Trans-splicing is an unusual process in which two separate RNA strands are spliced together to yield a mature mRNA. We present a novel computational approach which has an overall accuracy of 82% and can predict 92% of known trans-splicing sites. We have applied our method to chromosomes 1 and 3 of Leishmania major, with high-confidence predictions for 85% and 88% of annotated genes respectively. Suppression subtractive hybridization was used to identify differentially expressed genes in hemocytes from carpet-shell clam Ruditapes decussatus stimulated with a mixture of dead bacterial strains. Putative function could be assigned to 100 of the 253 sequenced cDNAs. Based on sequence homologies, 3.16% of the total identified genes were possibly related to immune functions. Clam myticin isoforms 1, 2 and 3, and clam mytilin, with similarity with myticins and mytilins previously reported on Mytilus galloprovincialis were identified and characterized for the first time in clams. The analysis of their expression levels by quantitative PCR showed that they were induced by bacterial challenge. The results obtained in this work could be the first step leading to the understanding of molecular mechanisms by which these economically important marine bivalves respond to pathogens. 相似文献

Rhodium and iridium complexes of the chiral triaza ligands, {N,N′-bis{[(2S)-(1-benzylpyrrolidinyl)]methyl}amine (2), N,N′-bis{[(2S)-(1-benzylpyrrolidinyl)]methyl}-N-propylamine (3), N,N′-bis{[(2S)-(1-benzylpyrrolidinyl)]methyl}-N-[3-(triethoxysilyl)propyl]amine (4)}, are described. All ligands form one to one [ML] species with the above metal ions. The structures of these complexes were elucidated by analytical and spectroscopic data (elemental analysis, mass spectroscopy, IR, 1H and 13C NMR). The fixation of the preformed triethoxysilyl-rhodium and iridium complexes, on mesoporous solids (MCM-41, SBA-15), and their use, under heterogeneous conditions, for the hydrogenation reactions are reported. The catalytic activity and selectivity of heterogenised complexes are higher to that observed under homogeneous conditions, as a consequence of the complex- and/or reagents-to-support interaction. The stable covalent bond between support and complex allows the recovery and recycling of the heterogenised catalysts for a number of cycles, moreover atomic absorption analysis of the reaction solutions shows that there is not any metal leaching into the solutions. 相似文献

The silver(I) complexes [Ag{C5H4N(NC)}]n(BF4)n (1), [Ag{C5H4N(NC)}2]n(BF4)n (2), [Ag{C6H4(NC)2}]n(BF4)n (3), and [Ag{C6H4(NC)2}2]n(BF4)n (4) have been synthesized using different Ag:L ratios of 2-isocyanopyridine (or 2-pyridylisocyanide, CNPy-2) or 1,2-phenylenediisocyanide ligands. The polymeric complex 2 has been characterized by X-ray diffraction revealing a polymeric chain structure. Breaking the polymeric structure of [Ag{C6H4(NC)2}]n(BF4)n (3) with acetonitrile, the dimeric complex [Ag{(CN)2C6H4)}(NCMe)2]2(BF4)2 (5) is formed, which has been also characterized by X-ray diffraction. 相似文献

A pilot-scale membrane bioreactor was used to treat urban wastewater using pure oxygen instead of air as a source of aeration, to study its influence on bacterial diversity and levels of enzyme activities (acid and alkaline phosphatases, glucosidase, protease, and esterase) in the sludge. The experimental work was developed in two stages influenced by seasonal temperature. Operational parameters (temperature, pH, BOD5, COD, total and volatile suspended solids) were daily monitored, and enzyme activities measured twice a week. Redundancy analysis (RDA) was used to reveal relationships between the level of enzyme activities and the variation of operational parameters, demonstrating a significant effect of temperature and volatile suspended solids. Bacterial diversity was analyzed by temperature-gradient gel electrophoresis of PCR-amplified partial 16S rRNA genes. Significant differences in community structure were observed between both stages. Sequence analysis revealed that the prevalent Bacteria populations were evolutively close to Alphaproteobacteria (44%), Betaproteobacteria (25%) and Firmicutes (17%). 相似文献

