A novel series of arylsulfonamide derivatives of (aryloxy)propyl piperidines was designed to obtain potent 5-HT7R antagonists. Among the compounds evaluated herein, 3-chloro-N-{1-[3-(1,1-biphenyl-2-yloxy)2-hydroxypropyl]piperidin-4-yl}benzenesulfonamide (25) exhibited antagonistic properties at 5-HT7R and showed selectivity over selected serotoninergic and dopaminergic receptors, as well as over serotonin, noradrenaline and dopamine transporters. Compound 25 demonstrated significant antidepressant-like activity in the forced swim test (0.625–2.5 mg/kg, i.p.) and in the tail suspension test (1.25 mg/kg, i.p.), augmented the antidepressant effect of inactive doses of escitalopram (selective serotonin reuptake inhibitor) and bupropion (dopamine reuptake inhibitor) in the FST in mice, and similarly to SB-269970, exerted pro-cognitive properties in the novel object recognition task in cognitively unimpaired conditions in rats (0.3 mg/kg, i.p.). Such an extended pharmacological profile, especially the augmentation effect of the identified 5-HT7R antagonist on SSRI activity, seems promising regarding the complexity of affective disorders and potentially improved outcomes, including mnemonic performance.  相似文献

A cytogenetic analysis was performed in experimental hybrids between species of Chagas disease transmitting bugs with remarkable differences in the amount and distribution of heterochromatin. Using C-banding technique, we identified the parental species chromosomes and analysed the meiotic behaviour in the male hybrids between Triatoma platensis and T. infestans, T. platensis and T. delpontei, and T. infestans and T. rubrovaria. The two former hybrids have an entirely normal meiotic behaviour despite the extensive differences in C-banded karyotypes observed in the parental species, indicating that heterochromatin differences between homeologous chromosomes are not a barrier that influences meiotic synapsis and recombination. On the contrary, the experimental hybrids between T. infestans and T. rubrovaria show failures in pairing of homeologous chromosomes that lead to the production of abnormal spermatids and hybrid sterility. Our data suggest that karyotypic repatterning within triatomines has involved at least two different pathways. Among closely related species, chromosomal changes have largely involved addition or deletion of heterochromatic regions. In more distant species, chromosomal rearrangements (i.e. inversions and translocations) have also arisen. Hybridisation data also allow to hypothesize about the origin and divergence of this taxonomic group, as well as the mechanisms that maintain species isolation.  相似文献

Mutation or multiplication of the alpha-synuclein (Syn)-encoding gene is frequent cause of early onset Parkinson''s disease (PD). Recent evidences point to the pathogenic role of excess Syn also in sporadic PD. Syn is a cytosolic protein, which has been shown to be released from neurons. Here we provide evidence that extracellular Syn induces an increase in surface-exposed glucose-related protein of 78 kDa (GRP78), which becomes clustered in microdomains of the neuronal plasma membrane. Upon interacting with Syn, GRP78 activates a signaling cascade leading to cofilin 1 inactivation and stabilization of microfilaments, thus affecting morphology and dynamics of actin cytoskeleton in cultured neurons. Downregulation of GRP78 abolishes the activity of exogenous Syn, indicating that it is the primary target of Syn. Inactivation of cofilin 1 and stabilization of actin cytoskeleton are present also in fibroblasts derived from genetic PD patients, which show a dramatic increase in stress fibers. Similar changes are displayed by control cells incubated with the medium of PD fibroblasts, only when Syn is present. The accumulation of Syn in the extracellular milieu, its interaction with the plasma membrane and Syn-driven clustering of GRP78 appear, therefore, responsible for the dysregulation of actin turnover, leading to early deficits in synaptic function that precede neurodegeneration.Alpha-synuclein (Syn), a small soluble protein abundant in the brain,1 is implicated in the pathogenesis of various neurodegenerative disorders, among them Parkinson''s disease (PD).2 Syn is the major component of intraneuronal inclusions found in the brain of PD patients.3 Distinct mutations4 as well as multiplications of the Syn-encoding gene (SNCA)5, 6, 7 have been identified in families with genetic forms of PD. Moreover, genome-wide association studies correlated polymorphisms in regions of the SNCA gene to sporadic cases of PD.8Syn is a natively unfolded protein enriched in presynaptic terminals, where it dynamically associates with synaptic vesicle membrane and lipid rafts.9, 10 A plethora of possible Syn interactors has been recognized11 and among them cytoskeletal proteins. The influence of Syn on the microtubule-based cytoskeleton has been characterized in vitro.12 Recently,13 Syn has been shown to depolymerize actin cytoskeleton, a master regulator of synaptic function,14 raising the possibility that the effects of Syn on synaptic vesicle dynamics are mediated by its interaction with actin.Syn is localized in the cytosolic compartment, is released from cultured neurons and is present in biological fluids15 indicating that Syn may also act extracellularly, participating in cell-to-cell transmission of pathology. Intraneuronal inclusions were found in healthy engrafted cells decades after transplantation in PD patients, and the host-to-graft transmission of Syn pre-formed fibrils has been demonstrated to occur in the mouse brain.16Here we show that exposure of neurons to extracellularly delivered wild-type (wt) or A30P-mutant monomeric Syn leads to cofilin 1 inactivation with the ensuing stabilization of microfilaments. These effects depend on Syn-driven clustering at the cell surface of the endoplasmic reticulum (ER) protein glucose-related protein of 78 kDa (GRP78). We propose actin cytoskeleton modification as the initial step in Syn-induced neurodegeneration, thereby actin and actin-binding proteins might constitute an effective target for screening and therapy of PD.  相似文献

