Many years' research into the concentration of unstable organic matter in ocean waters from many regions of the World Ocean is compiled. The relative amounts of this matter, as determined by counts of heterotrophic bacteria, are presented as three-dimensional illustrations, depicting specific ocean areas from the surface to the sea floor. The merits of using this information in oceanography are discussed, particularly in cases where other parameters are indistinct. In many cases, this is viewed as the best method for determining the origin of water layers at various depths, and of following the movement of water masses in the ocean.  相似文献

Using antibody‐related methods, we recently found that human thyroid cells express various proteins differently depending on whether they are cultured under normal gravity (1g) or simulated microgravity (s‐μg). In this study, we performed proteome analysis in order to identify more gravity‐sensitive thyroid proteins. Cells cultured under 1g or s‐μg conditions were sonicated. Proteins released into the supernatant and those remaining in the cell fragments were fractionated by free‐flow IEF. The fractions obtained were further separated by SDS‐gel electrophoresis. Selected gel pieces were excised and their proteins were determined by MS. A total of 235 different proteins were found. Out of 235 proteins, 37 appeared to be first identifications in human thyroid cells. Comparing SDS gel lanes of equally numbered free‐flow IEF fractions revealed similar patterns with a number of identical bands if proteins of a distinct cell line had been applied, irrespective of whether the cells had been cultured under 1g or s‐μg. Most of the identical band pairs contained identical proteins. However, the concentrations of some types of proteins were different within the two pieces of gel. Proteins that concentrated differently in such pieces of gel are considered as candidates for further investigations of gravitational sensitivity.  相似文献

Epidermal growth factor receptor localization in the rat and monkey testes   总被引:5,自引:0,他引:5  
Epidermal growth factors receptor (EGFR) was localized immunocytochemically in the testes of mature and immature rats and immature monkeys. One polyclonal antibody, recognizing the intracellular domain (RK2) of the receptor, was used to carry out the EGFR immunodetection. The RK2 antibody revealed the presence of the EGFR predominantly in Sertoli cells of mature and immature rats and of immature monkeys, although limited interstitial localization of the EGFR was also discerned in the mature rat. In cultured Sertoli cells of immature rats, grown in the absence of epidermal growth factor (EGF), the EGFR was randomly distributed at the cell surface, whereas after the addition of EGF the receptor became aggregated into distinct focal regions. In addition, EGFR of cultured Sertoli cells exhibited autophosphorylation activity upon stimulation with EGF, but failed to transcytose iodinated EGF across a permeability barrier formed by the cultured cells. Instead, all of the added iodinated EGF was internalized and degraded.  相似文献

In rodent brown adipose tissue, the beta-adrenergic signaling is believed, by an action on PGC-1alpha, to control UCP1 expression and mitochondriogenesis. We addressed this hypothesis using beta(1)/beta(2)/beta(3)-adrenoceptor knockout (beta-less) brown adipocytes in primary culture. In these cells: (a) proliferation and differentiation into multilocular cells were normal; (b) UCP1 mRNA expression was dramatically decreased (by 93%), whereas PGC-1alpha and mtTFA mRNA expressions were not; (c) UCP1, PGC-1alpha and COX IV protein expressions were decreased by 97%, 62% and 22%, respectively. Altogether the data show a dissociation between the control of UCP1, which is mostly beta-adrenoceptor-dependent and that of PGC-1alpha and of mitochondriogenesis which are not.  相似文献

Anthropogenic activity imposes increasing pressure on wildlife populations globally; these pressures can affect habitat suitability and function, modify wildlife space use, and influence population viability. Native mountain goat (Oreamnos americanus) populations can be negatively affected by anthropogenic disturbance and modify their space use in response to land development and recreational activity. From 2018 to 2020, we studied space use of mountain goats northeast of Smithers, British Columbia, Canada, an area that is subject to increasing anthropogenic development and yearlong recreational activities. We aimed to generate models that would improve our ability to identify habitat for mountain goats relative to existing survey data and established ungulate winter ranges. Using resource selection function (RSF) analyses generated from global positioning system (GPS) collar data, we identified influential habitat covariates and compared these covariates and RSF values to existing habitat models. Additionally, we compared the extent to which our models were congruent with existing resource selection probability functions, were congruent with aerial survey data, and overlapped existing ungulate winter ranges previously derived from predictive models inside and outside of the study area. Overall, our models noted higher RSF values among GPS data relative to aerial survey data for winter months, while results for summer habitats were comparable. In extending our RSFs outside of the study area and evaluating the overlap with ungulate winter ranges in adjacent areas, values were similar, albeit lower, as is expected given that the models were developed elsewhere. Ultimately, these models, combined with existing methods, improve the accuracy and reliability of identified, important areas of habitat for mountain goats. We recommend that the RSF models generated here be used in conjunction with aerial survey data and existing methods to delineate ungulate winter ranges for mountain goats in similar eco-regions in British Columbia. The models developed here support existing methods that have been used to delineate or validate ungulate winter ranges for mountain goats in British Columbia and help facilitate mitigation measures to support the continued use of important winter habitat and significant landscape features that play a role in ensuring population viability and resilience through time.  相似文献