The roles of adaptation, chance, and history on evolution of the toxic dinoflagellate Alexandrium minutum Halim, under selective conditions simulating global change, have been addressed. Two toxic strains (AL1V and AL2V), previously acclimated for two years at pH 8.0 and 20°C, were transferred to selective conditions: pH 7.5 to simulate acidification and 25°C. Cultures under selective conditions were propagated until growth rate and toxin cell quota achieved an invariant mean value at 720 days (ca. 250 and ca. 180 generations for strains AL1V and AL2V, respectively). Historical contingencies strongly constrained the evolution of growth rate and toxin cell quota, but the forces involved in the evolution were not the same for both traits. Growth rate was 1.5-1.6 times higher than the one measured in ancestral conditions. Genetic adaptation explained two-thirds of total adaptation while one-third was a consequence of physiological adaptation. On the other hand, the evolution of toxin cell quota showed a pattern attributable to neutral mutations because the final variances were significantly higher than those measured at the start of the experiment. It has been hypothesized that harmful algal blooms will increase under the future scenario of global change. Although this study might be considered an oversimplification of the reality, it can be hypothesized that toxic blooms will increase but no predictions can be advanced about toxicity. 相似文献

Replicated, factorial mesocosm experiments were conducted across Europe to study the effects of nutrient enrichment and fish
density on macrophytes and on periphyton chlorophyll a (chl-a) with regard to latitude. Periphyton chl-a densities and plant decline were significantly related to nutrient loading in all countries. Fish effects were significant
in a few sites only, mostly because of their contribution to the nutrient pool. A saturation-response type curve in periphyton
chl-a with nutrients was found, and northern lakes achieved higher densities than southern lakes. Nutrient concentration and phytoplankton
chl-a necessary for a 50% plant reduction followed a latitudinal gradient. Total phosphorus values for 50% plant disappearance
were similar from Sweden (0.27 mg L−1) to northern Spain (0.35 mg L−1), but with a sharp increase in southern Spain (0.9 mg L−1). Planktonic chl-a values for 50% plant reduction increased monotonically from Sweden (30 μg L−1) to València (150 μg L−1). Longer plant growing-season, higher light intensities and temperature, and strong water-level fluctuations characteristic
of southern latitudes can lead to greater persistence of macrophyte biomass at higher turbidities and nutrient concentration
than in northern lakes. Results support the evidence that latitudinal differences in the functioning of shallow lakes should

Glucosidase I is an important enzyme in N-linked glycoprotein processing, removing specifically distal alpha-1,2-linked glucose from the Glc3Man9GlcNAc2 precursor after its en bloc transfer from dolichyl diphosphate to a nascent polypeptide chain in the endoplasmic reticulum. We have identified a glucosidase I defect in a neonate with severe generalized hypotonia and dysmorphic features. The clinical course was progressive and was characterized by the occurrence of hepatomegaly, hypoventilation, feeding problems, seizures, and fatal outcome at age 74 d. The accumulation of the tetrasaccharide Glc(alpha1-2)Glc(alpha1-3)Glc(alpha1-3)Man in the patient's urine indicated a glycosylation disorder. Enzymological studies on liver tissue and cultured skin fibroblasts revealed a severe glucosidase I deficiency. The residual activity was <3% of that of controls. Glucosidase I activities in cultured skin fibroblasts from both parents were found to be 50% of those of controls. Tissues from the patient subjected to SDS-PAGE followed by immunoblotting revealed strongly decreased amounts of glucosidase I protein in the homogenate of the liver, and a less-severe decrease in cultured skin fibroblasts. Molecular studies showed that the patient was a compound heterozygote for two missense mutations in the glucosidase I gene: (1) one allele harbored a G-->C transition at nucleotide (nt) 1587, resulting in the substitution of Arg at position 486 by Thr (R486T), and (2) on the other allele a T-->C transition at nt 2085 resulted in the substitution of Phe at position 652 by Leu (F652L). The mother was heterozygous for the G-->C transition, whereas the father was heterozygous for the T-->C transition. These base changes were not seen in 100 control DNA samples. A causal relationship between the alpha-glucosidase I deficiency and the disease is postulated. 相似文献