Previous attempts to study the cytoarchitecture of cardiac Purkinje fibers with the scanning electron microscope (SEM) have been limited by the surrounding dense connective tissue. In this study the connective tissue was removed by treatment with 8N HCl, after adult sheep hearts were fixed in diastole or systole and tissue taken for SEM and transmission electron microscopy (TEM). In SEM, Purkinje fibers freely anastomosed in false tendons and formed a subendocardial plexus. In systole, medium and small-sized Purkinje fibers formed deep clefts not observed in diastole. The clefts are thought to be due to sarcolemmal folding and fiber buckling and may therefore affect conduction. The myofibrils beneath the laterally apposed sarcolemmas of adjacent Purkinje cells when fixed in systole were often observed as tightly curved arches in series. Similar configurations with expanded arches were observed in diastole. The formation of arches by myofibrils is unique to Purkinje fibers and is interpreted as the mechanism responsible for their compliance to stretch. The significance of contraction in producing the observed geometric changes in Purkinje fibers and the implications of their cytoarchitecture with respect to conduction are discussed.  相似文献

Mathematical models of the interaction between predator and host populations have been expressed as systems of nonlinear ordinary differential equations. Solutions of such systems may be periodic or aperiodic. Periodic, oscillatory solutions may depend on the initial conditions of the system or may be limit cycles. Aperiodic solutions can, but do not necessarily, exhibit oscillatory behavior. Therefore, it is important to characterize predatory-prey models on the basis of the possible types of solutions they may possess. This characterization can be accomplished using some well-known methods of nonlinear analysis. Examination of the system singular points and inspection of phase plane portraits have proved to be useful techniques for evaluating the effect of various modifications of early predator-prey models. Of particular interest is the existence of limit cycle oscillations in a model in which predator growth rate is a function of the concentration of prey.  相似文献

The predator-prey interactions between the protozoan Tetrahymena pyriformis and the bacterium Aerobacter aerogenes have been studied experimentally and mathematically. A mathematical model for the ciliates defines the mass distribution of cells within the population. The resulting model equations are solved by the use of multigroup theory. Experimental data from batch and continuous flow reactors are compared with the results of the numerical integration.  相似文献

