increasing shoot biomass and N, P and K contents in shoot tissues of R. lycioides . Both mycorrhizal inoculation treatments increased the nutrient contents in shoots of O. europaea and R. lycioides . In O. europaea plants, the inoculation treatments increased catalase, ascorbate peroxidase and dehydroascorbate reductase activities, but not monodehydroascorbate reductase and glutathione reductase activities. Inoculation with G. claroideum increased the activities of all antioxidant enzymes in R. sphaerocarpa . Monodehydroascorbate reductase, glutathione reductase and superoxide dismutase activities in R. lycioides leaves were preferentially increased by inoculation with the mixture of native AM fungi. This work support the view that increased antioxidant enzyme activities could be involved, at least in part, in the beneficial effects of mycorrhizal colonization on the performance of shrub species grown under semi-arid Mediterranean conditions.  相似文献

During a revegetation field experiment in Southeast Spain, we measured foliar carbon isotope ratios (delta13C) and gas exchange parameters in order to evaluate the influence of arbuscular mycorrhizal (AM) infection on the water use efficiency (WUE) of two semiarid woodland species. WUE during drought was significantly enhanced by inoculation with Glomus intraradices in Olea europaea ssp sylvestris, but not in Rhamnus lycioides. While Olea is a long-lived, slow-growing evergreen tree with a conservative water use strategy, Rhamnus is a drought-deciduous shrub with a shorter lifespan; these differences may explain their dissimilar patterns of physiological response to inoculation with the same AM fungus. Differences in delta13C and WUE between Olea and Rhamnus were larger when comparing AM inoculated than non-inoculated seedlings. This result suggests that some of the interspecific variability in delta13C observed for aridland plant communities may be due to different physiological responses to mycorrhization.  相似文献

Our aim was to examine the effect of water stress on plant growth and development of two native plant species (Tetraclinis articulata and Crithmum maritimum) and on microbial community composition and activity in the rhizosphere soil, following the addition of an organic amendment, namely sugar beet residue (SBR), and/or the inoculation with an arbuscular mycorrhizal (AM) fungus, namely Glomus mosseae, in a non-sterile heavy metal-polluted soil. The AM inoculation did not have any significant effect on plant growth of both species. In T. articulata, SBR increased shoot growth, foliar P, total phospholipid fatty acids (PLFA), fungi-related PLFA, AM fungi-related neutral lipid fatty acid, bacterial gram-positive/gram-negative PLFA ratio and the β-glucosidase and dehydrogenase activities. SBR and AM inoculation increased phosphatase activity in T. articulata plants grown under drought conditions. In both plants, there was a synergistic effect between AM inoculation and SBR on mycorrhizal colonisation under drought conditions. In C. maritimum, the increase produced by the SBR on total amounts of PLFA, bacterial gram-positive-related PLFA and bacterial gram-negative-related PLFA was considerably higher under drought conditions. Our results suggest that the effectiveness of the amendment with regard to stimulating microbial communities and plant growth was largely limited by drought, particularly for plant species with a low degree of mycorrhizal colonisation.  相似文献

Two tracksites yielding tridactyl footprints II-IV of theropods as well as supposedly sauropod tracks from the Causses Basin, briefly described in 2006 are reinvestigated. They are the Capelan tracksite (Meyrueis, Lozère) and the Garène tracksite (La Roque-Sainte-Marguerite, Aveyron), both Bathonian in age. In 2012, one of them (Le Capelan) was excavated and enlarged, allowing the discovery of numerous new tracks. In the Capelan tracksite, 102 concave epireliefs, 11 to 36 cm long, represent at least 21 trackways; the longest being visible along 37 meters. Footprints are almost as long as wide and show a large divarication angle II-IV (up to 70°). The largest of them (more than 24 cm long) were statistically compared with different Middle and Upper Jurassic ichnotaxa for which a synthesis is proposed. Tracks from the Causses Basin as well as those from the Capelan and the Garène tracksites can be ascribed to the ichnogenus Kayentapus. We propose a new ichnospecies Kubacensis nov. isp. Whose trackmakers were tetanurian theropods, probably megalosaurids. At the Capelan, some marks are tentatively interpreted as swim tracks of crocodylomorphs. The supposedly sauropod tracks are here reinterpreted and considered as erosion cups and as concavities indicating the location of tree trunks. The sedimentological analysis indicates a paralic environment including lagoons and freshwater to brackish bays. They were separated from dysoxic lagoons by calcarenitic coastal barrier spits, and by tidal flats where microbial mats contributed to the good preservation of trackways. Despite the presence of “mangroves”, the main track-bearing surface from the Capelan was quite open and visited by megalosaurid dinosaurs of various sizes, moving in all directions but preferably to the North. The regional palaeogeography of the early-middle Bathonian is in accordance with a coastal area with low relief, bordering the “Seuil Cévenol”, which was probably emerged.  相似文献