Dialysis-related amyloidosis is characterized by the deposition of insoluble fibrils of beta(2)-microglobulin (beta(2)-m) in the musculoskeletal system. Atomic force microscopy inspection of ex vivo amyloid material reveals the presence of bundles of fibrils often associated to collagen fibrils. Aggregation experiments were undertaken in vitro with the aim of reproducing the physiopathological fibrillation process. To this purpose, atomic force microscopy, fluorescence techniques, and NMR were employed. We found that in temperature and pH conditions similar to those occurring in periarticular tissues in the presence of flogistic processes, beta(2)-m fibrillogenesis takes place in the presence of fibrillar collagen, whereas no fibrils are obtained without collagen. Moreover, the morphology of beta(2)-m fibrils obtained in vitro in the presence of collagen is extremely similar to that observed in the ex vivo sample. This result indicates that collagen plays a crucial role in beta(2)-m amyloid deposition under physiopathological conditions and suggests an explanation for the strict specificity of dialysis-related amyloidosis for the tissues of the skeletal system. We hypothesize that positively charged regions along the collagen fiber could play a direct role in beta(2)-m fibrillogenesis. This hypothesis is sustained by aggregation experiments performed by replacing collagen with a poly-L-lysine-coated mica surface. As shown by NMR measurements, no similar process occurs when poly-L-lysine is dissolved in solution with beta(2)-m. Overall, the findings are consistent with the estimates resulting from a simplified collagen model whereby electrostatic effects can lead to high local concentrations of oppositely charged species, such as beta(2)-m, that decay on moving away from the fiber surface.  相似文献

Soft fruits are cultivated in many parts of the world and are heavily attacked by a huge range of arthropod pests, including many Tephritidae flies. Soft fruits are consumed by a broad array of consumers, including children, pregnant women and elderly people; thus, high safety levels are required to ensure their adequate protection. On this basis, ecofriendly control tools against Tephritids Infesting Soft Fruits (TISF) are urgently needed. Despite extensive research carried out on Tephritidae behavioral ecology, little is known about lek dynamics and cues evoking mating behavior in TISF. Here we provide a focused look at this issue, reviewing the current knowledge about sexual communication in TISF. First, we examine pre-courtship lek dynamics and the role of male-male aggression in TISF. Second, we review knowledge about mating behavior sequences, with special reference to chemical (mainly sex pheromones) and physical cues (both vibrational and visual) guiding mate choice dynamics. Third, current and potential Integrated Pest Management applications aimed at area-wide control of TISF are outlined, with special reference to improvement of monitoring, the sterile insect technique, and “lure and kill” tools, including the male annihilation technique based on the sprayable specialized pheromone and lure application technology.  相似文献

Dientamoeba fragilis is a pathogenic protozoan parasite with a world-wide distribution. Interestingly, a resistant cyst stage has not been demonstrated and it is still an unsolved problem how this parasite can survive successfully outside the human host. D. fragilis was found in 2% of approximately 2500 individuals unselected who submitted stools for parasitological examination during 2001 in Padua (Italy). The goal of this study was to detect the protozoan stages and the duration of persistence of this protozoa in faeces stored in different environmental conditions. The trophozoites of D. fragilis were detected up to 60 days after the collection of the faeces stored at 4 degrees C and Giemsa stained. The laboratory detection rate of the organism is greatly enhanced by use of preservative to fix stool specimens immediately after passage. Alternatively, a microscopic observation of the collected stool has to be performed immediately after passage followed by examination of permanently-stained smears. Demonstration of the charateristic "golf-club" and "acanthopodia-like" structures in unstained fixed faecal material by direct microscopy (400x) are suitable for a rapid identification of D. fragilis.  相似文献

Lysiphlebus testaceipes (Hymenoptera: Braconidae: Aphidiinae) is a generalist endoparasitoid attacking more than 100 aphid species. In L. testaceipes, wing fanning is a main male courtship display evoked by a female‐borne sex pheromone. However, no information is available on the characteristics and behavioral role of male fanning during courtship in this parasitoid. Here, the courtship behavior of a wild strain of L. testaceipes was quantified and the male wing fanning performances were analyzed through high‐speed video recordings and examined in relation to mating success. Courtship sequence of wild L. testaceipes did not substantially differ from that previously reported for other populations mass reared on aphids. We observed that the male courtship duration did not affect mating success. However, video analysis revealed that the males producing high‐frequency fanning signals achieved higher mating success over those that display low‐frequency fanning. Wing fanning before successful and unsuccessful courtship differed in amplitude of wing movements and alignment toward the mate, highlighting that frontal courtship positively influence the female mating decisions. This study increases knowledge on sexual behavior in a key parasitoid of aphids, highlighting the importance of wing fanning among the range of sensory modalities used in the sexual communication of L. testaceipes. From a practical point of view, this information is useful in L. testaceipes‐based biocontrol strategies, since it can help to establish parameters for quality checking of mass‐reared wasps over time.  相似文献
